In traditional Chinese Medicine (TCM), the licorice-yuanhua herbal pair is one of the most representative incompatible herbal pairs recorded in the “eighteen incompatible herbal pairs” theory. Previous studies of our research group have demonstrated several gut-related side-effects induced by the licorice-yuanhua herbal pair. In this study, we investigated whether and why this incompatible herbal pair could induce gut tissue damage. After licorice-yuanhua treatment, the duodenum, ileum, and colon and serum biomarkers of mice were examined by pathological staining, Western blot, and ELISA assays. The IEC-6 cells and LS174T cells were treated with licorice saponins, yuanhua flavonoids, and di-terpenes; iTRAQ-labeled proteomic technology was then used to explore their synergistic effects on mucosa cells, followed by verification of ZO-1 and MUC-2 protein expressions. The results showed that the licorice-yuanhua herbal pair induced ileum tissue injuries, including epithelial integrity loss, inflammation, and edema. These injuries were verified to be related to epithelial and mucous barrier weakening, such as downregulated ileum ZO-1 and MUC-2 protein expressions. Proteomic analysis also suggested that glycyrrhizic acid and genkwanin synergistically influence tight junction pathways in LS174T cells. Furthermore, licorice saponins, yuanhua flavonoids, and di-terpenes dose/structure-dependently downregulate ZO-1 and MUC-2 protein expressions in mucosa cells. Our study provides different insights into the incompatibility mechanisms and material basis of the licorice-yuanhua herbal pair, especially that besides toxic di-terpenes, licorice saponins and yuanhua flavonoids, which are commonly known to be non-toxic compounds, can also take part in the gut damage induced by the licorice-yuanhua herbal pair.

Traditional Chinese medicine (TCM) is an important part of Chinese culture and the Chinese pharmaceutical industry. The adoption of stricter requirements for the quality of medicines has resulted in the loss of conformity of some traditional techniques used to prepare TCMs modern pharmaceutical methods. The TCM industry has begun to seek out novel methods to improve product quality and increase economic efficiency. Membrane technology is efficient, energy saving, and environmentally friendly. It is widely used in food processing, desalination, wastewater treatment, and in the medical industry. Membrane technology has also been adopted by the TCM industry as an alternative to traditional technology and has achieved good results. At present, membrane technology has been used in the separation, purification and concentration of TCMs. This review describes the use of microfiltration, ultrafiltration, nanofiltration, reverse osmosis, and membrane distillation, in TCM processing that have been reported. Membrane fouling is the most important obstacle in the use of membrane-based processes by the TCM industry. The causes and control of membrane fouling associated with TCM extracts are also analyzed.

The aim of this work was to encapsulate sea buckthorn (Hippophae rhamnoides L.) pulp oil (SBPO) by spray drying. Gum Arabic (GA) and maltodextrins (MD) were used as wall materials. The effects of several factors, including GA to MD ratio, total solids content of emulsion, wall to core ratio, and inlet air temperature, on the microencapsulation efficiency (ME) were investigated. The optimization of operation conditions was realized by response surface methodology (RSM). The optimal conditions were as follows: GA to MD ratio 2.38, total solids content 39%, wall to core ratio 5.33, and inlet air temperature 154°C. Under the optimal conditions, the ME of SBPO microcapsules was 94.96 ± 1.42%. The physicochemical properties of microcapsules were also invested. SBPO microcapsules obtained had low water activity, low moisture content, high water solubility, and high bulk density. For the morphological characteristics, cracks and pores were not observed in most microcapsules, which was beneficial for the protection of ingredients in microcapsules. The particle size of SBPO microcapsules ranged from 0.01 to 5 μm, and the mean diameter d4,3 was 2.55 μm. The analysis results of fourier transform infrared spectroscopy (FTIR) informed the presence of SBPO in microcapsules. There were no significant differences in the content of the main fatty acids in SBPO before and after spray drying. The results of oxidative stability showed that the microencapsulation by spray drying could effectively protect SBPO from oxidation and extend the shelf life of SBPO.

Siji-kangbingdu mixture is an anti-inflammatory, anti-bacterial, and anti-viral herbal mixture which is frequently used by doctors to treat upper respiratory infections. It’s important to establish an efficient and economical quality-control method to ensure the quality consistency and efficacy stability of Siji-kangbingdu mixture. In this study, an integrated multi-evaluation method was established, sequentially involving UPLC-TripleTOF-MS analysis, UPLC fingerprint analysis, and the quantitative analysis of multi-components using the single-marker (QAMS) method. With one chromatographic condition, a total of 71 compounds were identified by MS and MS/MS information, with a mass error of less than 5 ppm; 49 peaks detected in 254 nm were selected to establish the fingerprint similarity model, and 7 chemical compounds were simultaneously determined, namely, chlorogenic acid, liquiritin, rutin, isochlorogenic acid A, forsythin, forsythoside A, and glycyrrhizic acid, with forsythoside A as the reference standard. There was no significant difference in the content of the seven compounds between the QAMS method and the external standard method (ESM). The established multi-evaluation method will largely promote the quality control and standardization process of Siji-kangbingdu mixture. It also provides a reference workflow for the overall evaluation of TCM patent medicines, from chemical profiling to fingerprint and quantitative analysis.

Baihe Zhimu Tang (BZT) is a famous traditional Chinese medicine recipe to treat dry coughing due to yin deficiency and for moisturizing the lungs. Zhimu is an essential ingredient in BZT used to treat inflammation, fever and diabetes. The most important active components in Zhimu are flavonoids such as neomangiferin, mangiferin, and steroid saponins (e.g., timosaponin BII, anemarsaponin BIII, timosaponin AIII). The aim of this study was to compare the pharmacokinetics of mangiferin, neomangiferin, timosaponin BII, anemarsaponin BIII and timosaponin AIII in rat plasma after oral administration of BZT and Zhimu extract (ZME). A sensitive, reliable and robust LC-MS/MS method to simultaneously determine steroid saponins and flavonoids in rat plasma was successfully validated. Significant differences (p < 0.05) were found in the pharmacokinetic parameters of timosaponin BII, anemarsaponin BIII and timosaponin AIII between BZT and ZME. It was surmised that formula compatibility could significantly influence the pharmacokinetics of BZT and our study is the first to study the administration of BZT based on pharmacokinetic studies.

This study examined the behavior and penetration mechanisms of typical phenolic (benzoic) acids, which determine their observed penetration rates during membrane separation, focusing on the influence of electrostatic and hydrophobic solute/membrane interactions. To understand the effects of hydrophobicity and electrostatic interaction on membrane filtration, the observed penetration of five structurally similar phenolic acids was compared with regenerated cellulose (RC) and polyamide (PA) membranes at different solute concentrations and solution pHs. Variation partitioning analysis (VPA) was performed to calculate the relative contributions of electrostatic and hydrophobic effects. The penetration of phenolic acids was mainly influenced by the electrostatic interaction, with salicylic acid having the highest penetration. Penetration of phenolic acids through the PA membrane decreased from 98% at pH 3.0 to 30–50% at pH 7.4, indicating the dominance of the electrostatic interaction. Moreover, based on its hydrophobicity and greater surface charge, the PA membrane could separate binary mixtures of protocatechuic/salicylic acid and 4-hydroxybenzoic/salicylic acid at pH 9.0, with separation factors of 1.81 and 1.78, respectively. These results provide a greater understanding of solute/membrane interactions and their effect on the penetration of phenolic acids through polymeric ultrafiltration membranes.
