Hydroponics is currently one of the primary methods for soilless cultivation. Although the phenotype and quality of vegetables differ between hydroponic and soil-based systems, limited research has been conducted on the selection and breeding of pakchoi cultivars specifically suited for hydroponics. In this study, principal component analysis (PCA) and cluster analysis were performed on the commercial traits, agronomic characteristics, and nutritional quality of 20 pakchoi parental lines grown under hydroponic conditions to classify and screen suitable germplasm for breeding. PCA reduced the 11 agronomic traits into two independent principal components, accounting for a cumulative contribution of 79.22%. Cluster analysis grouped the 20 parental lines into four categories based on the composite scores of agronomic traits and nutritional quality. Group 3 was selected for breeding programs aiming to develop high-yielding cultivars with a desirable morphology. For breeding targets emphasizing darker leaves and petiole coloration, Group 4 presented the most suitable germplasm. Group 1 was ideal for enhancing nutritional quality by offering parent lines rich in calcium, magnesium, vitamin C, and amino acids. Alternatively, Group 2 contained lines with high levels of soluble proteins, amino acids, and soluble sugars.

R2R3-MYB represents a substantial gene family that plays diverse roles in plant development. In this study, 102 SmR2R3-MYB genes were identified from eggplant fruit and classified into 31 subfamilies. Analysis indicated that segmental duplication events played a pivotal role in the expansion of the SmR2R3-MYB gene family. Furthermore, the prediction of miRNAs targeting SmR2R3-MYB genes revealed that 60 SmR2R3-MYBs are targeted by 57 miRNAs, with specific miRNAs displaying varying numbers of target genes, providing valuable insights into the regulatory functions of miRNAs in plant growth, development, and responses to stress conditions. Through expression profile analysis under various treatment conditions, including low temperature (4 °C), plant hormone (ABA, Abscisic acid), and drought stress (PEG, Polyethylene glycol), diverse and complex regulatory mechanisms governing SmR2R3-MYB gene expression were elucidated. Notably, EGP21875.1 and EGP21874.1 exhibited upregulation in expression under all treatment conditions. Transcriptome and metabolome analyses demonstrated that, apart from anthocyanins (delphinidin-3-O-glucoside, cyanidin-3-O-(6-O-p-coumaroyl)-glucoside, and malvidin-3-O-(6-O-p-coumaroyl)-glucoside), overexpression of SmMYB75 could also elevate the content of various beneficial compounds, such as flavonoids, phenolic acids, and terpenes, in eggplant pulp. This comprehensive study enhances our understanding of SmR2R3-MYB gene functions and provides a strong basis for further research on their roles in regulating anthocyanin synthesis and improving eggplant fruit quality.

Flag leaf angle (FLA) in rice (Oryza sativa L.) is one of the important traits affecting F1 seed production by mechanization. Here, we report the map-based cloning and functional characterization of the FLA1 (FLAG-LEAF-ANGLE 1) gene, which resides at a major-effect quantitative trait locus (QTL). Through cell morphological observations and exogenous hormone treatment assays, we demonstrate that gibberellin (GA) modulates rice FLA by altering both the number of cell layers and cell length. Combining genetic and molecular biological analyses with genetic complementation and gene overexpression assays, we elucidated and validated the biological function of FLA1. In addition, we found that FLA1 is constitutively expressed and encodes a protein localized to both the cell membrane and nucleus. Via RT-qPCR assays, we further demonstrated that the FLA1fla-R allele from the rice accession fla-R enhances GA biosynthesis by upregulating the expression of CLA1 and GA20ox2. Furthermore, yeast two-hybrid assays revealed that auxin-repressed protein 1 (ARP1) interacts with FLA1, suggesting a potential role of this interaction in the modulation of rice FLA. Collectively, our results demonstrate that optimizing rice FLA via molecular manipulation of FLA1 can resolve the problem of flag leaf shearing during F1 hybrid rice seed production without compromising F1 hybrid seed yield, thereby facilitating mechanized F1 hybrid rice seed production.
