Capillary zone electrophoresis-mass spectrometry (CE-MS) is a mature analytical tool for the efficient profiling of (highly) polar and ionizable compounds. However, the use of CE-MS in comparison to other separation techniques remains underrepresented in metabolomics, as this analytical approach is still perceived as technically challenging and less reproducible, notably for migration time. The latter is key for a reliable comparison of metabolic profiles and for unknown biomarker identification that is complementary to high resolution MS/MS. In this work, we present the results of a Metabo-ring trial involving 16 CE-MS platforms among 13 different laboratories spanning two continents. The goal was to assess the reproducibility and identification capability of CE-MS by employing effective electrophoretic mobility (µeff) as the key parameter in comparison to the relative migration time (RMT) approach. For this purpose, a representative cationic metabolite mixture in water, pretreated human plasma and urine samples spiked with the same metabolite mixture, were used and distributed for analysis by all laboratories. The µeff was determined for all metabolites spiked into each sample. The background electrolyte (BGE) was prepared and employed by each participating lab following the same protocol. All other parameters (capillary, interface, injection volume, voltage ramp, temperature, capillary conditioning and rinsing procedure, etc.) were left to the discretion of the contributing labs. The results revealed that the reproducibility of the µeff for 20 out of the 21 model compounds was below 3.1% vs. 10.9% for RMT, regardless of the huge heterogeneity in experimental conditions and platforms across the thirteen labs. Overall, this Metabo-ring trial demonstrated that CE-MS is a viable and reproducible approach for metabolomics.

Herein we describe the synthesis and evaluation of four novel HER2-targeting, cathepsin B-sensitive antibody–drug conjugates bearing a monomethylauristatin E (MMAE) cytotoxic payload, constructed via the conjugation of cleavable linkers to trastuzumab using a site-specific bioconjugation methodology. These linkers vary by both cleavable trigger motif and hydrophilicity, containing one of two cathepsin B sensitive dipeptides (Val-Cit and Val-Ala), and engendered with either hydrophilic or hydrophobic character via application of a PEG12 spacer. Through evaluation of physical properties, in vitro cytotoxicity, and receptor affinity of the resulting antibody–drug conjugates (ADCs), we have demonstrated that while both dipeptide triggers are effective, the increased hydrophobicity of the Val-Ala pair limits its utility within this type of linker. In addition, while PEGylation augments linker hydrophilicity, this change does not translate to more favourable ADC hydrophilicity or potency. While all described structures demonstrated excellent and similar in vitro cytotoxicity, the ADC with the ValCitPABMMAE linker shows the most promising combination of in vitro potency, structural homogeneity, and hydrophilicity, warranting further evaluation into its therapeutic potential.

Linum flavum hairy root lines were established from hypocotyl pieces using Agrobacterium rhizogenes strains LBA 9402 and ATCC 15834. Both strains were effective for transformation but induction of hairy root phenotype was more stable with strain ATCC 15834. Whereas similar accumulation patterns were observed in podophyllotoxin-related compounds (6-methoxy-podophyllotoxin, podophyllotoxin and deoxypodophyllotoxin), significant quantitative variations were noted between root lines. The influence of culture medium and various treatments (hormone, elicitation and precursor feeding) were evaluated. The highest accumulation was obtained in Gamborg B5 medium. Treatment with methyl jasmonate, and feeding using ferulic acid increased the accumulation of aryltetralin lignans. These results point to the use of hairy root culture lines of Linum flavum as potential sources for these valuable metabolites as an alternative, or as a complement to Podophyllum collected from wild stands.

The proton affinity ( PA) and protonation entropy, ΔpS°, of glycine (Gly), 1, aspartic acid (Asp), 2, asparagine (Asn), 3, histidine (His), 4, lysine (Lys), 5, glutamic acid (Glu), 6, and glutamine (Gln), 7, have been reinvestigated by the extended kinetic method, using the “isothermal point” method and the orthogonal distance regression technique. The proton affinity values of α-amino acids bearing a basic residue ( PA = 926.8, 965.2, 996.0, 993.9, 981.8 and 988.1 kJ mol−1 for 2–7, respectively) show significant deviation from the tabulated values. As expected from the effect of a strong intramolecular hydrogen bond in the protonated forms of these peculiar amino acids, negative protonation entropies are detected (ΔpS° = −36, −43, −37, −29, −95 and −55 J mol−1 K−1 for 2–7, respectively).

Chlordecone (CLD) is a persistent toxic chlorinated pesticide which contaminates different ecosystems in French West Indies. A soil remediation process including zero-valent iron (ZVI) has produced promising results but failed to completely degrade CLD, and the analytical procedures used yielded little information on the transformation products. To fill these gaps, dechlorination of aqueous CLD by micrometric particles of ZVI has been investigated. Aliquots of water with 25% (v/v) of acetone spiked with 100 ppm CLD were taken at different times during a 30-day ZVI treatment and directly analysed by ultra-high-performance liquid chromatography in negative electrospray ionisation mode. CLD has been totally transformed after 14 days into 14 dechlorinated degradation products, including 9 isomeric compounds. The maximum chloride concentrations appearing in the medium represent 44% of that which would result from total dechlorination of CLD. The CLD transformation products identified by accurate mass measurements on an ultra-high-resolution Q-TOF mass spectrometer (Q-TOF-MS) were C10H3Cl9O2, C10H4Cl8O2, C10H5Cl7O2, C10H6Cl6O2 and C10H7Cl5O2. The results show the interest of LC-Q-TOF-MS for identifying transformation products of organic contaminants, and the effectiveness of micrometric ZVI particles to totally transform CLD into less chlorinated products.
