Heat shock transcription factors (Hsfs) are a class of important transcription factors (TFs) which play crucial roles in the protection of plants from damages caused by various abiotic stresses. The present study aimed to characterize the Hsf genes in carnation (Dianthus caryophyllus), which is one of the four largest cut flowers worldwide. In this study, a total of 17 non-redundant Hsf genes were identified from the D. caryophyllus genome. Specifically, the gene structure and motifs of each DcaHsf were comprehensively analyzed. Phylogenetic analysis of the DcaHsf family distinctly separated nine class A, seven class B, and one class C Hsf genes. Additionally, promoter analysis indicated that the DcaHsf promoters included various cis-acting elements that were related to stress, hormones, as well as development processes. In addition, cis-elements, such as STRE, MYB, and ABRE binding sites, were identified in the promoters of most DcaHsf genes. According to qRT-PCR data, the expression of DcaHsfs varied in eight tissues and six flowering stages and among different DcaHsfs, even in the same class. Moreover, DcaHsf-A1, A2a, A9a, B2a, B3a revealed their putative involvement in the early flowering stages. The time-course expression profile of DcaHsf during stress responses illustrated that all the DcaHsfs were heat- and drought-responsive, and almost all DcaHsfs were down-regulated by cold, salt, and abscisic acid (ABA) stress. Meanwhile, DcaHsf-A3, A7, A9a, A9b, B3a were primarily up-regulated at an early stage in response to salicylic acid (SA). This study provides an overview of the Hsf gene family in D. caryophyllus and a basis for the breeding of stress-resistant carnation.

A pot experiment was performed to investigate the effect of phytoremediation (CK, using tall fescue), fungi remediation (GV, using Glomus versiforme), bacterial remediation (PS, using Pseudomonas fluorescens Ps2-6), and microbial-phytoremediation (GVPS, using three species) on removing polycyclic aromatic hydrocarbons (PAHs) and the microbial diversity in soils. Inoculation with G. versiforme and P. fluorescens could increase the biomass of tall fescue and the accumulation of phenanthrene (PHE) and pyrene (PYR) in plants. Among them, the highest PHE and PYR removal efficiencies and highest biomass of tall fescue were observed in the GVPS treatment and the microbial diversity in contaminated soil was changed, the result revealed that Proteobacteria and Ascomycota were the dominant bacterial phylum and fungal phylum in all treatments, while more Proteobacteria were detected in GVPS treatment. At the genus level, the abundance of Sphingomonas (3.17%), Pseudomonas (2.05%), and Fusarium (8.65%) treated with GVPS increased compared with other treatments. These pieces of evidence contribute to a better understanding of the mechanisms involved in the combined microbial-phytoremediation strategies for PAHs-contaminated soils, especially the effects of microbial-phytoremediation on rhizosphere microbial diversity. Inoculation with G. versiforme and P. fluorescens could increase PHE and PYR removal efficiencies, the biomass of tall fescue, microbial diversity, and the abundance of Proteobacteria, Sphingomonas, Pseudomonas, and Fusarium in (PHE + PYR)-contaminated soils.

A novel, highly selective and colorimetric strategy for pyruvic acid (PA) detection is proposed based on the conformational change of i-motif DNA and the salt-induced aggregation effects of unmodified gold nanoparticles. The addition of PA can induce the C-rich DNA to fold into a rigid close-packed i-motif DNA, which accordingly loses its ability to effectively protect and stabilize AuNPs from salt-induced aggregation. In the presence of pyruvate decarboxylase (PDC), PA can be efficiently converted to acetaldehyde and CO2 through nonoxidative decarboxylation, causing the solution pH to change from acidic to neutral. Thus, the C-rich DNA can still maintain its extended single strand status and can effectively stabilize the AuNPs from salt-induced aggregation. The absorbance difference before and after the addition of PDC depends on the concentration of PA and can be used for the quantitative determination of PA. The developed colorimetric sensor can achieve PA detection with a linear range from 5.6 × 10−6 M to 1.68 × 10−4 M and a detection limit of 3.0 μM. The as-established colorimetric strategy opens a new avenue for the specific detection of DNA conformation-responsive small biomolecules.

Low-temperature and weak-light conditions have major effects on the growth and flower quality of horticultural plants. A greenhouse pot experiment was performed to investigate the effects of arbuscular mycorrhizal fungi (AMF) (Funneliformis mosseae and Glomus versiforme) on the growth, photosynthesis, and chlorophyll fluorescence parameters of snapdragon (Antirrhinum majus L.) under low-temperature and weak-light stress. The growth and biomass of snapdragon were higher following inoculation with F. mosseae and G. versiforme compared with control plants. The percentage of mycorrhizal colonization and root activity were high in A. majus plants with AMF. AMF inoculation enhanced the net photosynthetic rate, transpiration rate, stomatal conductance, and water use efficiency of plants under low-temperature and weak-light conditions. Furthermore, the chlorophyll content, potential activity of photosystem II (PSII), effective photochemistry quantum efficiency of PSII, actual photochemical quantum efficiency of PSII, and photochemical quenching coefficient were higher in AMF-inoculated plants than in uninoculated plants. The application of AMF reduced the intercellular CO2 concentration and non-photochemical quenching coefficient. Thus, snapdragon plants treated with F. mosseae and G. versiforme are more resistant to low-temperature and weak-light stress than untreated plants.
