In this study, two Nepeta species, Nepeta viscida, N. nuda subsp. nuda, and their putative hybrids N. x tmolea were investigated for their trichome structure and density. For this purpose, fresh or herbarium specimens were used. SEM photographs were taken with the standard techniques in Balıkesir University. While N. x tmolea individuals are similar to N. nuda subsp. nuda in some respects (for example, absence long glandular trichomes on the stem, the absence of glandular hairs on the corolla), it shows some transition between N. viscida and N. nuda subp. nuda in some respects (for example, while the corolla of N. viscida does not contain long glandular hair, presence in a high density in N. nuda subsp. nuda and low density in N. x tmolea individuals).

Nepeta viscidaand N. nuda subsp. nuda and N. × tmolea were examined in this study. Mainly fresh leaf pieces, dried with silica grains, were used for DNA extraction procedures via DNA isolation kits. Standard PCR techniques were executed using three different primer sets (one nuclear DNA region (nrITS) and two chloroplast DNA regions (rpl32-trnL andtrnA(Leu)-trnA(Phe)-trnL-F). DNA sequences were analysed and evaluated using different molecular approaches and software. Consequently, the inconstant molecular structure and hybrid nature of N. × tmolea specimens were shown and interpreted in this study. According to our result, N. × tmolea have some intermediate characters compared to its parents. nrITS data give more information phylogenetically, and also the most polymorphic loci are seen in nrITS data. Morphological and molecular data contribute to define separation of N. × tmolea. Consequently, the inconstant molecular structure and hybrid nature of N. × tmolea specimens were shown and interpreted in this study.

This study aims to describe a new hybrid of Origanum, which occurs on Karpathos (Greece), by examining its morphology and molecular structure. Nuclear ribosomal DNA (ITS) data of the alleged hybrid specimens, and its putative parents, O. onites and O. vetteri, were examined. Hybrid and parent individuals sharing the same location with the new hybrid from Karpathos and different specimens belonging to ancestral taxa from Turkey, were used for molecular analysis. According to molecular data, there are some polymorphic nucleotide regions showing the hybridization evidence between the new hybrid and its parents. Molecular results show that the new hybrid is more similar to O. vetteri. Also, the new hybrid has some intermediate morphological characteristics ranging between its parents. Consequently, it was deemed appropriate to present the studied putative hybrid to the scientific world as a new hybrid taxon.
