Genistein (GEN), a natural isoflavone possesses a wide range of pharmacological properties and nutraceutical applications. GEN has been studied for its anticancer activity against different types of cancers, but its use in clinical practice is limited due to its low water solubility, rapid metabolism and excretion, lack of cancer cell targeting and poor bioavailability. In the present study, we investigated folate receptor-targeted and PEGylated poly(lactide-co-glycolide) nanoparticles (PLGA-PEG-FA NPs) containing GEN for targeted delivery to ovarian cancer cells. PLGA-PEG and PLGA-PEG-FA polymer conjugates were synthesized and characterized. Nano-precipitation method was employed for the fabrication of NPs of PLGA, PLGA-PEG and PLGA-PEG-FA containing GEN. GEN containing PLGA-PEG and PLGA-PEG-FA NPs prepared were small (104.17 ± 1.61 and 125.41 ± 3.11 nm, respectively) and exhibited sustained release of GEN for around six days. Folate-decorated PLGA-PEG NPs showed increased cellular uptake in comparison to non-targeted PLGA-PEG NPs. The GEN containing PLGA-PEG-FA NPs showed superior anticancer activity than non-targeted PLGA and PLGA-PEG NPs in folate receptor-overexpressing ovarian cancer cell line, SKOV-3. The IC50 of GEN, GEN encapsulated NPs of PLGA, PLGA-PEG and PLGA-PEG-FA were 51.48, 26.70, 23.43 and 11.98 µg/ml, respectively. Folate-targeted PLGA nanoparticles could be developed for potential target-specific delivery of GEN in the treatment of ovarian cancer.

Since time immemorial natural products have been a great source of medicine to mankind. The anti-viral activities from several ayurvedic herbal medicines (in the form of crude extract or fraction or isolated compounds) have been established but their effectiveness against coronavirus still needs to be explored. They can provide a rich resource of anti-SARS-CoV-2 drug candidates. In this paper, in-silico techniques have been used to identify the potential lead molecules against SARS-CoV-2. A list of flavonoids having anti-viral activity was prepared and evaluated against the selected target. Rhoifolin, 5,7-dimethoxyflavanone-4′-O-β-d-glucopyranoside, baicalin, astragalin, luteolin, and kaempferol showed good binding affinity and thus these could be promising compounds. In-silico screening such as ADMET prediction has been performed which predicted that the selected flavonoids have good pharmacokinetics and pharmacodynamics properties. Molecular dynamics simulation studies and MM-PBSA binding free energy calculations showed luteolin to be a more effective candidate against viral protein Mpro. The novelty of the approach mainly rests in the identification of potent anti-viral natural molecules from natural products flavonoid group of molecules to be effective against the latest coronavirus infection. Background. Sesbania grandiflora has been traditionally used as antidiabetic, antioxidant, antipyretic, and expectorant and in the management of various ailments. Materials and Methods. The study evaluates the antidiabetic activity of methanolic extract of Sesbania grandiflora (MESG) in type 2 diabetic rats induced by low dose streptozotocine and high fat diet. Diabetic rats were given vehicle, MESG (200 and 400 mg/kg, p.o.), and the standard drug, metformin (10 mg/kg), for 28 days. During the experimental period, body weight, abdominal girth, food intake, fasting serum glucose, urine analyses were measured. Insulin tolerance test was carried out on 25th day of drug treatment period. Serum analyses for lipid profile and SGOT and SGPT and serums creatinine, urea, protein, SOD, and MDA were also carried out. At the end of the experiment, animals were euthanized, the liver and pancreas were immediately dissected out, and the ratio of pancreas to body weight and hepatic glycogen were calculated. Results. MESG (200 and 400 mg/kg, p.o.) induced significant reduction () of raised blood glucose levels in diabetic rats and also restored other parameters to normal level. Conclusion. Therefore, it is concluded that MESG has potential antihyperglycemic and antihyperlipemic activities and alleviate insulin resistance conditions. Background. Background: Immunomodulators could alter the activity of the immune system. Most of the synthetic chemotherapeutic agents are immunosuppressants and produce side effects. Plant based immunomodulators are used as general adaptogens and immune strengthners without undesired effects. Present study established the mode of immunostimulatory action of punarnavine alkaloid (PA) isolated from Boerhaavia diffusa Linn. Root by examining various parameters after immunization with SRBC. Methods: PA was separated from B. diffusa and its acute toxicity was studied by up-down regulation method. Immunomodulatory activity of PA was determined in Albino mice by observing its effect on organ weight (liver, spleen, thymus and kidney), expression of cytokines, bone marrow cellularity and alpha-esterase positive cells. Plaque forming assay, delayed type hypersensitivity, and phagocytosis activity were also carried out to support the effect of PA. Results: PA did not exhibit any toxic effect in mice. In delayed type hypersensitivity study, the foot pad thickness due to influx of mononuclear cells at the site of inoculation was distinctly increased in PA treated mice. PA enhanced the phagocytic efficacy of the polymorphonuclear (PMN) cells by causing more engulfment of the Candida cells, thereby stimulating a non-specific immune response. Plaque forming assay confirmed that PA treatment elevated the humoral immune response due to synthesis of antibody which in turn is responsible for the enhancement of macrophages and B-lymphocyte subsets. The significant increase in the number of α-esterase positive cells and bone marrow cellularity due to immunomodulatory effect indicated the proliferation of stem cells. Different organ weight was also markedly improved by PA treatment compared to SRBC sensitized group. Further, in real time PCR studies treatment of Punarnavine significantly increased IL- 7, IL- 10, IL-12a and IL-12b mRNA gene expression. Conclusion: From these results it can be concluded that punarnavine could be developed as a potent immunomodulatory agent.
