A new series of quinazoline-4(3H)-ones are evaluated for anticonvulsant activity. After intraperitoneal (ip) injection to albino mice at a dose of 100 mg/kg body weight, synthesized quinazolin-4(3H)-ones (1–24) were examined in the maximal electroshock (MES) induced seizures and subcutaneous pentylenetetrazole (scPTZ) induced seizure models in mice. The Rotarod method was applied to determine the neurotoxicity. Most of the compounds displayed anticonvulsant activity in the scPTZ screen at a dose range of 0.204–0.376 mmol/mL. Out of twenty-four, compounds 8, 13 and 19 proved to be the most active with a remarkable protection (100%) against PTZ induced convulsions and four times more potent activity than ethosuximide. The structure-activity relationship concluded valuable pharmacophoric information, which was confirmed by the molecular docking studies using the target enzyme human carbon anhydrase II (HCA II). The studied quinazoline analogues suggested that the butyl substitution at position 3 has a significant effect on preventing the spread of seizure discharge and on raising the seizure threshold. However, benzyl substitution at position 3 has shown a strong anticonvulsant activity but with less seizure prevention compared to the butyl substitution.

Three new acylated quercetin rhamnosides have been isolated from the leaves and stem of Calliandra haematocephala Hassk. (Fabaceae) and their structures were established as quercitrin 2′′-O-caffeate (1), quercitrin 3′′-O-gallate (2) and quercitrin 2′′,3′′-di-O-gallate (3). Also, 17 known compounds were identified as gallic acid (4), methyl gallate (5), caffeic acid (6), myricitrin (7), quercitrin (8), myricetin 3-O-β-D-4C1-glucopyranoside (9), afzelin (10), isoquercitrin (11), myricetin 3-O-(6′′-O-galloyl)-β-D-glucopyranoside (12), myricitrin 2′′-O-gallate (13), quercitrin 2′′-O-gallate (14), afzelin 2′′-O-gallate (15), myricitrin 3′′-O-gallate (16), afzelin 3′′-O-gallate (17), 1,2,3,4,6-penta-O-galloyl-β-D-4C1-glucopyranose (18), myricitrin 2′′,3′′-di-O-gallate (19), quercetin 3-O-methyl ether (20), for the first time from the genus Calliandra except for 6. Compounds 7, 8, 13, 14, 16 and 19 exhibited moderate to strong radical scavenging properties on lipid peroxidation, hydroxyl radical, superoxide anion generation and DPPH radical in comparison with that of quercetin as a positive control in vitro. Compounds 7 and 8 exhibited lethal effect towards brine shrimp Artemia salina.

A new acylated avicularin, namely avicularin 2″-(4′″-O-n-pentanoyl)-gallate (1) along with 12 metabolites have been isolated from the ethyl acetate extract of the aerial parts of Jussiaea repens L. (Onagraceae). Their structures were established as trifolin 2″-O-gallate (2), quercetrin (3), guaijaverin (4), reynoutrin (5), juglanin (6), avicularin (7), hyperin (8), trifolin (9), hyperin 2″-O-gallate (10), rutin (11), kaempferol (12) and quercetin (13) on the basis of their chromatographic properties, chemical and spectroscopic evidences. The investigated ethyl acetate extract was found to be non-toxic (LD50 up to the maximum soluble dose 4 g kg−1 body wt.) and had significant antioxidant, hepatoprotective, anti-inflammatory and antidiabetic activities. Also some of the isolated flavonoids showed cytotoxic activity against Ehrlich ascitis carcinoma cells.

Background: Several quinazoline and triazole derivatives are reported to possess a wide-range of interesting pharmacological effects. Although various triazoloquinazoline subclasses having been synthesized and studied, the preparation of 1,2,4-triazolo[1,5-a]quinazolines as antihypertensive agent is still relatively unexplored. In continuation of our earlier research, we aimed at the synthesis and development of various potent antihypertensive 1,2,4-triazoloquinazoline derivatives. Results: A new series of 1,2,4-triazolo[1,5-a]quinazoline derivatives have been synthesized. Their structures were mainly established by spectroscopic methods of analysis (IR, MS, 1H and 13C NMR). Their in vivo antihypertensive activity was evaluated by tail cuff method using Muromachi Blood Pressure Monitor (Model MK 2000) for rats and mice. Some of the tested compounds were found to exhibit valuable effects in terms of heart rate and blood pressure. According to the biological results, some of tested derivatives have abolished completely the tachycardia of the parent compounds and may be studied and modified as potential adrenoblockers and cardiac stimulant. Conclusion: New series of fifteen 1,2,4-triazolo[1,5-a]quinazolines were synthesized by convenient methodology from four key molecules, whereby their structures were established by advanced spectroscopic analyses. Some lead compounds have abolished completely the tachycardia of the parent compounds, that may be examined as potent adrenoblockers and some other compounds seem to be a cardiac stimulant or may be modified to enhance their hypotensive activity.

Diabetes is an emerging metabolic disorder. α-Glucosidase inhibitors, such as acarbose, delay the hydrolysis of carbohydrates by interfering with the digestive enzymes. This action decreases the glucose absorption and the postprandial glucose level. We have synthesized 25 tricyclic 2-phenoxypyrido[3,2-e][1,2,4]triazolo[1,5-a]pyrimidin-5(4H)-ones hybrids and evaluated their α-glucosidase inhibitory activity. Compounds 6h and 6d have shown stronger activity than that of acarbose. Compound 6h exhibited the highest inhibition with an IC50 of 104.07 µM. Molecular modelling studies revealed that compound 6h inhibits α-glucosidase due to the formation of a stable ligand-α-glucosidase complex and extra hydrogen bond interactions, and directed in the binding site by Trp329.
