In this study, two bacterial species, Salipiger thiooxidans and Exiguobacterium aestuarii, were extracted and screened from the Saemangeum Reservoir. This study examined these species’ suitability as a probiotic by confirming the effects of S. thiooxidans and E. aestuarii added to rearing water for L. vannamei. Three experimental groups were evaluated for 6 weeks: (1) a control group reared in natural (i.e., untreated) water (CON), (2) an experimental group in which S. thiooxidans was added to natural water (SMG-A), and (3) natural water inoculated with E. aestuarii (SMG-B). The SMG-B group inoculated with E. aestuarii showed significantly higher final body weight, weight gain, specific growth rates, and feed efficiency than the control group. The SMG-B group inoculated with E. aestuarii exhibited significantly higher levels of serum lysozyme, and ACP and ALP activity than the control and SMG-A groups. The SMG-A and SMG-B groups inoculated with probiotics showed significantly lower total ammonia nitrogen and nitrite than the control group. Our findings suggest that S. thiooxidans and E. aestuarii extracted from the Saemangeum Reservoir can improve the water quality of aquaculture water, and, in particular, E. aestuarii is a potential probiotic for L. vannamei.

The spotted halibut is species that has a high potential market value in Korea, but the supply of seed is unstable because of the limited milt production of males. The objective of this research was to explore different aspects, such as CPAs, diluents, dilution ratio, and freezing rates, to develop an optimal sperm cryopreservation. The parameters assessed were movable sperm ratio, sperm activity index, survival rate, and DNA damage. The CPAs tested in this research were propylene glycol, dimethyl sulfoxide (DMSO), methanol, ethylene glycol, and glycerol. Different diluents, including 300 mM sucrose, 300 mM glucose, Stain’s solution, and Ringer’s solution, were investigated. The previous experiment showed that the optimal CPA for cryopreservation was DMSO with a concentration of 15% with 300 mM as diluent. To determine the effect of the dilution ratio, sperm was diluted to 1:1, 1:2, 1:10, 1:100, and 1:1000 with 300 mM sucrose containing DMSO at a final concentration of 15%. Lastly, the optimal freezing rate of the sperm was evaluated with four different freezing rates (−1, −5, −10, and −20 °C/min). Post-thaw sperm motility was higher with a dilution ratio lower than 1:2, and the freezing rate was less than −5 °C/min. In conclusion, these findings represent the development of a cryopreservation protocol for spotted halibut.

Ecological flow (E-flow) determination is an essential component of stream management and the preservation of aquatic ecosystems within a watershed. E-flow should be determined while considering the overall status of the watershed, including the hydrological cycle, hydraulic facility operation, and stream ecology. The purpose of this study is to determine E-flow by considering watershed status through coupled modeling with SWAT and PHABSIM. SWAT was calibrated to ensure reliability when coupling the two models, using observed data that included streamflow and dam inflows. The calibration result of SWAT showed that the averages of R2, NSE, and RMSE were 0.62, 0.57, and 1.68 mm/day, respectively, showing satisfactory results. Flow duration analysis using the SWAT results was performed to apply to discharge boundary conditions for PHABSIM. The averages of Q185 (mid-range flows) and Q275 (dry conditions) were suitable to simulate fish habitat. The habitat suitability index derived through a fish survey was applied to PHABSIM to estimate E-flow. E-flow was estimated at 20.0 m3/s using the coupled model and compared with the notified instream flow by the Ministry of Environment. The results demonstrate a high level of applicability for the coupled modeling approach between the watershed and physical habitat simulation models. Our attempt at coupled modeling can be utilized to determine E-flow considering the watershed status.
