As a major lignocellulosic biomass, which represented more than half of the world’s agricultural phytomass, crop residues have been considered as feedstock for biofuel production. However, large-scale application of this conventional biofuel process has been facing obstacles from cost efficiency, pretreatment procedure, and secondary pollution. To meet the growing demands for food, feed, and energy as the global population continues to grow, certain kinds of insects, many of which are voracious feeders of organic wastes that may help address environmental, economic, and health issues, have been highlighted as a source of protein and fat. The biorefinery studied includes initial corn stover degradation by yellow mealworm (Tenebrio molitor L.), followed by a second stage that employs black soldier fly (Hermetia illucens L.), to utilize the residues produced during the first stage. These two insect-based biorefinery yielded 8.50 g of insect biomass with a waste dry mass reduction rate of 51.32%, which resulted in 1.95 g crude grease from larval biomass that produced 1.76 g biodiesel, 6.55 g protein, and 111.59 g biofertilizer. The conversion rate of free fatty acids of crude grease into biodiesel reached 90%. The components of cellulose, hemicellulose, and lignin contained in corn stover hydrolyzed harmoniously, resulting in declines of 45.69, 51.85, and 58.35%, respectively. Moreover, fluctuations in lipid, protein, and reducing sugar were also analyzed. The investigation findings demonstrated that successive co-conversion of corn stover by insects possessing different feeding habits could be an attractive option for efficient utilization of lignocellulosic resources, and represents a potentially valuable solution to crop residues management, rise of global liquid energy, and animal feed demand.

Since the first X-ray structure of Cry3Aa was revealed in 1991, numerous structures of B. thuringiensis toxins have been determined and published. In recent years, functional studies on the mode of action and resistance mechanism have been proposed, which notably promoted the developments of biological insecticides and insect-resistant transgenic crops. With the exploration of known pore-forming toxins (PFTs) structures, similarities between PFTs and B. thuringiensis toxins have provided great insights into receptor binding interactions and conformational changes from water-soluble to membrane pore-forming state of B. thuringiensis toxins. This review mainly focuses on the latest discoveries of the toxin working mechanism, with the emphasis on structural related progress. Based on the structural features, B. thuringiensis Cry, Cyt and parasporin toxins could be divided into three categories: three-domain type α-PFTs, Cyt toxin type β-PFTs and aerolysin type β-PFTs. Structures from each group are elucidated and discussed in relation to the latest data, respectively.

There can be substantial negative consequences for insects colonizing a resource in the presence of competitors. We hypothesized that bacteria, associated with an oviposition resource and the insect eggs deposited on that resource, serve as a mechanism regulating subsequent insect attraction, colonization and potentially succession of insect species. We isolated and identified bacterial species associated with insects associated with vertebrate carrion and used these bacteria to measure their influence on the oviposition preference of adult black soldier flies which utilizes animal carcasses and is an important species in waste management and forensics. We also ascertained that utilizing a mixture of bacteria, rather than a single species, differentially influenced behavioral responses of the flies, as did bacterial concentration and the species of fly from which the bacteria originated. These studies provide insight into interkingdom interactions commonly occurring during decomposition, but not commonly studied.

The evolution of TLR-mediated innate immunity is a fundamental question in immunology. Here, we report the characterization and functional analysis of four TLR members in the lophotrochozoans Crassostrea gigas (CgTLRs). All CgTLRs bear a conserved domain organization and have a close relationship with TLRs in ancient non-vertebrate chordates. In HEK293 cells, every CgTLR could constitutively activate NF-κB responsive reporter, but none of the PAMPs tested could stimulate CgTLR-activated NF-κB induction. Subcellular localization showed that CgTLR members have similar and dual distribution on late endosomes and plasma membranes. Moreover, CgTLRs and CgMyD88 mRNA show a consistent response to multiple PAMP challenges in oyster hemocytes. As CgTLR-mediated NF-κB activation is dependent on CgMyD88, we designed a blocking peptide for CgTLR signaling that would inhibit CgTLR-CgMyD88 dependent NF-κB activation. This was used to demonstrate that a Vibrio parahaemolyticus infection-induced enhancement of degranulation and increase of cytokines TNF mRNA in hemocytes, could be inhibited by blocking CgTLR signaling. In summary, our study characterized the primitive TLRs in the lophotrocozoan C. gigas and demonstrated a fundamental role of TLR signaling in infection-induced hemocyte activation. This provides further evidence for an ancient origin of TLR-mediated innate immunity.
