Peanut sprout is a high-quality healthy food, which not only has beneficial effects, but also a higher phenol content than peanut seed. In this study, peanut sprout was treated with five cooking methods, namely boiling, steaming, microwave heating, roasting, and deep-frying, and the phenol content, monomeric phenol composition, and antioxidant activity were determined. The results showed that, compared with unripened peanut sprout, the total phenol content (TPC) and total flavonoid content (TFC) decreased significantly after the five ripening processes, and the highest retention of phenols and flavonoids was associated with microwave heating (82.05% for TPC; 85.35% for TFC). Compared with unripened peanut sprout, the monomeric phenol composition in germinated peanut was variable after heat processing. After microwave heating, except for a significant increase in the cinnamic acid content, no changes in the contents of resveratrol, ferulic acid, sinapic acid, and epicatechin were observed. Furthermore, there was a significant positive correlation of TPC and TFC with 2,2-diphenyl-1-picrylhydrazyl scavenging capacity, 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) scavenging capacity, and ferric ion reducing antioxidant power in germinated peanut, but not with hydroxyl free radical scavenging capacity, in which the main monomer phenolic compounds were resveratrol, catechin, and quercetin. The research results indicate that microwave heating can effectively retain the phenolic substances and antioxidant activity in germinated peanuts, making it a more suitable ripening and processing method for germinated peanuts.

Understanding the characteristics of the balanced nutrient requirements for peanut to achieve target yields is paramount when formulating fertilizer management strategies to increase yields and avoid fertilizer loss. Nutritional requirement estimation models can provide effective alternatives for the estimation of the optimum crop balanced nutrient requirements under varied agricultural conditions which are less time consuming and expensive. In the present study, the quantitative estimation of the optimum crop balanced nutrient requirements of peanut in China were obtained using quantitative evaluation of fertility of tropical soils (QUEFTS) model. The database covered the main agro-ecological region for peanut crops in China between 1993 and 2018. The predicted results of the QUEFTS model indicated that nutrient uptake requirements increased linearly with increasing pod yields until the yields had reached approximately 60% to 70% of the potential pod yields. It was found that with the increasing pod yields during the nutrient linear absorption stage, the plants had required 38.4 kg N, 4.3 kg P, and 14.0 kg K in total to produce 1000 kg of pods, and the corresponding internal efficiencies were 26.0 kg N/kg, 235.0 kg P/kg, and 71.6 kg K/kg, respectively. In addition, the balance rates of the removal nutrient in the pods were determined to be 29.4 kg N, 2.9 kg P, and 4.9 kg K per 1000 kg of pod yield, or approximately 76.5%, 67.4%, and 34.7% of N, P, and K in the total plants, respectively. This study’s field validation experiments verified the applicability and accuracy of the QUEFTS model. Therefore, it was considered to be an effective alternative for the estimation of the optimal balance N, P, and K uptake requirements for peanut crops. These findings will potentially be helpful when making future decisions regarding fertilizer recommendations for peanut crops in China.

High oleic acid peanut (HOAP) is extensively embraced in China because of its high nutritional value and enhanced oxidative stability. However, its dissemination has been severely constrained in high altitude and high latitude areas due to chilling stress during sowing, resulting in significant yield loss in these regions. Despite the lack of understanding of the molecular mechanisms underlying low temperature germination (LTG) in HOAP, discovering the quantitative trait loci (QTL) that confer this trait will undoubtedly benefit breeding efforts. In the present study, we identified putative genomic regions and single nucleotide polymorphisms (SNPs) that govern LTG tolerance of HOAP in an F2 population derived from the cross of chilling-tolerant YH65 and chilling-sensitive FL14 using bulk segregant analysis (BSA). Analysis of ΔSNP-index and Euclidean distance (ED) value association pinpointed the overlapped region to a 2.29 Mb interval on chromosome A05. The candidate interval showed that 122 genes were significantly related to response to abiotic stress and plant–pathogen interaction. Furthermore, an SNP site associated with LTG tolerance was discovered. The SNP site was employed as a Kompetitive Allele Specific PCR (KASP) marker and validated in a universal peanut panel. These findings may provide valuable insight into the molecular mechanism underpinning LTG tolerance and facilitate marker-assisted selective breeding in HOAP.

As one of the major oil crops worldwide, peanuts play a crucial role in ensuring the stability of global oil production and quality. Seed quality, a direct determinant of yield, is influenced by various factors, among which storage temperature and moisture content are critical. However, the mechanisms by which storage conditions affect peanut seedling development and final yield remain unclear. To address this, we conducted field plot experiments using different storage temperature regimes (0 °C, −10 °C, −20 °C, −40 °C) and seed moisture contents (5%, 10%, 15%) to evaluate their effects on seed quality, subsequent growth, and yield. The results showed that, at the same storage temperature, seed vigor declined with increasing seed moisture content. Conversely, at the same seed moisture content, seed vigor decreased with lower storage temperatures. Overall, the highest germination rate (99.21%) and emergence rate (96.79%) were observed under the 0 °C/5% treatment. Nutrient composition analysis revealed that, at a constant storage temperature, protein content was negatively correlated with seed moisture content, whereas linoleic acid content was positively correlated. After sowing, antioxidant enzyme activities in leaves were monitored throughout seedling development. Enzyme activities initially increased and then declined as plants matured. At the early seedling stage, the highest activities of superoxide dismutase (SOD) and peroxidase (POD) were detected under the 0 °C/5% treatment. In contrast, malondialdehyde (MDA) content increased significantly with decreasing storage temperature and increasing seed moisture content. From a yield perspective, these factors collectively influenced yield components under different treatments, with the maximum yield (6187.5 kg/ha) obtained under the 0 °C/5% treatment. In summary, the increase in nutrient content and peroxidase activity during the seedling stage of peanut seeds treated with 0 °C/15% water content improved seed quality and vitality, making seed preservation more suitable under these conditions. On the other hand, we conducted transcriptome sequencing on peanut varieties with different cold tolerance levels and identified a cold tolerance gene AhCOLD1, which was preliminarily validated to be involved in cold stress response. In summary, we have determined the optimal storage method for local peanut seeds and identified a cold resistant gene, providing effective technical support for stabilizing local peanut production.

Introduction: The microbial and mineral C pumps play crucial roles in soil organic C (SOC) stabilization via complicated organo-mineral interactions. Yet, how both of them regulate SOC dynamics during peanut cultivation remains elusive. Methods: The surface soils from five peanut fields of varying ages (0-, 2-, 5-, 10-, and 20-yr.) were selected to elucidate the temporal variations in SOC dynamics and its potential mineral-microbial synergistic regulation patterns, based on PLFAs, eco-enzymatic activity, amino sugars, lignin phenols and Fe oxides analysis, etc. Results: With increasing peanut cultivation ages, SOC, labile C pools, microbial biomass, hydrolase activities, plant‑ and microbial‑derived C (PC and MRC), all exhibited unimodal patterns, peaking at 2-yr. peanut field before subsequently declining. Furthermore, the decreased Cy/Pre (i.e., the ratio of cyclopropyl PLFAs to their monoenoic precursor) and elevated fungi/bacteria values in older peanut fields indicated intensified bacterial physiological stress and a community shift favoring fungal dominance in these fields. Long-term peanut cultivation (i.e., 10 and 20-yr.) decreased SOC lability, enhanced aggregate stability, and altered SOC origins (i.e., enlarged PC but reduced MRC contribution to SOC). As peanut cultivation ages increased, the aggravated soil acidification (evidenced by a decline in pH from 6.10 to 4.57) concurrently enhanced the activation and complexation of Fe oxides, which was attributed to elevated levels of reactive Fe oxides (i.e., Feo and Fep). This transition altered the Fe–C coupling modes from ‘weak’ adsorption-driven (C:Fe molar ratios < 1) in 0- and 2-yr. peanut fields, to ‘strong’ complexation (1 to 6) in the, 5-, 10-, and 20-yr. peanut fields. The PLS-PM and Mantel test further suggested that intensified soil acidification promote reactive Fe oxides accumulation, while labile C pools and microbial biomass positively affect the accumulation of PC and MRC, respectively. Notably, reactive Fe oxides play a significant role only in MRC, rather than in PC accumulation. Discussion: Overall, these results elucidate the coupled mineral‑microbial influences on SOC dynamics over the peanut cultivation chronosequence, thereby enhancing our understanding of SOC sequestration processes in China’s peanut-producing regions.

Peanut (Arachis hypogaea L.) is a globally vital oil and cash crop. However, frequent cold stress severely compromises its yield stability. To address this challenge, we conducted quantitative trait locus (QTL) mapping for two cold tolerance-associated traits, namely relative emergence rate (RER) and relative emergence index (REI), across four distinct environments using a recombinant inbred line (RIL) population derived from the cold-tolerant landrace Silihong and cold-sensitive cultivar Jinonghei 3. Two core QTLs associated with cold tolerance were detected, including qRER6 stable across all environments with PVE of 4.91–5.15% and a co-localized QTL qRER18.1/qREI18.2 on chromosome 18 that governs both target traits with PVE of 14.56–14.71% and 10.34%. Through integrated analysis of QTL mapping and Weighted gene co-expression network analysis (WGCNA), Arahy.657RUG was identified as a candidate cold tolerance gene and designated Ahcold18. Differential expression analysis and heterologous overexpression in Arabidopsis thaliana under freezing stress (−9 °C) showed that Ahcold18 enhances plant survival under low-temperature stress, suggesting a general role in cold tolerance that warrants further investigation in the context of peanut chilling tolerance. A gene-based KASP marker (KASP-2374669), developed from variant sites within Ahcold18, showed preliminary association with RER and REI in the RIL population; however, further validation in diverse germplasm is required to confirm its utility for marker-assisted selection. This study provides a critical genetic resource and a precise technical tool for marker-assisted breeding of cultivated peanut with cold tolerance.
