The NLRP3 inflammasome activation and neuroinflammation are known to be involved in the pathology of depression, whereas autophagy has multiple effects on immunity, which is partly mediated by the regulation of inflammasome and clearance of proinflammatory cytokines. Given the emerging evidence that autophagy dysfunction plays an essential role in depression, it is very likely that autophagy may interact with the inflammatory process in the development and treatment of depression. Salvianolic acid B (SalB), a naturally occurring compound extracted from Salvia miltiorrhiza, contains anti-inflammatory and antidepression properties and has recently been proven to modulate autophagy. In this study, we sought to investigate whether autophagy is involved in the inflammation-induced depression and the antidepressant effects of SalB. The effects of prolonged lipopolysaccharide (LPS) treatment and SalB administration on behavioral changes, neuroinflammation, autophagic markers and NLRP3 activation in rat hippocampus were determined by using behavioral tests, real-time PCR analysis, western blot, and immunostaining. Our data showed that periphery immune challenge by LPS for 2 weeks successfully induced the rats to a depression-like state, accompanied with enhanced expression of pro-inflammatory cytokines and NLRP3 inflammasome activation. Interestingly, autophagic markers, including Beclin-1, and the ratio of LC3II to LC3I were suppressed following prolonged LPS exposure. Meanwhile, co-treatment with SalB showed robust antidepressant effects and ameliorated the LPS-induced neuroinflammation. Additionally, SalB restored the compromised autophagy and overactivated NLRP3 inflammasome in LPS-treated rats. Collectively, these data suggest that autophagy may interact with NLRP3 activation to contribute to the development of depression, whereas SalB can promote autophagy and induce the clearance of NLRP3, thereby resulting in neuroprotective and antidepressant actions.

Purpose: This study aims to investigate the pathogenesis, clinical manifestations, and prognosis of interstitial lung disease (ILD) induced by cetuximab.Patients and methods: A single case of a colorectal cancer patient who underwent radical resection and descending colon-rectal anastomosis for sigmoid colon cancer on October 22, 2020, was selected. Postoperative pathology revealed moderately differentiated adenocarcinoma confined to the mucosa, measuring approximately 0.5×0.2×0.2 cm, with no cancer involvement in the proximal and distal resection margins, and metastatic carcinoma observed in 1 out of 5 pericolic lymph nodes. On July 29, 2021, due to a significant increase in CA19–9 levels, chest and abdominal CT scans, along with brain MRI, indicated enlarged retroperitoneal lymph nodes. The patient was afebrile and without respiratory symptoms such as cough, sputum production, chest tightness, or shortness of breath, and imaging suggested disease progression. With no significant abnormalities in C-reactive protein, blood routine, and no evidence of infection, cetuximab-related ILD was considered. Due to mild inflammation, the patient continued cetuximab combined with chemotherapy. Results: This study suggests that cetuximab can induce ILD, and the onset time may be associated with factors such as serum albumin levels and underlying pulmonary diseases. Conclusion: The incidence of cetuximab-induced ILD is low and relatively rare, with non-specific clinical symptoms. Clinically, treatment plans should be tailored to the patient’s actual condition to control disease progression and improve patient prognosis.

Purpose: This study assesses the cost-effectiveness of rimegepant for the on-demand acute treatment of migraine in the Chinese population, focusing on headache relief within a 2 h timeframe. Utilizing data from Phase III clinical trials on rimegepant involving Asian populations, this analysis aims to provide essential insights for healthcare decision-making in the context of migraine management in China.Patients and methods: Employing a decision tree model, this research evaluates the cost-effectiveness of rimegepant over a concise 2 h period, exclusively considering its direct market price of 219.00 CNY per dose for on-demand, single-use treatment upon approval in China. This model is based on pain relief outcomes from a clinical trial, categorizing health outcomes by the achievement of pain freedom and alleviation from the most bothersome symptom within two hours post-administration.Results: The study unveils that rimegepant adds 0.0018 quality-adjusted life days (QALD) with an incremental cost-effectiveness ratio (ICER) of 122,166.07 CNY/QALD. Against a daily cost-effectiveness threshold derived from the 2023 per capita GDP of China (734.45 CNY/day), rimegepant falls short of proving its cost-effectiveness. A significant price reduction to approximately 1.32 CNY per dose is required for rimegepant to be considered cost-effective within this framework. Furthermore, a series of sensitivity analyses were conducted to validate the robustness of these results. Conclusion: While rimegepant shows clinical efficacy in providing rapid relief from migraine symptoms, its current pricing exceeds the threshold for cost-effectiveness in the Chinese healthcare setting. This study underscores the need for price adjustments to enhance the accessibility and economic viability of new migraine treatments.

A co-factor of multiple metalloenzymes that promote normal physiological functions, such as the immune system, neurological system, human hematological function, bone health, and antioxidant action, copper is one of the most important trace metals in the human body. The copper content in the human body maintains a relatively constant level; too much copper can cause Wilson’s disease, and too little can cause Menkes disease. Copper is involved in the physiological functions of many body systems and has a very complex transport system with many transporters. In order to offer fresh concepts and a theoretical foundation for cancer treatment, this article will go into great detail about the distinct metabolic pathways of copper and the function of cell death, particularly the function of copper-controlled cell death, especially cuproptosis, in cancer detection and treatment. Cuproptosis, a copper-dependent cell death distinct from necrosis, apoptosis, and ferroptosis, is triggered by excessive intracellular copper binding to lipoylated tricarboxylic acid (TCA) cycle proteins, leading to abnormal protein aggregation and loss of iron-sulfur clusters in respiratory chain complexes. Future research may focus on optimizing the specificity of copper metabolism-related biomarkers (eg, CTR1, SOD1) for early tumor screening, developing targeted copper ionophores/chelators with low systemic toxicity, and exploring the crosstalk between copper metabolism and tumor immunity to establish multi-dimensional diagnosis and treatment strategies.
