Background: Previous studies have shown that exogenous short-chain fatty acids (SCFAs) introduction attenuated the body fat deposition in conventional mice and pigs. However, limited studies have evaluated the effects of exogenously introduced SCFAs on the lipid and glucose metabolism independently of the gut microbiota. This study was to investigate the effects of exogenous introduction of SCFAs on the lipid and glucose metabolism in a germ-free (GF) pig model. Methods: Twelve hysterectomy-derived newborn pigs were reared in six sterile isolators. All pigs were hand-fed with sterile milk powder for 21 d, then the sterile feed was introduced to pigs for another 21 d. In the second 21-d period, six pigs were orally administrated with 25 mL/kg sterile saline per day and considered as the GF group, while the other six pigs were orally administrated with 25 mL/kg SCFAs mixture (acetic, propionic, and butyric acids, 45, 15, and 11 mmol/L, respectively) per day and regarded as FA group. Results: Orally administrated with SCFAs tended to increase the adiponectin concentration in serum, enhance the CPT-1 activity in longissimus dorsi, and upregulate the ANGPTL4 mRNA expression level in colon (P < 0.10). Meanwhile, the mRNA abundances of ACC, FAS, and SREBP-1C in liver and CD36 in longissimus dorsi of the FA group were decreased (P < 0.05) compared with those in the GF group. Besides, the mRNA expression of PGC-1α in liver and LPL in longissimus dorsi tended to (P < 0.10) upregulate and downregulate respectively in the FA group. Moreover, oral administration of SCFAs tended to increase the protein level of GPR43 (P < 0.10) and decrease the protein level of ACC (P < 0.10) in liver. Also, oral administration of SCFAs upregulated the p-AMPK/AMPK ratio and the mRNA expressions of GLUT-2 and GYS2 in liver (P < 0.05). In addition, the metabolic pathway associated with the biosynthesis of unsaturated fatty acids was most significantly promoted (P < 0.05) by oral administration of SCFAs. Conclusions: Exogenous introduction of SCFAs might attenuate the fat deposition and to some extent improve the glucose control in the pig model, which occurred independently of the gut microbiota.

In the fabrication of 3-D complex tissues for implantation, layer-by-layer (LBL) electrospun nanofibrous scaffolds have recently received intensive interest. However, poor cell adhesion and cell expansion between the layers in an LBL stack remain important issues. In this study, we report a mussel-inspired, biomimetic approach to functionalize the surface of PCL/gelatin nanofibrous membranes coated with poly (dopamine) (PDA). Our study demonstrates that a PDA coating on electrospun PCL/gelatin nanofibers leads to a significant change in their surface properties and a higher adhesion force. Furthermore, we found that PDA coating promotes the adhesion and growth of adipose stem cells (ADSCs). In 3-D LBL stacked scaffolds, more cells survived in a PAD-coated scaffold than in a non-coated one. The PDA coating was further demonstrated to promote the osteogenic differentiation of ADSCs in LBL paper-stacking membranes. Our study suggests that PDA-coated paper-stacking nanofiber membranes present a facile and economic method for the development of 3D tissue engineering.

Genesis of novel gene regulatory modules is largely responsible for morphological and functional evolution. De novo generation of novel cis-regulatory elements (CREs) is much rarer than genomic events that alter existing CREs such as transposition, promoter switching or co-option. Only one case of de novo generation has been reported to date, in fish and without involvement of phenotype alteration. Yet, this event likely occurs in other animals and helps drive genetic/phenotypic variation. Using a porcine model of spontaneous hearing loss not previously characterized we performed gene mapping and mutation screening to determine the genetic foundation of the phenotype. We identified a mutation in the non-regulatory region of the melanocyte-specific promoter of microphthalmia-associated transcription factor (MITF) gene that generated a novel silencer. The consequent elimination of expression of the MITF-M isoform led to early degeneration of the intermediate cells of the cochlear stria vascularis and profound hearing loss, as well as depigmentation, all of which resemble the typical phenotype of Waardenburg syndrome in humans. The mutation exclusively affected MITF-M and no other isoforms. The essential function of Mitf-m in hearing development was further validated using a knock-out mouse model. Elimination of the MITF-M isoform alone is sufficient to cause deafness and depigmentation. To our knowledge, this study provides the first evidence of a de novo CRE in mammals that produces a systemic functional effect.

Although the importance of the intestinal microbiota in host growth and health is well known, the relationship between microbiota colonization and muscle development is unclear. In this study, the direct causal effects of the colonization of gut microorganisms on the muscle tissue of piglets were investigated. The body weight and lean mass of germ-free (GF) piglets were approximately 40% lower than those of normal piglets. The deletion of the intestinal microbiota led to weakened muscle function and a reduction in myogenic regulatory proteins, such as MyoG and MyoD, in GF piglets. In addition, the blinded IGF1/AKT/mTOR pathway in GF piglets caused muscle atrophy and autophagy, which were characterized by the high expression of Murf-1 and KLF15. Gut microbiota introduced to GF piglets via fecal microbiota transplantation not only colonized the gut but also partially restored muscle growth and development. Furthermore, the proportion of slow-twitch muscle fibers was lower in the muscle of GF piglets, which was caused by the reduced short-chain fatty acid content in the circulation and impaired mitochondrial function in muscle. Collectively, these findings suggest that the growth, development and function of skeletal muscle in animals are mediated by the intestinal microbiota.

Accurately predicting the air quality in a piggery and taking control measures in advance are important issues for pig farm production and local environmental management. In this experiment, the NH3 concentration in a semi-automatic piggery was studied. First, the random forest algorithm (RF) and Pearson correlation analysis were combined to analyze the environmental parameters, and nine input schemes for the model feature parameters were identified. Three kinds of deep learning and three kinds of conventional machine learning algorithms were applied to the prediction of NH3 in the piggery. Through comparative experiments, appropriate environmental parameters (CO2, H2O, P, and outdoor temperature) and superior algorithms (LSTM and RNN) were selected. On this basis, the PSO algorithm was used to optimize the hyperparameters of the algorithms, and their prediction performance was also evaluated. The results showed that the R2 values of PSO-LSTM and PSO-RNN were 0.9487 and 0.9458, respectively. These models had good accuracy when predicting NH3 concentration in the piggery 0.5 h, 1 h, 1.5 h, and 2 h in advance. This study can provide a reference for the prediction of air concentrations in pig house environments.
