The Debus–Radziszewski imidazole synthesis was adapted to directly yield long-chain imidazolium ionic liquids. Imidazolium acetate ionic liquids with side-chains up to sixteen carbon atoms were synthesised in excellent yields via an on-water, one-pot reaction. The imidazolium acetate ILs acted as surfactants when dissolved in various solvents. The imidazolium acetate ionic liquids were also derivatised via an acid metathesis to the chloride, nitrate, and hydrogen oxalate derivatives. The thermal behaviour of all the ionic liquids was determined via thermogravimetric and calorimetric analysis.

In this work we investigate the behaviour of molecules at the nanoscale using scanning tunnelling microscopy in order to explore the origin of the cooperativity in the formation of self-assembled molecular networks (SAMNs) at the liquid/solid interface. By studying concentration dependence of alkoxylated dimethylbenzene, a molecular analogue to 5-alkoxylated isophthalic derivatives, but without hydrogen bonding moieties, we show that the cooperativity effect can be experimentally evaluated even for low-interacting systems and that the cooperativity in SAMN formation is its fundamental trait. We conclude that cooperativity must be a local effect and use the nearest-neighbor Ising model to reproduce the coverage vs. concentration curves. The Ising model offers a direct link between statistical thermodynamics and experimental parameters, making it a valuable tool for assessing the thermodynamics of SAMN formation.

Surfactants are extremely versatile ubiquitous compounds with a wide range of applications. Traditional surfactants are based on non-renewable sources, while the alternative surfactants from natural feedstock remain underexplored. In our work, we synthesized and characterized a library of bioderived compounds with different structural properties. Namely, amide and ester derivatives of C10–C16 fatty acids and the amino or ⍺-hydroxy acids, including methionine, aspartic, glutamic, malic and citric acid. To further elucidate the structure-propery relationship, we also included iminodiacetic acid, a non-natural acid. The surfactant molecular structures varied in the number of carboxylic groups (one to three), the length of the hydrophobic chain and the type of linkage between the two parts of the molecule (ester, secondary or tertiary amide). The structural differences had a pronounced impact on their foaming properties, critical micelle concentration (CMC), and maximum tolerable concentration (MTC) in aquatic life, studied using zebrafish as model animals. The compounds exhibited a broad range of foaming properties across the whole pH range. Their respective CMC values spanned several orders of magnitude, and a linear relationship between the logarithm of CMC and the hydrophobic chain length was observed. Several compounds showed very high MTC values. The obtained results provide a basis for further development of bioderived surfactants and their use in different domains.

Background/Objectives: RMC-113, a 3-alkynyl-6-aryl-disubstituted isothiazolo[4,3-b]pyridine, is a dual inhibitor of the lipid kinases PIKfyve and PIP4K2C with broad-spectrum antiviral activity. The aim was to study the structure–activity relationship (SAR) of isothiazolo[4,3-b]pyridines as dual PIKfyve/PIP4K2C inhibitors. Methods: A series of isothiazolo[4,3-b]pyridines was synthesized by introducing structural variety at positions 3 and 6 of the central scaffold. The primary assay to guide the synthetic chemistry was a biochemical PIKfyve assay, with a number of analogues also tested for PIP4K2C binding affinity. Finally, isothiazolo[4,3-b]pyridines were also evaluated for antiviral and antitumoral activity in cell-based assays. Results: PIKfyve inhibition tolerated a wide variety of substituents on the aryl ring at position 6 of the isothiazolo[4,3-b]pyridine scaffold, with the 4-carboxamide analogue emerging as the most potent (IC50 = 1 nM). The SAR at position 3 was more restricted, although the introduction of electron-donating groups (such as a methyl and methoxy) on the pyridinyl ring yielded potent PIKfyve inhibitors, with IC50 values in the low nM range. The acetylenic moiety was essential for PIKfyve inhibition, and only the saturated ethyl linker displayed potent PIKfyve inhibition, albeit less active than the acetylene counterpart. The compounds were 2- to 5-fold less potent on PIP4K2C relative to PIKfyve. These dual PIKfyve/PIP4K2C inhibitors displayed antiviral activity against both the venezuelan equine encephalitis virus (VEEV) and the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). A screening against a panel of cancer cell lines revealed antitumoral activity, although some of the potent PIKfyve/PIP5K2C inhibitors lacked antitumoral activity. Conclusions: Isothiazolo[4,3-b]pyridines are dual PIKfyve/PIP4K2C inhibitors displaying broad-spectrum antiviral, as well as antitumoral, activity.
