Diabetes mellitus (DM) is a chronic inflammatory disease, and the rapidly increasing DM is becoming a major problem of global public health. Traditional Chinese medicine (TCM) has a long history of treating diabetes. It has been developed and utilized because of its good efficacy and no toxic side effects. Lobeliae chinensis is a traditional whole grass herbal. With the continuous deepening of pharmacological research on TCM, the active ingredients of Lobeliae chinensis are continuously revealed, which contained the alkaloids, flavonoids, flavonoid glycosides and amino acids that have the good effects of anti-inflammatory, anti-viral and anti-diabetic. In order to further explore the targets of active ingredients and its anti-diabetic mechanism, a feasible network pharmacology analysis model based on chemical, pharmacokinetic and pharmacological data was developed by network construction method to clarify the anti-diabetic mechanism of Lobeliae chinensis. The present study conducted by gas chromatography-mass spectrometer (GC/MS), which identified 208 metabolites of Lobeliae chinensis, of which 23 ingredients may have effective pharmacological effects after ADME screening. Network pharmacological analysis on the active ingredients revealed that 5-Hydroxymethylfurfural in Lobeliae chinensis affects the insulin resistance signaling pathway by acting on GSK3B, TNF, and MAPK1, acacetin affects the diabetic pathway by acting on INSR, DPP4, GSK3B, that regulate type 2 diabetes, non-insulin-dependent diabetes mellitus and inflammatory diseases. These results successfully indicated the potential anti-diabetic mechanism of the active ingredients of Lobeliae chinensis.

Bombyx mori bidensovirus (BmBDV), which had been assigned to Densovirinae in Parvoviridae previously, replicates mainly in silkworm midgut olumnar cells and causes the fatal flacheric disease. In contrast to parvovirus, this virus possesses two single-stranded DNA genome segments and encodes a putative protein-primed DNA polymerase. The accumulating evidence suggests that it has unique characteristics different to parvovirus and adopts its own mechanisms for replication. So far, little is known about the replication mechanisms of BmBDV. In this review, we focus on the pathology associated with this virus and the viral biology such as viral genome structure, viral genes, and viral replication and expression strategies.

Objective:Bombyx mori parvo-like virus is a type of virus containing two single-stranded linear DNA molecules (VD1, VD2). In the present work, the structural proteins of B. mori parvo-like virus (China Zhenjiang isolate) (BmDNV-Z) were identified. Methods: The virions were purified by cesium chloride density gradient centrifugation from the feces of B. mori larvae infected with BmDNV-Z and analyzed by SDS-PAGE, Western blotting and MALDI-TOF/mass spectometry. Results: There were seven polypeptides of the virions, named P1 to P7, which reacted with the anti-virion serum. In addition, two polypeptides, P5 and P6, were recognized by the antibody against a viral protein encoded by VD1-ORF3. Conclusion: The results showed that the viral polypeptides were mainly composed of 7 proteins named P1 to P7. Western blotting with anti-virion serum indicated that structural proteins P5, P6 and P7 had molecular weights of around 50, 55 and 130 kDa, respectively. They were the major immunogens of the virion. Among them, P5 and P6 were coded by VD1-ORF3 and P7 was encoded by VD2-ORF1. Taken together, three viral structural proteins encoded by VD1-ORF3 and VD2-ORF1 were identified in BmDNV-Z for the first time.

Background: The basic helix-loop-helix (bHLH) transcription factors and their homologs form a superfamily that plays essential roles in transcriptional networks of multiple developmental processes. bHLH family members have been identified in over 20 organisms, including fruit fly, zebrafish, human and mouse. Result: In this study, we conducted a genome-wide survey for bHLH sequences, and identified 57 bHLH sequences encoded in complete genome sequence of the ponerine ant, Harpegnathos saltator. Phylogenetic analysis of the bHLH domain sequences classified these genes into 38 bHLH families with 23, 14, 10, 1, 8 and 1 members in group A, B, C, D, E and F, respectively. The number of PabHLHs (ponerine ant bHLHs) with introns is higher than many other insect species, and they are found to have introns with average lengths only inferior to those of pea aphid. In addition, two H. saltator bHLHs named PaCrp1 and PaSide locate on two separate contigs in the genome. Conclusions: A putative full set of PabHLH genes is comparable with other insect species and genes encoding Oligo, MyoRb and Figα were not found in genomes of all insect species of which bHLH family members have been identified. Moreover, in-family phylogenetic analyses indicate that the PabHLH genes are more closely related with Apis mellifera than others. The present study will serve as a solid foundation for further investigations into the structure and function of bHLH proteins in the regulation of H. saltator development.

The basic helix-loop-helix (bHLH) proteins play essential roles in a wide range of developmental processes in higher organisms. bHLH family members have been identified in over 20 organisms, including fruit fly, zebrafish, and human. This study identified 54 bHLH family members in the pea aphid, Acyrthosiphon pisum (Harris) (Hemiptera: Aphididae), genome. Phylogenetic analyses revealed that they belong to 37 bHLH families with 21, 13, 9, 1, 9, and 1 members in group A, B, C, D, E, and F, respectively. Through in-group phylogenetic analyses, all of the identified A. pisum bHLH members were assigned into their correspondent bHLH families with confidence, among which 51 were defined according to phylogenetic analyses with orthologs from Drosophila melanogaster Meigen (Diptera: Drosophilidae), and 3 of them were defined according to phylogenetic analyses with orthologs from Bombyx mori L. (Lepidoptera: Bombycidae) and Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae). Analyses on genomic coding regions revealed that the number and average length of introns in A. pisum bHLH motifs are higher than those in other insects. The present study provides useful background information for future studies on structure and function of bHLH proteins in the regulation of A. pisum development.
