Energy intake prepartum is critically important to health, milk performance, and profitability of dairy cows. The objective of this study was to determine the effect of reduced energy density of close-up diets on dry matter intake (DMI), lactation performance and energy balance (EB) in multiparous Holstein cows which were housed in a free-stall barn and fed for ad libitum intake. Thirty-nine dry cows were blocked and assigned randomly to three groups fed a high energy density diet [HD, n = 13; 6.8 MJ of net energy for lactation (NEL)/kg; 14.0% crude protein (CP) ], or a middle energy density diet (MD, n = 13; 6.2 MJ NEL/kg; 14.0% CP), or a low energy density diet (LD, n = 13; 5.4 MJ NEL/kg; 14.0% CP) from d 21 before expected day of calving. After parturition, all cows were fed the same lactation diet to d 70 in milk (DIM). The DMI and NEL intake prepartum were decreased by the reduced energy density diets (P < 0.05). The LD group consumed 1.3 kg/d (DM) more diet compared with HD group in the last 24 h before calving. The milk yield and the postpartum DMI were increased by the reduced energy density diet prepartum (P < 0.05). The changes in BCS and BW prepartum and postpartum were not affected by prepartum diets. HD group had higher milk fat content and lower lactose content compared with LD group during the first 3 wk of lactation (P < 0.05). The energy consumption for HD, MD and LD groups were 149.8%, 126.2% and 101.1% of their calculated energy requirements prepartum (P < 0.05), and 72.7%, 73.1% and 75.2% during the first 4 wk postpartum, respectively. In conclusion, the low energy density prepartum diet was effective in controlling NEL intake prepartum, and was beneficial in increasing DMI and milk yield, and alleviating negative EB postpartum.

The objective of this study was to determine the effects of heat stress (HS) on physiological, blood biochemical, and energy metabolism in Dazu black goats. Six wether adult Dazu black goats were subjected to 3 experimental periods: high HS (group H, temperature-humidity index [THI] > 88) for 15 d, moderate HS (group M, THI was 79-88) for 15 d, and no HS (group L, THI < 72) for 15 d. Rectal temperature (RT) and respiratory rate (RR) were determined on d 7 and 15 of each period, and blood samples were collected on d 15 of each period. All goats received glucose (GLU) tolerance test (GTT) and insulin (INS) tolerance test on d 7 and d 10 of each period. The results showed that HS decreased dry matter intake (DMI) and INS concentrations (p < 0.05), and increased RT, RR, non-esterified fatty acid (NEFA), cortisol (COR), and total protein (TP) concentrations (p < 0.05). Compared to group L, the urea nitrogen (BUN) concentration increased and GLU concentration decreased in group H (p < 0.05).During the GTT, the area under the curve (AUC) of GLU concentrations increased by 12.26% (p > 0.05) and 40.78% (p < 0.05), and AUC of INS concentrations decreased by 26.04% and 14.41% (p < 0.05) in groups H and M compared to group L, respectively. The INS concentrations were not significant among the three groups (p > 0.05) during the ITT. A total of 60 differentially expressed metabolites were identified in response to groups H and M. In HS, changes in metabolites related to carbohydrate metabolism and glycolysis were identified (p < 0.05). The metabolites related to fatty acid βoxidation accumulated, glycogenic and ketogenic amino acids were significantly increased, while glycerophospholipid metabolites were decreased in HS (p < 0.05). HS significantly increased 1-methylhistidine, creatinine, betaine, taurine, taurolithocholic acid, inosine, and hypoxanthine, while decreasing vitamin E in blood metabolites (p < 0.05). In summary, HS changed the metabolism of fat, protein, and energy, impaired GLU tolerance, and mainly increased amino acid metabolism to provide energy in Dazu black goats.

Conjugated linoleic acids (CLAs) play a major role in adipocyte differentiation and lipid metabolism in animals. MicroRNAs (miRNAs) appear to be involved in many biological processes in adipose tissue. However, the specific influence on miRNAs by CLA supplementation in porcine adipose tissue remains unclear. Thus, we continuously added 1.5% CLA to the pig diet from the embryo stage to the finishing period and conducted a high-throughput sequencing approach to analyse the changes in adipose tissue miRNAs. We identified 283 known porcine miRNAs, and 14 miRNAs were differentially expressed in response to CLA treatment. A Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that the targets of the 14 differentially expressed miRNAs were involved in the Wnt signalling pathway. The CLA treatment downregulated the gene expression of PPARγ, C/EBPα, FAS, and FATP1 in both subcutaneous and abdominal fat tissues; the analysis showed that ssc-miR-21 expression was significantly correlated with PPARγ expression (p<0.05), and speculated that ssc-miR-21 might influence adipogenesis through PPARγ. In conclusion, our study analysed the miRNA profiles in porcine adipose tissues by CLA treatment, and demonstrated that miRNAs are important regulators of fat lipogenesis. This study provides valuable information for the molecular regulatory mechanism of CLA on adipose tissue.

The objective of this study was to determine the effect of close-up diets with increased ratios of neutral detergent fiber (NDF) to non-fibrous carbohydrates (NFC) on feeding behavior in transition cows. Thirty-nine dry cows were blocked, assigned randomly to three groups and fed diets with a low (1.07, LF), medium (1.41, MF) or high (2.08, HF) NDF:NFC ratio from 21 d prior to the expected day of calving. After parturition, all cows were fed the same diet for a further 35 d. The average DMI decreased as the ratio of NDF:NFC increased at -3 weeks (Wk) and -2 Wk and increased at +5 Wk plessthan 0.05). The average daily feeding time was lower and the average feeding rate was higher (plessthan 0.05) for LF cows compared with MF and HF cows during prepartum. In conclusion, a prepartum diet with increased ratios of NDF:NFC decreased the DMI and feeding rate, increased daily feeding times and visits prepartum and was beneficial in increasing DMI postpartum.
