Background: Aegle marmelos (L.) Corr. (Rutaceae), commonly known as bael, is used to treat fevers, abdomen pain, palpitation of the heart, urinary troubles, melancholia, anorexia, dyspepsia, diabetes and diarrhea in Indian traditional systems of medicine. The object of the present study was to evaluate the antidiabetic, antihyperlipidemic and antioxidant oxidative stress of umbelliferone β-D-galactopyranoside (UFG) from stem bark of Aegle marmelos Correa. in STZ (streptozotocin) induced diabetic rat. Methods: Diabetes was induced in rat by single intraperitoneal injection of STZ (60 mg/kg). The rat was divided into the following groups; I – normal control, II – diabetic control, III – UFG (10 mg/kg), IV – UFG (20 mg/kg), V – UFG (40 mg/kg), VI – Glibenclamide (10 mg/kg, p.o., once a daily dose). Diabetes was measured by change the level blood glucose, plasma insulin and the oxidative stress were assessed in the liver by estimation of the level of antioxidant markers i.e. superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) and Malondialdehyde (MDA) and antihyperlipidemic effect was measured by estimation of total cholesterol, triglycerides, LDL (low density lipoprotein) cholesterol, HDL (high density lipoprotein) cholesterol, VLDL (very low density lipoprotein) cholesterol. However in a study, the increased body weight was observed and utilization of glucose was in the oral glucose tolerance test. Result: Daily oral administration of different dose of UFG for 28 days showed significantly (P < 0.001) decreased in fasting blood glucose level and improve plasma insulin level as compared to the diabetic control group. Also it significantly (P < 0.001) decreased the level of glycated hemoglobin, glucose-6-phosphatase, fructose-1-6-biphosphate and increased the level of hexokinase. UFG treatment decreased liver MDA and increased the level of SOD, GPx and CAT. UFG treatment of lipids it’s increased the level of cholesterol, triglycerides, VLDL, LDL cholesterol and decreased the level of HDL cholesterol. Histologically, inflammatory cell in blood vessels, intercalated disc, fat degeneration and focal necrosis observed in diabetic rat organ but was less obvious in UFG treated groups. The mechanism of action of UFG may be due to the increased level of pancreatic insulin secretion and effect on the antioxidant marker. Conclusion: UFG posses an antidiabetic, antioxidant and antihyperlipidemic effect on the STZ induced diabetic rat. Hence it could be the better choice to cure the diabetes.

The current perspective for the search of 3-hydroxy-3-methyl-glutaryl-coenzyme A (HMG-CoA) reductase inhibitor has been shifted towards a natural agent also having antioxidant property. Thus, this study was intended to isolate and identify the bioactive compounds from methanolic extract of Ficus virens bark (FVBM) and to evaluate their antioxidant, HMG-CoA reductase inhibitory and hypolipidemic activity. Bioactivity guided fractionation and isolation of bioactive compound from FVBM extract has been done to isolate and characterize the potent HMG-CoA reductase (HMGR) inhibitor with antioxidant activity by using repeated extensive column chromatography followed by spectroscopic methods, including Infrared (IR), 1H & 13C nuclear magnetic resonance (NMR) and Mass spectrum analysis. The in vitro HMGR inhibition and enzyme kinetic assay was determined using HMG-CoA as substrate. In addition, antioxidant activity of the new isolated compound, was measured using 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay and FRAP value. In-silico molecular informatics of HMGR enzyme type inhibition and pharmacokinetics data of the new compound was further evaluated through molecular docking and ADME-T studies. Further, in-vivo hypolipidemic property of FVBM extract and newly isolated compound was also analyzed in triton-WR 1339 induced rats. Thereby, we report the discovery of n-Octadecanyl-O-α-D-glucopyranosyl(6′ → 1″)-O-α-D-glucopyranoside (F18) as a novel HMG-CoA reductase inhibitor with strong antioxidant property. This inhibitor exhibited not only higher free radical scavenging activity but also marked HMG-CoA reductase inhibitory activity with an IC50 value of 84 ± 2.8 ng/ml. This inhibitory activity concurred with kinetic study that showed inhibition constant (Ki) of 84 ng/ml via an uncompetitive mode of inhibition. The inhibition was also corroborated by molecular docking analysis and in silico pharmacokinetics data. The in vivo study revealed that administration of FVBM extract (at higher dose, 100 mg/rat) and the inhibitor (1 mg/rat) to Triton WR-1339-induced hyperlipidemic rats significantly ameliorated the altered levels of plasma lipids and lipoproteins including hepatic HMG-CoA reductase activity; this effect was comparable to the effect of standard drug atorvastatin. The in vitro, in silico and in vivo results clearly demonstrated the antioxidant potential and therapeutic efficacy of the inhibitor as an alternate drug against hyperlipidemia.

Fumaria parviflora Lam. (Fumaraceae) is widely used in traditional as well as folkloric system of medicine from ancient. It is commonly known as ‘Pitpapra’ or ‘Shahtrah’ in Indian traditional system of medicine and used for treating numerous ailments like diarrhea, fever, influenza, blood purifier and other complications. The object of the present study was to evaluate the Antileishmanial, antibacterial, antifungal and cytotoxic potential of isolated compound.

Phytochemical investigation of the ethanolic extracts of the seeds of Carica papaya L. (Caricaceae) led to the isolation of 2,3,4-trihydroxytoluene (caricaphenyl triol) and glyceryl-1-(2’,3’,4’-trihydroxybenzoyl)-2,3-dioleate (papayaglyceride) as the new phytoconstituents along with the known components glyceryl-1-oleiyl-2,3-dilinoleiate, glyceryl-1-oleiyl-2,3-diarachidate, glyceryl-1-linoleiyl-2,3-distearate, carpaine, glyceryl-1,2-dipalmitate, glyceryl trimyristate, glyceryl tristearate, glyceryl-1,2-dipalmityl-3-myristate, glyceryl-1-oleiyl-2,3-dimyristate, β-sitosterol glucoside, glyceryl-1-oleiyl-3-phosphate, glyceryl-1-oleiyl-2-lauryl-3-phosphate and glyceryl-1,2-distearyl-3-phosphate. The structures of all these compounds have been elucidated by spectral data analysis and chemical reactions. The methanolic extract of the seeds and 2,3,4-trihydroxytoluene (200 μg/ml) showed antifungal activity against Aspergillus flavus, Candida albicans and Penicillium citrinium.
