Zoonotic protozoan parasites in dogs represent a significant public health concern because of their ability to infect humans. This study aimed to detect and determine the prevalence of zoonotic protozoan infections, including Balantidium coli, Entamoeba histolytica, Giardia duodenalis, Cryptosporidium spp., Sarcocystis spp. and Cystoisospora spp. in stray and owned dogs with diarrhea in Trabzon Province, Türkiye. Fecal samples were obtained from 450 dogs of various ages, sexes, and breeds. Samples were preserved in 70% alcohol for molecular analysis and incubated in 2.5% potassium dichromate for microscopic examination. Parasite DNA was analyzed using polymerase chain reaction (PCR) and identified by restriction fragment length polymorphism (RFLP). Microscopic examination identified the Cystoisospora ohioensis group, Cystoisospora canis, and Sarcocystis spp. in 56 samples (12.44%). Specifically, Cystoisospora spp. and Sarcocystis spp. were detected in 36 (8.00%) and 20 (4.44%) of the samples, respectively. A mixed infection involving C. canis and the C. ohioensis group was observed in one sample. Molecular analyses detected Cystoisospora spp. DNA in 36 samples (8.00%) and Sarcocystis spp. DNA in 20 samples (4.44%). RFLP analysis confirmed that all detected Cystoisospora spp. DNA belonged to C. ohioensis, while Sarcocystis spp. could not be differentiated at the species level. This study reports the first microscopic detection of Sarcocystis spp. and C. ohioensis in dogs in Trabzon Province and constitutes the first molecular identification of C. ohioensis in Türkiye.

Amaç: Güncel çalışmada, kedilerin fibrosarkomlarında hücre proliferasyon indeksi, p53 tümor baskılayıcı gen durumu, tümor metastazı, invazyon kapasitesi ile hücresel orjinin ortaya konulması amacıyla Prolifere olan hücre nükleus antijeni (PCNA), Matriks Metalloproteinaz-9 (MMP-9), Vimentin, Alfa-Düz Kas Aktini (α-SMA), S-100, Desmin ve Pan- Sitokeratin (Pan-SK) gibi kanser markerları ve intermedier filamentlerin ekspresyonlarının immunohistokimyasal olarak değerlendirilmesi amaçlanmıştır. Gereç ve Yöntem: Bu çalışmada, Patoloji Anabilim Dalı’na rutin histopatolojik inceleme amacıyla getirilen 7 adet kediden alınan tümoral doku örnekleri kullanılmıştır. Histopatolojik değişikliklerin değerlendirilebilmesi amacıyla kesitlere Hematoksilen & Eozin boyaması uygulandı. İmmunohistokimyasal boyamalarda streptavidin-biotin peroksidaz kompleksi yöntemi kullanıldı. Bulgular: 7 spontane kutanöz fibrosarkom vakasının 5’i evre I, 1’i evre II ve 1’i ise evre III olarak saptandı. Tümöral dokulardaki belirgin kollajen bantlarının varlığı Masson Trikrom boyama ile ortaya konuldu. Vimentin ve α-SMA boyanmaları yönünden tüm vakalar pozitifti. Pan SK, S-100 ve Desmin boyanmaları yönünden tümör hücreleri negatif reaksiyon verdi. Tüm vakalar PCNA yönünden pozitifti. Sadece 2 vaka p53 pozitif boyanma gösterdi. Bu 2 vaka, ileri evrelerdi. Sadece bir fibrosarkom vakasında (Evre III) tümör tipi dev hücrelerinde intrasitoplazmik MMP-9 ekspresyonları tespit edildi. Öneri: Sonuç olarak, çalışmada kullanılan tümör örneklem sayısı az olsa da, PCNA, p53 ve MMP-9’un kedilerin spontane fibrosarkomlarında hücre proliferasyonunun belirlenmesi, malignitenin derecelendirilmesi, tümör agresifliği ve davranışı ile prognozun saptanmasında oldukça kullanışlı markerlar olduğunu ortaya koymuştur.

In this study, intermediate filaments, Pan Cytokeratin (Pan CK), Vimentin, Desmin and S-100 were evaluated to reveal the cellular origin of feline SCCs. Alpha-Smooth Muscle Actin (α-SMA) for cancer-related fibroblasts (CAFs) in the tumor microenvironment, and p53, a tumor suppressor gene, were investigated. Proliferating Cell Nuclear Antigen (PCNA) expression was evaluated for the cell proliferation index. Matrix Metalloproteinase-9 (MMP-9) immunoreactivity was evaluated for the metastasis and invasion capacity. In addition, it is aimed to reveal the expressions of proapoptotic Bax gene, antiapoptotic Bcl-2 gene, caspase-dependent pathway Caspase-3 and caspase-independent pathway Apoptosis Inducing Factor (AIF) for apoptosis mechanism. Biopsy samples taken from 7 cats brought to Department of Pathology for routine histopathological examination were used in this study. Tumor tissue samples were fixed in 10% formaldehyde solution. Serial sections of 5 μm thickness were taken from the paraffin blocks prepared after routine tissue follow-up procedures. Hematoxylin & Eosin (H&E) staining was performed on the sections. Avidin-Biotin Peroxidase-Technique (ABC) was used as immunohistochemical staining. It was determined that the tumors had epithelial-mesenchymal transition, exhibited a very high proliferation index, had p53 mutation, and showed low metastasis/high invasion capacity. It was revealed that Bax/Bcl-2 ratio increased in favor of proapoptotic Bax, and caspase-independent apoptosis was more dominant than caspase-dependent apoptosis.

Background: Mastitis is one of the most prevalent and economically significant diseases in dairy cattle, causing to inflammation of the mammary gland and reduced milk yield. Objectives: This study investigated the therapeutic efficacy of Vitex pseudo‐negundo (VPN) extracts in an experimentally induced rat mastitis model. Methods: Sixty female Wistar rats were randomly assigned into six groups of 10. Group I served as the control group. Mastitis was induced by intramammary injection of Staphylococcus aureus (SA) (1.5 × 10 8 cfu/mL; 10 µL per gland) into the L4, L5, R4 and R5 mammary glands. Group III received intramammary gentamicin (0.25 mg/10 µL/day) for 5 days. Groups IV–VI received VPN extracts—water (0.2603 µg/kg/day), ethanol (0.0803 µg/kg/day) and petroleum ether (0.1042 µg/kg/day), administered at 10 µL per gland for the same duration. Rats were clinically monitored three times daily from the onset of clinical signs until euthanasia. Result: SA infection significantly increased mammary gland weight, total oxidative status (TOS) and bacterial load and induced tissue pathology, while decreasing total antioxidant status (TAS). C‐reactive protein levels remained unchanged. VPN significantly improved mammary gland weight, TOS, TAS, bacterial load and histological parameters ( p < 0.001). Mammary gland weight, oxidative stress, bacterial load and histopathological lesion severity all decreased markedly following treatment. Histological examination showed near‐normal restoration of mammary gland architecture in all treatment groups. Conclusion: Preparations of VPN, particularly the water extract, demonstrated strong potential for restoring altered microbiological, biochemical and histological parameters in this rat mastitis model.
