Yellow nutsedge (Cyperus esculentus, known as ‘YouShaDou’ in China, YSD) and purple nutsedge (Cyperus rotundus, known as ‘XiangFuZi’ in China, XFZ), closely related Cyperaceae species, exhibit significant differences in triacylglycerol (TAG) accumulation within their tubers, a key factor in carbon flux repartitioning that highly impact the total lipid, carbohydrate and protein metabolisms. Previous studies have attempted to elucidate the carbon anabolic discrepancies between these two species, however, a lack of comprehensive genome-wide annotation has hindered a detailed understanding of the underlying molecular mechanisms. This study utilizes transcriptomic analyses, supported by a comprehensive YSD reference genome, and metabolomic profiling to uncover the mechanisms underlying the major carbon perturbations between the developing tubers of YSD and XFZ germplasms harvested in Yunnan province, China, where the plant biodiveristy is renowned worldwide and may contain more genetic variations relative to their counterparts in other places. Our findings indicate distinct expression patterns of key regulatory genes involved in TAG biosynthesis and lipid droplet formation, including transcriptional factors and structural genes such as ABI3 transcriptional factor, rate-limiting enzymes GPAT3/6/9 and DGAT2/3, and oleosin and caleosin homologs. Furthermore, our omics data suggest that these differences in gene expression are not the sole contributors to the diverse tuber compositions. Instead, complex interactions among highly regulated catalytic reactions, governing carbohydrate, protein, and species-specific metabolite metabolisms, such as starch and sucrose metabolic pathways, flavonoid and amino acids biosynthetic pathways, collectively contribute to the pronounced carbon anabolic differentiation primarily evident in TAG accumulation, as well as the starch properties in mature tubers. This study offers new metabolic insights into the high-value underground non-photosynthetic tissues of Cyperaceae species, which harbors not only high biomass productivity but also abundant nutrients as favorable food or industrial sources in the modern agriculture. The detailed omics analyses aim to deepen our understanding of the Cyperaceae species, which may potentially broaden their application values and facilitate the molecular breeding of better varieties to ameliorate the food safety problem.

Food poisoning caused by potato glycoside alkaloids (SGA) remains a critical factor that affects potato production safety. The potato tuber moth (Phthorimaea operculella) is a notorious pest that displays good adaptability to SGA in potato tissues. Studies that explore the mechanisms underlying SGA homeostasis in potato tuber moth larvae are urgently needed. In this study, ultra-high-performance liquid chromatography (UHPLC)-triple quadrupole mass spectrometry (MS/MS) was applied to detect the dominant SGA substances α-solanine and α-chaconine in potato leaves and PTM larval tissues. From the highest to lowest SGA concentrations, the potato cultivars studied were ranked as follows: DS47, LS6, DS23 and QS9. To exclude the influence of nutrients within different potato varieties, different SGA containing (0%, 0.1%, 0.2%, 0.3% and 0.4%) the artificial diet treatment groups were added. DS47 and 0.3% SGA-containing artificial diets presented the best conditions for PTM growth, development and reproduction compared to other potato cultivars and artificial diet controls. The potato tuber moth larva tissues were dissected and the SGA content within different tissues were detected using an UHPLC machine. The results showed that α-chaconine was dispersed in the feces, midgut, hindgut, head and cuticle, and α-solanine was distributed only in the feces and midgut. Antibiotic-treated insects exhibited higher concentrations of SGA than the normal microbiome group. Furthermore, the SGA concentrations of 100 newly-hatched PTM larvae and puparia were detected, with both of them found to contain small amounts of SGA. The results showed that ecdysis and the excretion process were effective approaches used by the potato tuber moth to equilibrate internal SGA accumulation. The microorganism-decreased SGA concentrations were excited in their gut. SGA may transfer from adults to the next generation, and SGAs in PTM are inheritable. In this study, we demonstrated that the potato tuber moth possessed an effective method to preliminarily decrease high SGA accumulation in potato.

Macadamia integrifolia is an economically important woody oil crop valued for its high monounsaturated fatty acid content and excellent kernel nutritional quality. Although it has great industrial potential, variety-related differences in lipid metabolism and kernel quality are still poorly understood. Resolving these differences will support targeted quality improvement and molecular-breeding efforts. In this study, three representative macadamia cultivars, namely DS1, GR1 and O.C., which are widely cultivated in Yunnan province of China, were employed to investigate the dynamic variations in kernel quality traits across three physiological stages. Integrated transcriptomic, metabolomic, and lipidomic analyses were applied at the kernel development stage to characterize the metabolic differentiation. Multi-omics results revealed distinct carbon allocation and lipid accumulation patterns observed among the three cultivars. Variety-related differences in oil yield and nutrient profiles were associated with altered gene expressions and metabolite levels linked to fatty acid biosynthesis, triacylglycerol (TG) assembly and oil-body formation. Differences in lipid and carbon metabolite accumulation also contributed to these phenotypic variations. This study preliminarily identified candidate genes and pathways associated with TG-centered lipid metabolism in developing macadamia kernels at a critical developmental stage, based on samples collected from single trees per cultivar.
