The significant accumulation of protein-rich agro-industrial waste requires the urgent development of biotechnological methods for its processing and usage. Strains of B. subtilis and B. paralicheniformis were isolated and identified from soil samples obtained from a chicken farm in the Akmola region and the city of Taraz. These strains produce proteases with peak activity at 70 °C and pH 10.5 for B. subtilis, and at 60 °C and pH 9.0 for B. paralicheniformis. The protease, keratinase, and collagenase activity of B. subtilis enzymes are 158.8 ± 2.5 U/mL, 109.3 U/mL, and 10.4 U/mL, respectively. The enzyme values for B. paralicheniformis were 323 ± 4.8 U/mL, 122.6 U/mL, and 23.6 U/mL, respectively. Enzymatic hydrolysis of poultry and livestock waste, including chicken feathers, hooves, and cattle hides, was conducted using the B. paralicheniformis strain. Following 96 hours of treatment, the hydrolysis rates of feathers, hooves, and hides were 100%, 29.6%, and 97.2%, respectively. Examination of the hydrolyzed feathers using scanning electron microscopy revealed the degradation of essential structural components of the feathers and the adherence of bacterial cells to the surface of the feather keratin. The acquired data suggest the potential of the B. paralicheniformis strain as a producer of alkaline proteolytic enzymes and its use in technologies for processing protein-rich agro-industrial waste.

Phytases are industrial enzymes that enhance the availability of phytate-bound phosphorus and can reduce the environmental burden associated with undigested phytate. Bacillus paralicheniformis T7 is a spore-forming strain exhibiting multiple hydrolytic activities, including phytase activity. The phyL gene encoding β-propeller phytase was cloned from B. paralicheniformis T7 and expressed in Escherichia coli, followed by purification and biochemical characterization of the recombinant enzyme—rPhyL. The molecular mass of rPhyL was 41.0 kDa, and maximal activity was observed at pH 8.0 and 60 °C. The enzyme displayed alkaline stability, retaining 90% and 93% of its initial activity after 5 h at pH 8.0 and 10.0, respectively. Following 5 h of incubation at 50 °C and 55 °C, residual activities were 83% and 46%, respectively. rPhyL was strongly Ca2+-dependent, with maximal activity at 0.6–1.0 mM CaCl2. Kinetic analysis using sodium phytate as the substrate yielded an apparent Km of 1.21 mM, an apparent Vmax of 328.6 μM·min−1, and an apparent Ki of 1.34 mM under the specified assay conditions. Under the tested conditions, rPhyL exhibited detectable activity toward sodium phytate, whereas no detectable activity was observed toward ATP, sodium pyrophosphate, β-glycerophosphate, or p-nitrophenyl phosphate. These findings characterize rPhyL as a Ca2+-dependent bacillary β-propeller phytase with alkaline stability, expanding the enzymatic repertoire of the spore-forming B. paralicheniformis T7 strain.

In the present study, two metalloproteases M14 and M42 from a Kazakh strain of Bacillus paralicheniformis T7 with high proteolytic activity were successfully cloned, expressed and purified. Protease genes were amplified by polymerase chain reaction (PCR) using specific primers selected on the basis of full genome sequencing data of B. paralicheniformis strain N7 (GenBank accession #CP124861). The amplified fragments were cloned in the expression vector pET-28c(+). Recombinant analogs of the metalloproteases M14 and M42 were obtained by plasmid expression in Escherichia coli cells of the strain ArcticExpressRP(DE3). Using metal affinity chromatography, rM14 and rM42 proteins were purified and polyclonal antibodies were produced by immunization of rabbits. Using anti-rM14 and anti-rM42 antibodies, the expression level of native metalloproteases M14 and M42 in cell lysate and culture fluid of B. paralicheniformis T7 strain was examined after culturing the strain on keratin-containing media: chicken feather, horn, hoof, hide and wool of cattle. Substrate-dependent expression of metalloprotease genes of B. paralicheniformis strain T7 was established. The obtained results indicate the prospective use B. paralicheniformis strain T7 as a producer of metalloproteases M14 and M42.
