Identifying the appropriate harvesting stage of jujube fruit is of high interest since the changes in physiological indicators during the fruit maturation may influence the fruit nutritional quality. The present study aimed at assessing the effect of the maturation stages on the nutritional composition and quality of the Chinese jujube variety ‘Dongzao.’ Fresh fruits were harvested at three consecutive maturity stages: white (W), semi-red (SR), and full-red (R) stage, for analyzing a wide range of nutritional constituents. Our results showed significant effects of the maturity stage on the fruit nutritional constituents. Soluble sugars and dietary fibers (mainly lignin and hemicellulose) highly increased in the fruit. Total flavonoids, carotenoids, and saponin contributing to the antioxidant capacity of the fruit were fairly decreased over the maturity stages. Except for citric, ascorbic, and succinic acids, all the organic acids increased in the fruit. Interestingly, jujube fruit is a good source of essential amino acids (26% of the total amino acids), and also gradually accumulated cyclic nucleotides (cAMP and cGMP) during the maturation. From our results, the semi-red stage seems to be the most appropriate for harvesting the jujube fruit in order to preserve its bioactive compounds.

Phytoplasmas are obligate cell wall-less prokaryotic bacteria that primarily multiply in plant phloem tissue. Jujube witches’ broom (JWB) associated with phytoplasma is a destructive disease of jujube (Ziziphus jujuba Mill.). Here we report the complete ‘Candidatus Phytoplasma ziziphi’ chromosome of strain Hebei-2018, which is a circular genome of 764,108-base pairs with 735 predicted CDS. Notably, extra 19,825 bp (from 621,995 to 641,819 bp) compared to the previously reported one complements the genes involved in glycolysis, such as pdhA, pdhB, pdhC, pdhD, ackA, pduL and LDH. The synonymous codon usage bias (CUB) patterns by using comparative genomics analysis among the 9 phytoplasmas were similar for most codons. The ENc-GC3s analysis among the 9 phytoplasmas showed a greater effect under the selection on the CUBs of phytoplasmas genes than mutation and other factors. The genome exhibited a strongly reduced ability in metabolic synthesis, while the genes encoding transporter systems were well developed. The genes involved in sec-dependent protein translocation system were also identified. The expressions of nine FtsHs encoding membrane associated ATP-dependent Zn proteases and Mn-SodA with redox capacity in the Ca. P. ziziphi was positively correlated with the phytoplasma concentration. Taken together, the genome will not only expand the number of phytoplasma species and provide some new information about Ca. P. ziziphi, but also contribute to exploring its pathogenic mechanism.

One of the most important attributes of a genome is genome size, which can to a large extent reflect the evolutionary history and diversity of a plant species. However, studies on genome size diversity within a species are still very limited. This study aims to clarify the variation in genome sizes of Chinese jujube and sour jujube, and to characterize if there exists an association between genome sizes and geographical variation. We measured the genome sizes of 301 cultivars of Chinese jujube and 81 genotypes of sour jujube by flow cytometry. Ten fruit traits, including weight, vertical diameter, horizontal diameter, size, total acids, total sugar, monosaccharide, disaccharide, soluble solids, and ascorbic acid were measured in 243 cultivars of Chinese jujube. The estimated genome sizes of Chinese jujube cultivars ranged from 300.77 Mb to 640.94 Mb, with an average of 408.54 Mb, with the highest number of cultivars (20.93%) falling in the range of 334.787 to 368.804 Mb. The genome size is somewhat different with geographical distribution. The results showed weakly significant positive correlation (p < 0.05) between genome size and fruit size, vertical diameter, horizontal diameter, and weight in the Chinese jujube. The estimated sour jujube genome sizes ranged from 346.93 Mb to 489.44 Mb, with the highest number of genotypes (24.69%) falling in the range of 418.185 to 432.436 Mb. The average genome size of sour jujube genotypes is 423.55 Mb, 15 Mb larger than that of Chinese jujube. There exists a high level of variation in genome sizes within both Chinese jujube cultivars and sour jujube genotypes. Genome contraction may have been occurred during the domestication of Chinese jujube. This study is the first large-scale investigation of genome size variation in both Chinese jujube and sour jujube, which has provided useful resources and data for the characterization of genome evolution within a species and during domestication in plants.

Chinese jujube (Ziziphus jujuba Mill.) is attracting more and more attention worldwide due to their tasty and nutritious fruit with extremely high contents of vitamin C (Vc) and soluble sugar. In order to find out the main factors that influence jujube growth and reproductive adaptability, the phenological periods, vegetative growths, fruiting abilities, and fruit qualities of eight newly released cultivars were compared and comprehensively analyzed in three representative ecological sites of the three main jujube-producing regions including Fuping (Hebei), Taigu (Shanxi), and Alar (Xinjiang) in China. Our results showed that the characteristics of jujube cultivars were significantly affected by soil and meteorological factors. The fruit number per bearing shoot was much more affected by temperature, light, and rainfall. The fruit number per bearing shoot, contents of soluble solids, and soluble sugar and Vc contents in fruits were influenced more by meteorological factors. The content of flavonoids was affected by both soil and meteorological factors. A principal component analysis (PCA) indicated that cultivars suitable for planting in Fuping (Hebei) were Yuhong and Lengbaiyu. Zaocuimi, Fucuimi, and Zaoqiuhong were suitable to be cultivated in Taigu (Shanxi), while Zaocuimi, Yuhong, Yulu, Luzao 2, and Yueguang behaved better in Alar (Xinjiang). This study provides insights of the environmental factors on jujube yield and quality and therefore provides references for highly efficient jujube cultivation.
