Exploring suitable maize straw-return measures is essential for the new double-cropping system of maize (Zea mays L.)–rice (Oryza sativa L.) rotation in the middle reaches of Yangtze River in China, which can increase crop yield by improving soil quality. In this study, four straw-return measures were evaluated by investigating the soil bulk density (BD), organic matter (OM), microbial community, and nutrients from 2016 to 2018. The four straw-return treatments were as follows: (1) no straw-return (CK), (2) only rice straw incorporated into the field (M0Ri), (3) both maize and rice straw incorporated to field (MiRi), and (4) maize straw mulched and rice straw incorporated into the field (MmRi). Compared to CK, two-season crop straw-return treatments changed soil microbial community composition, and increased soil total organic carbon (TOC) and dissolved organic carbon (DOC), microbial biomass carbon (MBC), mineralized nitrogen (Nmin), available phosphorus (P) and exchangeable potassium (K) in the 0–20 cm soil layer by 3.6%, 63.4%, 38.8%, 12.4%, 39.7%, and 21.6%, respectively, averaged across MmRi and MiRi treatments. In addition, MmRi and MiRi increased annual yield by 9.1% and 15.2% in 2017 and 11.7% and 12.9% compared to CK in 2018, respectively. MmRi exhibited superiority in the soil microbial community, enzyme activities, DOC, MBC, Nmin, available P, and exchangeable K in contrast to MiRi. We concluded that MmRi is the best measure to implement for straw-return in maize–rice rotation systems.

Drought stress is a major environmental constraint that limits rice (Oryza sativa L.) production worldwide. In this study, we investigated the effects of drought stress at the booting stage on rice leaf physiological characteristics and yield. The results showed that drought stress would lead to a significant decrease in chlorophyll content and photosynthesis in rice leaves, which would affect rice yield. Three different rice varieties were used in this study, namely Hanyou73 (HY73), Huanghuazhan (HHZ), and IRAT109. Under drought stress, the chlorophyll content of all cultivars decreased significantly: 11.1% and 32.2% decreases in chlorophyll a and chlorophyll b in HHZ cultivars, 14.1% and 28.5% decreases in IRAT109 cultivars, and 22.9% and 18.6% decreases in HY73 cultivars, respectively. In addition, drought stress also led to a significant decrease in leaf water potential, a significant increase in antioxidant enzyme activity, and an increase in malondialdehyde (MDA) content, suggesting that rice activated a defense mechanism to cope with drought-induced oxidative stress. This study also found that drought stress significantly reduced the net photosynthetic rate and stomatal conductance of rice, which, in turn, affected the yield of rice. Under drought stress, the yield of the HHZ cultivars decreased most significantly, reaching 30.2%, while the yields of IRAT109 and HY73 cultivars decreased by 13.0% and 18.2%, respectively. The analysis of yield composition showed that the number of grains per panicle, seed-setting rate, and 1000-grain weight were the key factors affecting yield formation. A correlation analysis showed that there was a significant positive correlation between yield and net photosynthetic rate, stomatal conductance, chla/chlb ratio, Rubisco activity, and Fv/Fm, but there was a negative correlation with MDA and non-photochemical quenching (NPQ). In summary, the effects of drought stress on rice yield are multifaceted, involving changes in multiple agronomic traits. The results highlight the importance of selecting and nurturing rice varieties with a high drought tolerance, which should have efficient antioxidant systems and high photosynthetic efficiency. Future research should focus on the genetic mechanisms of these physiological responses in order to develop molecular markers to assist in the breeding of drought-tolerant rice varieties.

Due to relatively low yield as well as low resources use efficiency with double rice (Oryza sativa L.) cropping systems (RR), exploring new cropping systems to increase yield and resources use efficiency simultaneously has become a large challenge of the middle reaches of the Yangtze River (MRYR). Our previous study demonstrated that the maize (Zea mays L.)–rice cropping system (MR) exhibited higher superiority of yield and resource use efficiency compared with the conventional double-rice cropping system. However, the reason for the yield increases in both maize and rice and the physiological processes involved in those two crops under MR are poorly understood. A 3-year field experiment was conducted at two sites (Wuxue and Jingmen) from 2016 to 2018 to examine the differences in dry matter (DM) accumulation, soil properties, and resources use efficiency between the MR and RR cropping systems. Compared with RR, the annual yield of MR was 18.2–26.3% and 15.4–31.5% higher across three years at Wuxue and Jingmen, respectively. The average yield of maize in MR was 36.5% and 21.9% higher than that of early rice in RR at Wuxue and Jingmen, respectively. The yield increase for maize was mainly attributed to the 29.7% (Wuxue) and 28.5% (Jingmen) increases in post-silking DM accumulation due to the higher plant growth rate promoted by the higher net assimilation rate and radiation use efficiency. For the late rice in MR, the average yield was 10.9% and 14.5% higher than that of late rice in RR at Wuxue and Jingmen, respectively, which was promoted by the 7.8–23.3% increase in pre-anthesis DM accumulation due to improved soil properties. Compared with RR, the MR cropping system exhibited increased soil pH, total organic carbon, and mineral nitrogen, and decreased the bulk density in the late rice season. As a result of greater yield in both seasons under MR, the annual accumulated temperature and radiation use efficiency, partial factor productivity from applied nitrogen, and water use efficiency of MR were 17.7–26.4%, 22.2–25.5%, 5.5–7.8%, and 33.6–48.7% higher than those of RR, respectively. We conclude that the higher yield in the MR than in the RR cropping system was mainly attributed to the accumulation of post-silking biomass due to maximized use of radiation in the maize season, and the accumulation of pre-anthesis biomass due to improved soil nutrients in the late rice season.

Plant immune receptor proteins, including PRRs and NLRs, are essential for various aspects of plant growth and development, including microbial sensing, and immune responses. However, a comprehensive and systematic understanding of immune-related gene families in alfalfa plants is lacking. This study aims to identify and characterize these genes in alfalfa to fill this knowledge gap. We identified 736 RLK, 407 RLP, and 870 NLR genes in the alfalfa genome and analyzed their structural and genetic variation. Phylogenetic analysis classified RLKs, RLPs, and NLRs into 22, 21, and three subgroups, respectively. Chromosomal location and synteny analyses revealed that single-gene duplication events contributed to the evolution of these genes. Expression profiling, promoter cis-acting elements, and associated network analyses demonstrated their critical roles in plant growth and stress prevention. Specifically, the alfalfa RLKs gene (MsRLK725) was selected for functional validation due to its numerous genetic variants and significant upregulation under biotic stress. Overexpression of MsRLK725 in Nicotiana benthamiana enhanced resistance to Rhizoctonia solani and Phoma medicaginis, and the hairy root transient transformation system improved alfalfa’s salt tolerance. Our findings offer valuable insights for future research on using immune-related genes in alfalfa, highlighting their potential in molecular breeding for stress tolerance and disease resistance.
