Soil enzymes and microbial communities are key factors in forest soil ecosystem functions and are affected by stand age. In this study, we studied soil enzyme activities, composition and diversity of bacterial and fungal communities and relevant physicochemical properties at 0–10 cm depth (D1), 10–20 cm depth (D2) and 20–30 cm depth (D3) soil layers in 3-(3a), 6-(6a), 12-(12a), 18-(18a), 25-(25a), 32-(32a) and 49-year-old (49a) Chinese fir plantations to further reveal the effects of stand age on soil biotic properties. Spectrophotometry and high-throughput sequencing was used to assess the soil enzyme activity and microbial community composition and diversity of Chinese fir plantation of different stand ages, respectively. We found that soil catalase activity increased as the stand age of Chinese fir plantations increased, whereas the activities of urease, sucrase and β-glucosidase in 12a, 18a and 25a were lower than those in 6a, 32a and 49a. Shannon and Chao1 indices of bacterial and fungal communities first decreased gradually from 6a to 18a or 25a and then increased gradually from 25a to 49a. Interestingly, the sucrase and β-glucosidase activities and the Shannon and Chao1 indices in 3a were all lower than 6a. We found that the relative abundance of dominant microbial phyla differed among stand ages and soil depths. The proportion of Acidobacteria first increased and then decreased from low forest age to high forest age, and its relative abundance in 12a, 18a and 25a were higher than 3a, 32a and 49a, but the proportion of Proteobacteria was opposite. The proportion of Ascomycota first decreased and then increased from 6a to 49a, and its relative abundance in 12a, 18a and 25a was lower than 3a, 6a, 32a and 49a. Our results indicate that soil enzyme activities and the richness and diversity of the microbial community are limited in the middle stand age (from 12a to 25a), which is important for developing forest management strategies to mitigate the impacts of degradation of soil biological activities.

Phytoremediation using woody plants can effectively reduce heavy metal (HM) concentrations in soils. However, the remediation capacity of woody plants depends greatly on plant species and soil environmental conditions. In order to evaluate the HM remediation potential of woody plants from different tailing areas, the HM accumulation characteristics of roots, shoots, and leaves of 12 dominant native woody plants growing in iron and lead-zinc tailing areas were analyzed. The results showed that the concentrations of Cd, As, Ni, Mn, and Cr in most plants in the two tailing areas exceeded the level of normal plants. The distribution of different elements in plants was generally as follows: root > leaf > shoot for Pb and As; root > shoot > leaf for Cr; and leaf > shoot > root for Zn, Ni, and Mn. The distribution of Cu and Cd in plants varied with the type of HM pollution in the two tailing areas. There were significant (p < 0.05) negative correlations between available phosphorus in the soil and Pb, Cd, and Zn in the plant roots when the soil was heavily polluted with Pb, Cd, and As; similarly, there were significant (p < 0.01) negative correlations between readily available potassium in the soil and Pb, Zn, and Ni in plant roots. Based on the higher than average concentration of HMs in plants, and higher bioconcentration factors and translocation factors, some plants were considered woody plant species with phytoremediation. Slash pine (Pinus elliottii) and indian azalea (Rhododendron simsii) had strong enrichment and translocation abilities for Cd, oriental white oak (Quercus glauca) and beautiful sweetgum (Liquidambar formosana) for Mn and paulownia (Paulownia fortunei) for Zn. The plants listed above can be used as potential species for phytoremediation in iron and lead-zinc tailing areas.

The structure and function of forest ecosystems are directly or indirectly affected by their stand density. However, what effect the density of Chinese fir plantations has on the functional diversity of the soil microbial community remains unclear. The microbial metabolic functional diversity of soils sampled at the topsoil (0–20 cm) of 35-year-old Chinese fir plantations of five initial densities (D1: 1667 stems∙hm−2, D2: 3333 stems∙hm−2, D3: 5000 stems∙hm−2, D4: 6667 stems∙hm−2, and D5: 10,000 stems∙hm−2) was studied by using Biolog ECO technology. The results showed that the soil pH, oxidizable organic carbon (SOOC), available N (AN), available P (AP), and available K (AK) contents all showed a gradual increase from D1 to D4 and a decrease from D4 to D5, while the number of culturable bacteria and total microorganisms, the average well color development (AWCD) values for the single carbon substrate and six types of carbon sources used by the microbial community, as well as the Shannon-Wiener diversity index (H’), Pielou evenness index (J), and McIntosh Diversity Index (U), were the opposite, suggesting that low-densities favored C and N mineralization and the nutrient cycle. The density of Chinese fir plantations had a significant effect on the use of carbohydrates, amino acids, carboxylic acids, and phenolic acids by the soil microbial community, but it had no significant effect on the use of polymers (p < 0.05). Principal component analysis (PCA) revealed that carbohydrates, polymers, and phenolic acids were sensitive carbon sources that caused differences in the metabolic functions of soil microbial communities in Chinese fir plantations. Redundancy analysis (RDA) showed that physicochemical factors have a significant influence on the metabolic function of soil microbial communities (RDA1 and RDA2 explained >85% variance). The changes in density affected the soil physicochemical properties, the composition, and the metabolic functional diversity of microbial communities in Chinese fir plantations, which is certainly useful for the stand density regulation of Chinese fir plantations.

Purpose: This research was aimed to screen and identify multifunctional phosphorus-dissolving bacteria of a Chinese fir (Cunninghamia lanceolata) plantation and study its phosphorus-dissolving characteristics in order to provide strain resources and a theoretical basis for developing the appropriate bacterial fertilizer of a Chinese fir plantation. Methods: First, phosphorus-dissolving bacteria were isolated from the woodland soil of a Chinese fir plantation by Pikovskava inorganic phosphorus medium (PVK). Then, some growth-promoting indicators of primary screening strains were determined, including the capacity of phosphorus-solubilized, nitrogenase activity, 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activity, production of indole-3-acetic acid (IAA), secretion of iron carrier and so on. Finally, the screening multifunctional phosphorus-dissolving bacteria were identified, which were combined with colony characteristics, physiological and biochemical tests and molecular biotechnology. Results: (1) Thirteen phosphorus-dissolving bacteria were isolated and screened in total, and P5 (195.61 mg·L−1) had the strongest capacity of phosphorus-solubilized. Five phosphorus-dissolving bacteria were provided with nitrogenase activity, and the highest activity of nitrogenase was P10 and P5 (71.90 C2H4 nmol·mL−1·h−1 and 71.00 C2H4 nmol·mL−1·h−1, respectively). Four strains were provided with ACC deaminase activity, and the highest activity of ACC deaminase was P5 and P9, (0.74 μmol·mg−1·h−1 and 0.54 μmol·mg−1·h−1, respectively). Most strains could secrete IAA, and three strains of bacteria had a strong secretory ability, which could secrete IAA with a concentration greater than 15 mg·mL−1, and P5 was 18.00, P2 was 17.30, P6 was 15.59 (mg·mL−1). P5 produced carriers of iron better than others, and the ratio of the diameter of the iron production carrier ring to the diameter of the colony was 1.80, respectively, which was significantly higher than other strains. Combining all kinds of factors, P5 multifunctional phosphorus-dissolving bacteria were screened for eventual further study. (2) Strain P5 was identified as Burkholderia ubonensis, based on the colony characteristics, physiological and biochemical tests, 16SrDNA sequence analysis and phylogenetic tree construction. Conclusion: P5 has a variety of high-efficiency growth-promoting capabilities, and the ability to produce IAA, ACC deaminase activity and siderophore performance are significantly higher than other strains, which had great potential in the development of microbial fertilizer.

Chinese fir (Cunninghamia lanceolata (Lamb.) Hook.) is an important plantation tree species in China, and seed germination is a key step in forest tree cultivation. To reveal the gene expression network and molecular mechanisms in the germination of Chinese fir seeds, physiological indexes were measured and transcriptome and metabolome analyses were performed on Chinese fir seeds in four stages of germination (imbibition stage, preliminary stage, emergence stage, and germination stage). All six physiological indicators had significant differences at different developmental stages. In transcriptome and metabolome analysis, we identified a large number of differentially expressed genes (DEGs) and differentially accumulated metabolites (DAMs). Gene Ontology (GO) analysis showed a large number of DEGs associated with cell growth, and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis showed that DEGs were significantly enriched in the flavonoid biosynthesis, phenylpropanoid biosynthesis, and plant hormone signal transduction pathways. The KEGG enrichment results of DAMs were similar to those of DEGs. The joint analysis of DEGs and DAMs indicated that flavonoid biosynthesis and phenylpropanoid biosynthesis were the key pathways of Chinese fir seed germination. Our study revealed a number of key genes and key metabolites, laying the foundation for further studies on the gene regulatory network of Chinese fir seed germination.
