The avian leukosis virus subgroup J (ALV-J) belongs to the chicken retrovirus that causes enormous economic losses in the poultry industry. Interferon-stimulated genes (ISGs) are critical for controlling virus infections. Here, we identified 897 type I ISGs induced by interferon-α (IFN-α) in chicken peripheral blood mononuclear cell (PBMC) by RNA-Seq. In addition, we further identified 152 potential anti-ALV-J chicken type I ISGs. Among these potential anti-ALV-J ISGs, chicken cholesterol 25-hydroxylase (chCH25H) was selected for further antiviral mechanism studies in chicken embryo fibroblast cell lines (DF1). The gene chCH25H is located on chromosome 6 and clustered in a distinct group with mammals CH25H in the phylogenetic tree. The core promoter region of chCH25H was located within −75/−1 sequence. We found that chCH25H was induced by chicken IFN-α and ALV-J in DF1 cells. The overexpression of chCH25H significantly inhibited ALV-J replication in DF1 cells at 48 h post infection (hpi). In addition, ALV-J replication was significantly enhanced in the chCH25H- knockout DF1 cells. Furthermore, we demonstrated that chCH25H restricted ALV-J infection through the production of 25-hydroxycholesterol (25HC), rather than type I and II interferon. Our results identified 152 potential anti-ALV-J chicken type I ISGs and revealed that 25HC, the product of chCH25H, could be used as a natural antiviral agent to control ALV-J infection.

The striped catfish, Pangasianodon hypophthalmus is an important freshwater aquaculture species in Southeast Asian countries due to its rapid growth. The growth hormone receptor (GHR) is a significant regulatory factor for the growth axis and has great potential applications in animal genetic breeding. This study aims to characterize the GHRb cDNA of the striped catfish and analyze the distribution of its mRNA. Screening of single nucleotide polymorphisms’ (SNPs) loci and diplotypes was performed to provide basic information for the assisted selection of molecular markers in genetic breeding. The results showed that the GHRb cDNA of striped catfish had 2791 bp, which encoded for 569 amino acids. In a phylogenyic study, the ghrb of the striped catfish was clustered with those of other catfish and they were highly homologous. Quantitative real-time PCR (qRT-PCR) experiments showed that GHRb mRNA was expressed in ten different organs of the striped catfish, with the highest expression level in the liver. Five SNP and a haplotype block were identified in the 3′UTR of the GHRb gene using the direct sequencing of 307 individuals. Three haplotypes were found and four diplotypes were constructed. The association analysis revealed that these polymorphisms were significantly associated with growth traits in the striped catfish (p < 0.05). These polymorphisms will provide a valuable reference for future molecular genetic marker-assisted breeding of striped catfish.

In this study, the compound aquaculture model of red tilapia (O. mossambicus albina × O. urolepis hornorum) and water spinach (Ipomoea aquatica) was used to investigate the effect of water spinach rafts on the water quality, antioxidant system, non-specific immune response, and growth performance of red tilapia and the carbon balance of payments. Red tilapia is characterized by its high adaptability to different production environments and food sources, as one of the most productive fish in aquaculture, and is well accepted in the market due to its nutritional and organoleptic characteristics. The experiment lasted for nine weeks and included two systems: the red tilapia-water spinach raft aquaponics (AP) system with 10% cover ratio with water spinach floating beds, and the aquatic monoculture (AM) system with only red tilapia. The total phosphorus (TP), total nitrogen (TN), and nitrate nitrogen (NO−3-N) in the AM were higher than those in AP from the fifth to ninth week. On the second, third, fifth, and sixth weeks, the ammonia nitrogen (NH4+-N), in the AM was higher than those in the AP. From the seventh week, the pH of the AM was significantly lower than the AP, while the nitrite nitrogen (NO−2-N) was significantly higher than the AP. The water quality index of the AP was better than that of the AM, indicating that water spinach can remove the nutrients from aquaculture water bodies. The average daily gain and specific growth rate (SGR) of fish in AP were higher than those in the AM. The acid phosphatase (ACP), alkaline phosphatase (AKP), and catalase (CAT) activities in the hepatopancreas of red tilapia in the AP were also significantly higher than those in the AM, while the malondialdehyde (MDA) in the AP was lower than the AM. The serum ACP and CAT of red tilapia in the AP were also higher than those in the AM, while the MDA of fish in the AP was lower than the AM. The results showed that both the experimental group and the control group were carbon sources and released greenhouse gases into the atmosphere, but the total carbon emissions of the red tilapia and the water spinach symbiotic system in the experimental group was significantly lower than that of the control group (p < 0.05). These results demonstrated that the application of water spinach rafts in aquaponics can not only improve the water quality, but also improve the growth performance, antioxidant system and non-specific immune responses of red tilapia, while promoting the utilization of organic matter in the aquaculture system, improving the ecological benefits in terms of the carbon income and expenditure.

Hypoxia is a common environmental stressor in aquatic ecosystems, and during the cultivation process, Megalobrama amblycephala is prone to death because it is hypoxia-intolerant, which brings huge economic losses to farmers. The pituitary gland is a crucial endocrine gland in fish, and it is mainly involved in the secretion, storage, and regulation of hormones. In the present study, we compared the transcriptional responses to serious hypoxia in the pituitary gland among hypoxia-sensitive (HS) and hypoxia-tolerant (HT) M. amblycephala and a control group that received a normal oxygen supply (C0). The fish were categorized according to the time required to lose balance during a hypoxia treatment. A total of 129,251,170 raw reads were obtained. After raw sequence filtering, 43,461,745, 42,609,567, and 42,730,282 clean reads were obtained for the HS, HT, and C0 groups, respectively. A transcriptomic comparison revealed 1234 genes that were differentially expressed in C0 vs. HS, while 1646 differentially expressed genes were obtained for C0 vs. HT. In addition, the results for HS vs. HT showed that 367 upregulated and 41 downregulated differentially expressed genes were obtained for a total of 408 differentially expressed genes. A KEGG analysis of C0 vs. HS, C0 vs. HT, and HS vs. HT identified 315, 322, and 219 enriched pathways, respectively. Similar hypoxia-induced transcription patterns suggested that the downregulated DEGs and enriched pathways were related to pathways of neurodegeneration in multiple diseases, pathways in cancer, thermogenesis, microRNAs in cancer, diabetic cardiomyopathy, and renin secretion. However, in the upregulated DEGs, the PI3K-Akt signaling pathway (C0 vs. HS), microRNAs in cancer (C0 vs. HT), and HIF-1 signaling pathway (HS vs. HT) were significantly enriched. There is a lack of clarity regarding the role of the pituitary gland in hypoxic stress. These results not only provide new insights into the mechanism by which pituitary tissue copes with hypoxia stress in M. amblycephala but also offer a basis for breeding M. amblycephala with hypoxia-resistant traits.

Pacific white shrimp (Penaeus vannamei) is one of the main shrimp species cultivated around the world. Despite its high yields and easy handling, water pollution from intensive shrimp cultivation remains a serious problem in China. In this study, a compound aquaculture model of P. vannamei and water spinach (Ipomoea aquatica) was used to investigate the effect of a water spinach floating raft on water quality, antioxidants, non-specific immune response, growth performance, and microbial diversity. The experimental design of this study consisted of two groups with three replicates for each, i.e., control group: aquatic monoculture (AM) system with only P. vannamei; treatment group: P. vannamei-I. aquatica raft aquaponics (AP) system with a 50% cover ratio with a water spinach floating raft. The experiment lasted for seven weeks. The results show that the concentrations of total phosphorus (TP), total nitrogen (TN), nitrate nitrogen (NO3−-N), ammonia nitrogen (NH4+-N), nitrite nitrogen (NO2−-N), and active phosphorus (AP) in the AM group were higher than those in the AP group at different sampling times. The water quality index of the AP group was better than that of the AM group, indicating that water spinach can remove the nutrients from aquaculture water bodies. The average daily gain and survival rate of shrimp in the AP group were higher than those in the AM group. The total antioxidant capacity (T-AOC), catalase (CAT), superoxide dismutase (SOD), malondialdehyde (MDA), and acid phosphatase (ACP) in the AP group were better than those in the AM group. The Shannon–Wiener and Simpson indices of the gut, water, and sediment of the AP system were significantly higher than those in the AM system, which implied a higher abundance of microorganisms in the AP system. These results demonstrate that the application of a water spinach floating raft in aquaponics can not only improve the water quality, but also improve the growth performance, antioxidant system, and non-specific immune responses of Pacific white shrimp, while increasing the abundance of microorganisms in the aquaculture system and improving the ecological benefits in terms of the expenditure.
