Mongolian oak (Quercus mongolica) is a common building material and landscaping tree species in northern China, with significant economic and ecological value. Its seedling growth is inhibited by high light intensity, but the mechanism by which light stress affects the growth and development of its seedlings remains unclear. In this study, we investigated the phenotypes, physiological processes, and molecular responses of 3-year-old Mongolian oak seedlings under different light treatments: full light (Sck), light shading (S1; 40% light), moderate shading (S2; 20% light), and severe shading (S3; 3% light). Compared to Sck, the S1 and S2 treatments resulted in higher leaf area, photosynthetic pigment content, photosynthesis rates, soluble sugar contents, and soluble protein contents in Mongolian oak seedlings. The S1 and S2 treatments also promoted seedling height and diameter growth and resulted in lower degrees of membrane lipid peroxidation, cell membrane permeability, and antioxidant enzyme activity. In contrast, severe shading (S3) significantly inhibited seedling height and diameter growth due to the lower net photosynthetic rate, and exhibiting higher degrees of membrane lipid peroxidation and cell membrane permeability. Shading treatments (S1 and S2) alleviated the negative effects of strong light on the growth and development of Mongolian oak seedlings, with the S2 treatment having the greatest effect. However, severe shading (S3) inhibited growth and development. A total of 3726 differentially expressed genes (DEGs) were detected in leaves under different shading treatments in RNA sequencing analysis. Among these, 1691, 3150, and 824 DEGs were detected in the Sck-S1, Sck-S2, and S1-S2 comparison groups, respectively. The different shading treatments determined common expression regulation pathways, including carotenoid biosynthesis, photosynthetic antenna proteins, and mitogen activated protein kinase (MAPK) signal transduction. Shading induced increases in gene expression levels in light harvesting complexes, which are related to changes in gene expression in the photosynthetic system, leading to changes in photosynthetic physiology. The expression levels of genes related to reactive oxygen species signal perception and activation enzymes were upregulated in Sck. Together, these findings revealed the response mechanisms of Mongolian oak seedlings to different shading levels at the physiological and molecular levels, providing a scientific basis and technical support for the cultivation and large-scale production of Mongolian oak seedlings.

Elm (Ulmus) species are important components of forest resources with significant ecological and economic value. As tall hardwood trees that are drought-resistant, poor-soil-tolerant, and highly adaptable, Ulmus species are an excellent choice for ecologically protected forests and urban landscaping. Additionally, the bioactive substances identified in the fruits, leaves, bark, and roots of Ulmus have potential applications in the food and medical fields and as raw materials in industrial and cosmetic applications. However, the survival of Ulmus species in the natural environment has been threatened by recurrent outbreaks of Dutch elm disease, which have led to the death of large numbers of Ulmus trees. In addition, severe damage to the natural habitats of some Ulmus species is driving their populations to extinction. Omics technology has become an important tool for the collection, protection, and biological characteristic analysis of Ulmus species and their resources due to its recent advances. This article summarizes the current research and application status of omics technology in Ulmus. The remaining problems are noted, and future research directions are proposed. Our review is aimed at providing a reference for resource conservation of Ulmus and for scientific research into this genus.

Poplar is an important shelterbelt, timber stand, and city tree species that has been the focus of forestry research. The regulatory role of the long non-coding RNA molecule (lncRNA; length > 200 nt) has been a research hotspot in plants. In this study, seedlings of 741 poplar were irradiated with LED blue and white light, and the Illumina HiSeq 2000 sequencing platform was used to identify lncRNAs. |logFC| > 1 and p < 0.05 were considered to indicate differentially expressed lncRNAs, and nine differentially expressed lncRNAs were screened, the target genes of which were predicted, and three functionally annotated target genes were obtained. The differentially expressed lncRNAs were identified as miRNA targets. Six lncRNAs were determined to be target sites for twelve mRNAs in six miRNA families. LncRNAs and their target genes, including lncRNA MSTRG.20413.1-ptc-miR396e-5p-GRF9, were verified using quantitative real-time polymerase chain reaction analysis, and the expression patterns were analyzed. The analysis showed that the ptc-miR396e-5p expression was downregulated, while lncRNA MSTRG.20413.1 and GRF9 expression was upregulated, after blue light exposure. These results indicate that lncRNAs interact with miRNAs to regulate gene expression and affect plant growth and development.
