Background: Catalpa bungei is an important tree species used for timber in China and widely cultivated for economic and ornamental purposes. A high-density linkage map of C. bungei would be an efficient tool not only for identifying key quantitative trait loci (QTLs) that affect important traits, such as plant growth and leaf traits, but also for other genetic studies. Results: Restriction site-associated DNA sequencing (RAD-seq) was used to identify molecular markers and construct a genetic map. Approximately 280.77 Gb of clean data were obtained after sequencing, and in total, 25,614,295 single nucleotide polymorphisms (SNPs) and 2,871,647 insertions-deletions (InDels) were initially identified in the genomes of 200 individuals of a C. bungei (7080) × Catalpa duclouxii (16-PJ-3) F1 population and their parents. Finally, 9072 SNP and 521 InDel markers that satisfied the requirements for constructing a genetic map were obtained. The integrated genetic map contained 9593 pleomorphic markers in 20 linkage groups and spanned 3151.63 cM, with an average distance between adjacent markers of 0.32 cM. Twenty QTLs for seven leaf traits and 13 QTLs for plant height at five successive time points were identified using our genetic map by inclusive composite interval mapping (ICIM). Q16–60 was identified as a QTL for five leaf traits, and three significant QTLs (Q9–1, Q18–66 and Q18–73) associated with plant growth were detected at least twice. Genome annotation suggested that a cyclin gene participates in leaf trait development, while the growth of C. bungei may be influenced by CDC48C and genes associated with phytohormone synthesis. Conclusions: This is the first genetic map constructed in C. bungei and will be a useful tool for further genetic study, molecular marker-assisted breeding and genome assembly.

Clones of Catalpa bungei and Catalpa fargesii f. duclouxii were studied over several years in central China to explore genetic variation in growth traits and to identify clones of high wood yield and high stability. The genetic parameters for height, diameter at breast height (DBH), and stem volume of clones, were estimated. The effect of clone × year on the increment of stem volume in the two species was analyzed by genotype and genotype × environment (GGE) biplot methods. Significant differences in growth traits among clones and between species were found. The growth of C. bungei exceeded that of C. fargesii f. duclouxii after 4 years. Furthermore, from the 5th year, the repeatability and genetic variation coefficient (GCV) of the C. bungei clones were higher than those of the C. fargesii f. duclouxii clones in most cases. The phenotypic variation coefficient (PCV) of the C. fargesii f. duclouxii clones was significantly lower than that of the C. bungei clones. The repeatability of stem volume was intermediate or high in the two species. ANOVA revealed significant effects of the clone by year interaction in these two species. GGE biplot analysis revealed that wood yield and stability were largely independent in C. bungei; clones 22-03, 19-27, and 20-01 were the optimal clones in this species. In contrast, the optimal clones 63 and 128 of C. fargesii f. duclouxii combined the desired characteristics of high yield and high stability. In conclusion, our results indicated that the height and stem volume of C. bungei was under strong genetic control, whereas that of C. fargesii f. duclouxii was influenced by the environment more than by genetic effects. Genetic improvement by clone selection can be expected to be effective, as the repeatability of stem volume was high. Francis and Kannenberg’s method and GGE biplot analysis were used in combination to evaluate the clones. C. bungei clone 22-03 and C. fargesii f. duclouxii clones 63 and 128 were identified as the optimal clones, which exhibited both a high increment of stem volume and high stability.

Catalpa bungei is an economically important tree with high-quality wood and highly valuable to the study of wood formation. In this work, the xylem microstructure of C. bungei tension wood (TW) was observed, and we performed transcriptomics, proteomics and Raman spectroscopy of TW, opposite wood (OW) and normal wood (NW). The results showed that there was no obvious gelatinous layer (G-layer) in the TW of C. bungei and that the secondary wall deposition in the TW was reduced compared with that in the OW and NW. We found that most of the differentially expressed mRNAs and proteins were involved in carbohydrate polysaccharide synthesis. Raman spectroscopy results indicated that the cellulose and pectin content and pectin methylation in the TW were lower than those in the OW and NW, and many genes and proteins involved in the metabolic pathways of cellulose and pectin, such as galacturonosyltransferase (GAUT), polygalacturonase (PG), endoglucanase (CLE) and β-glucosidase (BGLU) genes, were significantly upregulated in TW. In addition, we found that the MYB2 transcription factor may regulate the pectin degradation genes PG1 and PG3, and ARF, ERF, SBP and MYB1 may be the key transcription factors regulating the synthesis and decomposition of cellulose. In contrast to previous studies on TW with a G-layer, our results revealed a change in metabolism in TW without a G-layer, and we inferred that the change in the pectin type, esterification and cellulose characteristics in the TW of C. bungei may contribute to high tensile stress. These results will enrich the understanding of the mechanism of TW formation.

Morus alba L. is an important tree species planted widely in China because of its economic value. In this report, we investigated the influence of two arbuscular mycorrhizal fungal (AMF) species, Glomus mosseae and Glomus intraradices, alone and together, on the growth of M. alba L. seedlings under greenhouse conditions. The growth parameters and physiological performance of M. alba L. seedlings were evaluated 90 days after colonization with the fungi. The growth and physiological performance of M. alba L. seedlings were significantly affected by the AMF species. The mycorrhizal seedlings were taller, had longer roots, more leaves and a greater biomass than the non-mycorrhizae-treated seedlings. In addition, the AMF species-inoculated seedlings had increased root activity and a higher chlorophyll content compared to non-inoculated seedlings. Furthermore, AMF species colonization increased the phosphorus and nitrogen contents of the seedlings. In addition, simultaneous root colonization by the two AMF species did not improve the growth of M. alba L. seedlings compared with inoculation with either species alone. Based on these results, these AMF species may be applicable to mulberry seedling cultivation.

The tree species Pinus tabulaeformis Carr. (P. tabulaeformis) is commonly planted in China due to its economic and ecological value. In order to identify one or more ectomycorrhizal (ECM) fungal species for future P. tabulaeformis afforestation, we investigated the effects of five ECM fungal species: Laccaria laccata, Boletus edulis, Gomphidius viscidus, Suillus grevillei, and Suillus luteus on the growth of P. tabulaeformis seedlings under greenhouse conditions. The growth parameters of P. tabulaeformis seedlings were evaluated 90 days following fungal colonisation. The majority of seedlings were significantly affected by ECM inoculation. Mycorrhizal inoculated seedlings were taller, had more lateral roots, and a greater biomass compared with the non-mycorrhizal (CK) seedlings. With the exception of G. viscidus, inoculated seedlings exhibited higher phosphorus, potassium, and nitrogen content compared with the CK seedlings. In addition, ECM colonisation increased the enzymatic activity of catalase, acidic phosphatase, protease, and the urease content in the rhizosphere soil. Our study showed that Laccaria laccata, Suillus grevillei, and Suillus luteus may be useful for improving the growth and cultivation of P. tabulaeformis seedlings. Furthermore, we observed that S. luteus inoculation increased the gas exchange parameters of P. tabulaeformis seedlings under field conditions.

Etiolation (a process of growing plants in partial or complete absence of light) promotes adventitious root formation in tetraploid black locust (Robinia pseudoacacia L.) cuttings. We investigated the mechanism underlying how etiolation treatment promotes adventitious root formation in tetraploid black locust and assessed global transcriptional changes after etiolation treatment. Solexa paired-end sequencing of complementary DNAs (cDNAs) from control (non-etiolated, NE) and etiolated (E) samples resulted in 107,564 unigenes. In total, 52,590 transcripts were annotated and 474 transcripts (211 upregulated and 263 downregulated) potentially involved in etiolation were differentially regulated. These genes were associated with hormone metabolism and response, photosynthesis, signaling pathways, and starch and sucrose metabolism. In addition, we also found significant differences of phytohormone contents, activity of following enzymes i.e., peroxidase, polyphenol oxidase and indole acetic acid oxidase between NE and E tissues during some cottage periods. The genes responsive to etiolation stimulus identified in this study will provide the base for further understanding how etiolation triggers adventitious roots formation in tetraploid black locus.
