Due to soil changes, high density planting, and the use of straw-returning methods, wheat common root rot (spot blotch), Fusarium crown rot (FCR), and sharp eyespot (sheath blight) have become severe threats to global wheat production. Only a few wheat genotypes show moderate resistance to these root and crown rot fungal diseases, and the genetic determinants of wheat resistance to these devastating diseases are poorly understood. This review summarizes recent results of genetic studies of wheat resistance to common root rot, Fusarium crown rot, and sharp eyespot. Wheat germplasm with relatively higher resistance are highlighted and genetic loci controlling the resistance to each disease are summarized.

Background: Drought-resistant varieties are an important way to address the conflict between wheat’s high water demand and the scarcity of water resources in the North China Plain (NCP). Drought stress impacts many morphological and physiological indicators in winter wheat. To increase the effectiveness of breeding drought-tolerant varieties, choosing indices that can accurately indicate a variety’s drought resistance is advantageous. Results: From 2019 to 2021, 16 representative winter wheat cultivars were cultivated in the field, and 24 traits, including morphological, photosynthetic, physiological, canopy, and yield component traits, were measured to evaluate the drought tolerance of the cultivars. Principal component analysis (PCA) was used to transform 24 conventional traits into 7 independent, comprehensive indices, and 10 drought tolerance indicators were screened out by regression analysis. The 10 drought tolerance indicators were plant height (PH), spike number (SN), spikelet per spike(SP), canopy temperature (CT), leaf water content (LWC), photosynthetic rate (A), intercellular CO2 concentration (Ci), peroxidase activity (POD), malondialdehyde content (MDA), and abscisic acid (ABA). In addition, through membership function and cluster analysis, 16 wheat varieties were divided into 3 categories: drought-resistant, drought weak sensitive, and drought-sensitive.Conclusion: JM418, HM19,SM22, H4399, HG35, and GY2018 exhibited excellent drought tolerance and, therefore, can be used as ideal references to study the drought tolerance mechanism in wheat and breeding drought-tolerant wheat cultivars.

Leaf rust, caused by Puccinia triticina , is one of the most destructive fungal diseases in wheat production worldwide. The hypersensitive reaction (HR) is an important defence response against P. triticina infection. In this study, the physiological races 165 and 260 of P. triticina were combined with a line derived from the bread wheat cultivar Thatcher with the leaf rust resistance locus Lr26 to form compatible and incompatible combinations, respectively. Based on an RNA‐Seq database of the interaction systems, a new wheat cysteine‐rich receptor‐like kinase gene, TaCRK2 , is specifically induced and up‐regulated in the incompatible combination. We identified that TaCRK2 was regulated in a Ca 2+ ‐dependent manner. Knockdown of TaCRK2 by virus‐induced gene silencing and RNAi leads to a dramatic increase in HR area and the number of haustorial mother cells at the single infection site. In addition, urediniospores, a P. triticina‐ specific pathogenic marker in compatible combinations, were observed on leaf surfaces of silenced plants at approximately 15 days after inoculation in the incompatible combination. Moreover, transcription levels of TaPR1, TaPR2 , and TaPR5 were obviously reduced in TaCRK2 ‐silenced plants. TaCRK2 overexpression in Nicotiana benthamiana induced strong HR‐like cell death. Finally, transient expression of green fluorescent protein fused with TaCRK2 in N. benthamiana indicated that TaCRK2 localizes in the endoplasmic reticulum. Thus, TaCRK2 plays an important role in the resistance to P. triticina infection and has a positive regulation effect on the HR cell death process induced by P. triticina .

Drought stress, a major abiotic stress, significantly affects wheat (Triticum aestivum L.) production globally. To identify genes and metabolic pathways crucial for responding to short-term drought stress, we conducted transcriptomic and metabolomic analyses of winter wheat cultivar Jimai 418 at four developmental stages: jointing (GS31), booting (GS45), anthesis (GS65), and 8 days after anthesis (DAA8). Transcriptomic analysis identified 14,232 differentially expressed genes (DEGs) under drought stress compared to the control. Specifically, 1387, 4573, 7380, and 892 DEGs were identified at the four developmental stages, respectively. Enriched pathways associated with these DEGs included plant hormone signal transduction, mitogen-activated protein kinase (MAPK) signaling, galactose metabolism, and starch and sucrose metabolism. Totals of 222, 633, 358, and 38 differentially accumulated metabolites (DAMs) were identified at the four stages, respectively. Correlation analysis of both datasets revealed DEGs and DAMs associated with plant hormone signal transduction, arginine and proline metabolism, ABC transporters, and amino acid biosynthesis. These findings offer significant insights into Jimai 418’s molecular response to short-term drought stress. The identified DEGs, DAMs, and enriched pathways contribute to our understanding of wheat drought tolerance. This research will facilitate further investigations into drought tolerance mechanisms and guide the breeding of wheat varieties with enhanced drought resistance.

Flag leaves, plant height (PH), and spike-related traits are key determinants contributing to yield potential in wheat. In this study, we developed a recombinant inbred line (RIL) population with 94 lines derived from the cross between ‘AS985472’ and ‘Sumai 3.’ A genetic map spanned 3553.69 cM in length were constructed using 1978 DArT markers. Severn traits including flag leaf width (FLW), flag leaf length (FLL), PH, anthesis date (AD), spike length (SL), spikelet number spike (SNS), and spike density (SD) were evaluated against this RIL population under three different environments. Combined phenotypic data and genetic map, we identified quantitative trait loci (QTL) for each trait. A total of four major and stably expressed QTLs for FLW, PH, and SD were detected on chromosomes 2D and 4B. Of them, the major QTLs individually explained 10.10 – 30.68% of the phenotypic variation. QTLs with pleiotropic effects were identified on chromosomes 4A and 6D as well. Furthermore, the genetic relationships between seven yield-related traits were detected and discussed. A few genes related to leaf growth and development at the interval of a major locus for FLW on chromosome 2D were predicated. Overall, the present study provided useful information for understanding the genetic basis of yield-related traits and will be useful for marker-assisted selection in wheat breeding.
