This study aimed to evaluate the osteogenic activity of Endosequence Root Repair Material (ERRM) putty using rat mesenchymal stem cells (MSCs). The extract of set ERRM and ProRoot-mineral trioxide aggregate (MTA) (control) was cocultured with rat MSCs and incubated for one, three, and seven days. The cell viability and proliferation were assessed. A quantitative real-time polymerase chain reaction for bone morphogenetic protein-2 (BMP-2), alkaline phosphatase, bone sialoprotein, and osteocalcin gene expression was performed. Both materials enhanced cell viability and proliferation, which increased over time. On day seven, the cells treated with either material exhibited significantly greater cell viability compared with control untreated cells. MSCs treated with either material showed deeper alkaline phosphatase staining after three days compared to control untreated cells. Treated MSCs also exhibited upregulation of the gene expression of bone morphogenetic protein-2, alkaline phosphatase, bone sialoprotein, and osteocalcin. Both ERRM and ProRoot-MTA enhance the osteogenic differentiation of MSCs.

Oral squamous cell carcinoma (OSCC) is a malignant tumour of Head and Neck Cancer (HNC). The recent therapeutic approaches used to treat cancer have adverse side effects. The natural agents exhibiting anticancer activities are generally considered to have a robust therapeutic potential. Curcuminoids, one of the major active compounds of the turmeric herb, are used as a therapeutic agent for several diseases including cancer. In this study, the cytotoxicity of curcuminoids was investigated against OSCC cell line HNO97. Our data showed that curcuminoids significantly inhibits the proliferation of HNO97 in a time and dose-dependent manner (IC50=35 μM). Cell cycle analysis demonstrated that curcuminoids increased the percentage of G2/M phase cell populations in the treated groups. Treating HNO97 cells with curcuminoids led to cell shrinking and increased detached cells, which are the typical appearance of apoptotic cells. Moreover, flow cytometry analysis revealed that curcuminoids significantly induced apoptosis in a time-dependent manner. Furthermore, as a response to curcuminoids treatment, comet tails were formed in cell nuclei due to the induction of DNA damage. Curcuminoids treatment reduced the colony formation capacity of HNO97 cells and induced morphological changes. Overall, these findings demonstrate that curcuminoids can in vitro inhibit HNC proliferation and metastasis and induce apoptosis.

This study involved the synthesis of the UiO-67 metal-organic framework; UiO-67 is a well-known type of MOF obtained by coordinating the Zr6O4(OH)4 metal unit with the 4,42-biphenyldicarboxylate organic linker, using the hydrothermal technique. The novelty of the current work is to synthesize UiO-67 MOFs, and their application as biological agents for antibacterial and cancer cells. Subsequently, the composite material UiO-67 was subjected to a comprehensive characterization process involving Fourier-transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), thermal gravimetric analyses (TGA) and surface area analysis, and the results showed the successful synthesis of the UiO-67 MOFs, with a high specific surface area of 1415 m2/g. The synthesized UiO-67 for its antibacterial properties tested against five pathogenic bacterial strains, which include three gram-positive and methicillin-resistant pairs including MRSA, S. aureus and Enterococcus faecalis, and Two gram-negative bacteria E. colli and S. typhimurium using the agar well diffusion method. These findings have shown enhanced, strong antibacterial activity against all the five used gram-positive and gram-negative bacterial strains. Furthermore, the anticancer efficacy of UiO-67 was evaluated on two distinct types of cancer cells: We are using MCF-7 (human breast cancer cell line) and HepG2 (human liver cancer cells). The experiments prove that UiO-67 has the potential of cytotoxicity against both Glioblastoma and H460 cancer lines with the ability to inhibit apoptosis at the same time.
