Compared with open oesophagectomy (OE), minimally invasive oesophagectomy (MIO) proves to have benefits in reducing the risk of pulmonary complications for patients with resectable oesophageal cancer. However, it is unknown whether MIO has superiority in reducing the occurrence of in-hospital mortality (IHM). The objective of this meta-analysis was to explore the effect of MIO vs. OE on the occurrence of in-hospital mortality (IHM). Sources such as Medline (through December 31, 2014), Embase (through December 31, 2014), Wiley Online Library (through December 31, 2014), and the Cochrane Library (through December 31, 2014) were searched. Data of randomized and non-randomized clinical trials related to MIO versus OE were included. Eligible studies were those that reported patients who underwent MIO procedure. The control group included patients undergoing conventional OE. Fixed or random -effects models were used to calculate summary odds ratios (ORs) or relative risks (RRs) for quantification of associations. Heterogeneity among studies was evaluated by using Cochran’s Q and I2 statistics. A total of 48 studies involving 14,311 cases of resectable oesophageal cancer were included in the meta-analysis. Compared to patients undergoing OE, patients undergoing MIO had statistically reduced occurrence of IHM (OR=0.69, 95%CI =0.55 -0.86). Patients undergoing MIO also had significantly reduced incidence of pulmonary complications (PCs) (RR=0.73, 95%CI = 0.63-0.86), pulmonary embolism (PE) (OR=0.71, 95%CI= 0.51-0.99) and arrhythmia (OR=0.79, 95%CI = 0.68-0.92). Non-significant reductions were observed among the included studies in the occurrence of anastomotic leak (AL) (OR=0.93, 95%CI =0.78-1.11), or Gastric Tip Necrosis (GTN) (OR=0.89, 95%CI =0.54-1.49). Most of the included studies were non-randomized case-control studies, with a diversity of study designs, demographics of participants and surgical intervention. Minimally invasive oesophagectomy (MIO) has superiority over open oesophagectomy (OE) in terms of the occurrence of in-hospital mortality (IHM) and should be the first-choice surgical procedure in esophageal surgery.

Background: The progressive, multifactorial and multistep dynamic process of metastasis is the primary cause of breast cancer (BC) lethality. PROX1 (Prospero-related homeobox 1), as a type of transcription factor that plays a key role in the formation of lymphatic vessels in animal embryonic development, has been proven to promote or suppress cancer in a variety of malignant tumors. However, molecular mechanisms behind PROX1 induced breast cancer metastases remain elusive.

Compared with open esophagectomy (OE), minimally invasive esophagectomy (MIE) proves to have clear benefits in reducing the risk of pulmonary complications for patients with resectable esophageal cancer. The objectives of our study were to explore the superiority of MIE in reducing the occurrence of anastomotic leakages (ALs) when compared to OE. A systematic review and meta-analysis was performed to assess the superiority of MIE on the occurrence of ALs over OE, by searching many sources (through December, 2014) such as Medline, Embase, Wiley Online Library, and Cochrane Library. Fixed-effects model was used to calculate summary odds ratios (ORs) to quantify associations between OE and MIE groups. Cochran’s Q and I 2 statistics were used to evaluate heterogeneity among studies. Among a total of 43 studies involving 5537 patients included in the meta-analysis, 2527 (45.6 %) cases underwent MIE and 3010 (54.4 %) cases underwent OE. Compared to patients undergoing OE, patients undergoing MIE did not have statistical significance in reduced occurrence of ALs (OR = 0.97, 95 % CI = 0.80–1.17). Insignificant reduced occurrence of ALs was not associated with anastomotic location (OR = 0.90, 95 % CI = 0.71–1.13) or anastomotic procedure (OR = 1.02, 95 % CI = 0.79–1.30). More proofs are needed to clarify the strengths or weaknesses of MIE in preventing anastomotic leakages after esophagectomy for cancer. A largely randomized, controlled trial should be undertaken to resolve this contentious issue urgently.

Introduction: Aquaporins (AQPs), also called water channels, have been shown to have functions in the migration, invasion, and proliferation of human breast tumor cells. Most AQP mRNA expression levels were tested by cell lines, mouse models, and even human breast tissues. However, the mRNA expression of individual AQPs in different clinicopathologic characteristics and prognostic values according to different kinds of classifications of breast cancer patients remains unclear. Materials and methods: In the current study, we used the Oncomine database, Breast cancer Gene-Expression Miner v4.1, Kaplan–Meier Plotter, and cBioPortal to investigate the expression distribution and prognostic values of AQPs in breast cancer patients. Results: Our study revealed that the mRNA expression levels of AQP8, AQP9, and AQP10 were upregulated, while those of AQP3, AQP4, AQP5, and AQP7 were downregulated in breast cancer. The clinical database showed that lower mRNA levels of AQP1 were associated with a high Scarff–Bloom–Richardson grade, but AQP9 showed the opposite trend. Further survival analyses indicated that high mRNA expression levels of AQP0, AQP1, AQP2, AQP4, AQP6, AQP8, AQP10, and AQP11 were significantly associated with better relapse-free survival (RFS). Conversely, AQP3 and AQP9 were associated with worse RFS in breast cancer patients, suggesting that these two genes might be potential targets in future chemotherapy. Discussion: These significant AQP members might be further explored as new biomarkers for breast cancer prognosis, but this needs further study.

Background: As the most popular beverage in East Asia, green tea (GT) has various biological activities effects such as anti-mutation, anti-oxidation, and anti-tumor. In this study, we aimed to evaluate whether GT consumption could be an effective way to decrease the risk of breast cancer. Methods: We had performed a systematic review and updated meta-analysis of published case–control studies to evaluate the association between GT intake and the risk of breast cancer. Searching strategies were performed by the following keywords “Breast cancer,” “breast neoplasm,” and “green tea,” with derivations and different combinations. The following databases were searched: PubMed, Cochrane Library, EMBASE, Web of science, China National Knowledge Infrastructure, WanFang, and China Biology Medicine disc. Studies published in both English and Chinese were considered for inclusion. Risk of bias was assessed through the Newcastle-Ottawa Scale (NOS). All data were analyzed through using Review Manager 5.1 software. Results: Fourteen studies fulfilled inclusion criteria for meta-analysis, yielding a total of 14,058 breast cancer patients and 15,043 control subjects. Individuals with the habit of drinking GT were found to have a negative association with the risk of future breast cancer (odds ratio 0.83; 95% confidence interval: 0.72–0.96) despite significant heterogeneity. In subgroup analyses, the negative correlation was still found in studies using registry-based controls, NOS grades ≥6 and the number of cases <500. Conclusions: GT consumption may have a decreased incidence of breast cancer despite significant heterogeneity. However, owing to the quality of available studies, more properly designed trials are warranted to clarify the association between GT consumption and breast cancer.

Multiple cancers have been reported to be associated with angiogenesis and are sensitive to anti‐angiogenic therapies. Vascular normalization, by restoring proper tumor perfusion and oxygenation, could limit tumor cell invasiveness and improve the effectiveness of anticancer treatments. However, the underlying anticancer mechanisms of antiangiogenic drugs are still unknown. Metformin (MET) and simvastatin (SVA), two metabolic‐related drugs, have been shown to play important roles in modulating the hypoxic tumor microenvironment and angiogenesis. Whether the combination of MET and SVA could exert a more effective antitumor effect than individual treatments has not been examined. The antitumor effect of the synergism of SVA and MET was detected in mouse models, breast cancer patient‐derived organoids, and multiple tumor cell lines compared with untreated, SVA, or MET alone. RNA sequencing revealed that the combination of MET and SVA (but not MET or SVA alone) inhibited the expression of endothelin 1 (ET‐1), an important regulator of angiogenesis and the hypoxia‐related pathway. We demonstrate that the MET and SVA combination showed synergistic effects on inhibiting tumor cell proliferation, promoting apoptosis, alleviating hypoxia, decreasing angiogenesis, and increasing vessel normalization compared with the use of a single agent alone. The MET and SVA combination suppressed ET‐1‐induced hypoxia‐inducible factor 1α expression by increasing prolyl hydroxylase 2 (PHD2) expression. Furthermore, the MET and SVA combination showed a more potent anticancer effect compared with bosentan. Together, our findings suggest the potential application of the MET and SVA combination in antitumor therapy.
