Particulate matter (PM), a major air pollutant in Beijing in recent years, poses a formidable public health threat. Even through many studies have documented the chemical and biological characteristics of PM, less is known about these characteristics on hazardous haze days (Air Quality Index, AQI 301–500) and the difference with sunny or unhealthy haze day (AQI 151–200) characteristics. Herein, studies were performed during a red alert air pollution event (continuous hazardous haze days) and the first few days following the event (sunny days first and then unhealthy haze days) in Beijing from December 19 to 25, 2016. A laser particle counter and an ANDERSEM-6 sampler were used to study the concentration and size distributions dynamics of the PM and the culturable airborne bacteria and fungi, respectively. PM2.5 was sampled by a high-volume air sampler and the chemical compositions, bacterial and fungal community structures, and endotoxin levels were analyzed. The results showed that the PM concentrations on the hazardous haze days and unhealthy haze days were 10.7 and 8.0 times higher, respectively, than those on the sunny days. The chemical composition of PM2.5 was highly correlated with the AQI. The concentration and percentage of water-soluble inorganic ions (WSII), which dominated the PM2.5 constituents, as well as the levels of endotoxin were higher on hazardous haze days than on unhealthy haze days and sunny days. Interestingly, the abundances of bacteria and fungi demonstrated the following order: unhealthy haze days> sunny days> hazardous haze days. Most culturable bacteria and fungi were distributed in particles with aerodynamic diameters of 2.1–4.7 μm. Redundancy analysis found total organic carbon explained 30.0% and 27.1% of total variations in bacterial composition and fungal composition at the genera level, respectively. Our results facilitate a better understanding of the biological and chemical composition dynamics of PM in Beijing.

Ebola virus emerged in West Africa in December 2013. The high population mobility and poor public health infrastructure in this region led to the development of the largest Ebola virus disease (EVD) outbreak to date. On September 26, 2014, China dispatched a Mobile Biosafety Level-3 Laboratory (MBSL-3 Lab) and a well-trained diagnostic team to Sierra Leone to assist in EVD diagnosis using quantitative real-time PCR, which allowed the diagnosis of suspected EVD cases in less than 4 hours from the time of sample receiving. This laboratory was composed of three container vehicles equipped with advanced ventilation system, communication system, electricity and gas supply system. We strictly applied multiple safety precautions to reduce exposure risks. Personnel, materials, water and air flow management were the key elements of the biosafety measures in the MBSL-3 Lab. Air samples were regularly collected from the MBSL-3 Lab, but no evidence of Ebola virus infectious aerosols was detected. Potentially contaminated objects were also tested by collecting swabs. On one occasion, a pipette tested positive for EVD. A total of 1,635 suspected EVD cases (824 positive [50.4%]) were tested from September 28 to November 11, 2014, and no member of the diagnostic team was infected with Ebola virus or other pathogens, including Lassa fever. The specimens tested included blood (69.2%) and oral swabs (30.8%) with positivity rates of 54.2% and 41.9%, respectively. The China mobile laboratory was thus instrumental in the EVD outbreak response by providing timely and reliable diagnostics. The MBSL-3 Lab significantly contributed to establishing a suitable laboratory response capacity during the emergence of EVD in Sierra Leone. The China mobile laboratory was thus instrumental in the EVD outbreak response by providing timely and accurate diagnostics. Therefore, the MBSL-3 Lab played a significant role in establishing a suitable laboratory response capacity during the emergence of EVD in Sierra Leone.

Bioaerosols from swine confinement buildings (SCBs) pose a challenge to public health, and microorganisms within the SCBs bioaerosols originate from swine feces, of which the microbial composition is associated with the production phase. The present study adopted the whole metagenome shotgun sequencing approach, to assess the effects of the production phase on the composition and functional potential of microbial populations in SCBs bioaerosols. Most annotated proteins were assigned into domain bacteria, within which the predominant phylum was Firmicutes. The taxonomical profiles of bioaerosols from different types of piggeries showed that buildings housing weaning piglets (WP) exhibited higher abundances of Bacteroidetes and Proteobacteria than buildings housing finishing pigs (FP), gestating sows (GS), farrowing sows (FS), and breeding boars (BB). Regarding the functional potential, the WP bioaerosol had more genes involved in the protein turnover and fewer genes involved in the carbohydrate metabolism than bioaerosols from other types of SCBs. Furthermore, production phase influenced the antibiotic resistance genes (ARGs) profile of the SCBs bioaerosols. Bioaerosol microbiome of BB, shared a high similarity with GS, and WP bioaerosol microbiome was more similar to FP than other types of SCBs. Our study suggests that the production phase plays a key role in the SCBs bioaerosol microbiome.

Timely detection of highly infectious pathogens is essential for preventing and controlling public health risks. However, most traditional testing instruments require multiple tedious steps and ultimately testing in hospitals and third-party laboratories. The sample transfer process significantly prolongs the time to obtain test results. To tackle this aspect, a portable fiber optic surface plasmon resonance (FO-SPR) device was developed for the real-time detection of infectious pathogens. The portable device innovatively integrated a compact FO-SPR sensing component, a signal acquisition and processing system, and an embedded power supply unit. A gold-plated fiber is used as the FO-SPR sensing probe. Compared with traditional SPR sensing systems, the device is smaller size, lighter weight, and higher convenience. To enhance the detection capacity of pathogens, a monolayer graphene was coated on the sensing region of the FO-SPR sensing probe. Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) was used to evaluate the performance of the portable device. The device can accurately detect the SARS-CoV-2 spike S1 protein in phosphate-buffered saline (PBS) and artificial saliva within just 20 min, and the device successfully detected cultured SARS-CoV-2 virus. Furthermore, the FO-SPR probe has long-term stability, remaining stable for up to 8 days. It could distinguish between the SARS-CoV-2 spike protein and the MERS-CoV spike protein. Hence, this FO-SPR device provides reliable, rapid, and portable access to test results. It provides a promising point-of-care testing (POCT) tool for on-site screening of infectious pathogens.

Influenza virus is a serious threat to global human health and public health security. There is an urgent need to develop new anti-influenza drugs. Lentinan (LNT) has attracted increasing attention in recent years. As potential protective agent, LNT has been shown to have anti-tumor, anti-inflammatory, and antiviral properties. However, there has been no further research into the anti-influenza action of lentinan in vivo, and the mechanism is still not fully understood. In this study, the anti-influenza effect and mechanism of Lentinan were studied in the Institute of Cancer Research (ICR) mouse model. The results showed that Lentinan had a high degree of protection in mice against infection with influenza A virus, delayed the emergence of clinical manifestations, improved the survival rate of mice, significantly prolonged the middle survival days, attenuated the weight loss, and reduced the lung coefficient of mice. It alleviated the pathological damage of mice infected with the influenza virus and improved blood indices. Lentinan treatment considerably inhibited inflammatory cytokine (TNF-α, IL-1β, IL-4, IL-5, IL-6) levels in the serum and lung and improved IFN-γ cytokine levels, which reduced cytokine storms caused by influenza virus infection. The underlying mechanisms of action involved Lentinan inhibiting the inflammatory response by regulating the TLR4/MyD88 signaling pathway. This study provides a foundation for the clinical application of Lentinan, and provides new insight into the development of novel immunomodulators.

Background: Influenza A virus (IAV) continues to pose serious threats to public health. The current prophylaxis and therapeutic interventions for IAV requires frequent changes due to the continuous antigenic drift and antigenic shift of IAV. Emerging evidence indicates that the host microRNAs (miRNAs) play critical roles in intricate host-pathogen interaction networks. Cellular miRNAs may directly target virus to inhibit its infection and be developed as potential anti-virus drugs. Methods: In this study, we established a broad-spectrum anti-IAV miRNA screening method using miRanda software. The screened miRNAs were further verified by luciferase assay, viral protein expression assay and virus replication assay. Results: Five cellular miRNAs (miR-188-3p, miR-345-5p, miR-3183, miR-15-3p and miR-769-3p), targeting 99.96, 95.31, 92.9, 94.58 and 97.24% of human IAV strains recorded in NCBI, respectively, were chosen for further experimental verification. Finally, we found that miR-188-3p downregulated PB2 expression at both mRNA and protein levels by directly targeted the predicted sites on PB2 and effectively inhibited the replication of IAV (H1N1, H5N6 and H7N9) in A549 cells. Conclusions: This is the first report screening cellular miRNAs that broad-spectrum inhibiting IAV infection. These findings suggested that cellular miR-188-3p could be used for RNAi-mediated anti-IAV therapeutic strategies.
