Single-nucleus gene expression: Identifying the genes expressed at the level of a single cell nucleus can better help us understand the human brain. Blue et al. developed a single-nuclei sequencing technique, which they applied to cells in classically defined Brodmann areas from a postmortem brain. Clustering of gene expression showed concordance with the area of origin and defining 16 neuronal subtypes. Both excitatory and inhibitory neuronal subtypes show regional variations that define distinct cortical areas and exhibit how gene expression clusters may distinguish between distinct cortical areas. This method opens the door to widespread sampling of the genes expressed in a diseased brain and other tissues of interest. Science , this issue p. 1586

In order to approximate the underlying process of temporal data, time series prediction has been a hot research topic for decades. Developing predictive models plays an important role in interpreting complex real-world elements. With the sharp increase in the quantity and dimensionality of data, new challenges, such as extracting deep features, recognizing deep latent patterns have emerged, demanding novel approaches and effective solutions. Deep learning, composed of multiple processing layers to learn with multiple levels of abstraction, are now commonly deployed for overcoming the newly arisen difficulties. This paper reviews state-of-the-art developments in deep learning for time series prediction. Based on modeling for the perspective of conditional or joint probability, we categorize them into discriminative, generative and hybrids models. Experiments are implemented on both benchmarks and real-world data to elaborate the performance of representative deep learning-based prediction methods. Finally, we conclude with comments on possible future perspectives and ongoing challenges with time series prediction.

The vehicular announcement network is one of the most promising utilities in the communications of smart vehicles and in the smart transportation systems. In general, there are two major issues in building an effective vehicular announcement network. First, it is difficult to forward reliable announcements without revealing users' identities. Second, users usually lack the motivation to forward announcements. In this paper, we endeavor to resolve these two issues through proposing an effective announcement network called CreditCoin, a novel privacy-preserving incentive announcement network based on Blockchain via an efficient anonymous vehicular announcement aggregation protocol. On the one hand, CreditCoin allows nondeterministic different signers (i.e., users) to generate the signatures and to send announcements anonymously in the nonfully trusted environment. On the other hand, with Blockchain, CreditCoin motivates users with incentives to share traffic information. In addition, transactions and account information in CreditCoin are tamper-resistant. CreditCoin also achieves conditional privacy since Trace manager in CreditCoin traces malicious users' identities in anonymous announcements with related transactions. CreditCoin thus is able to motivate users to forward announcements anonymously and reliably. Extensive experimental results show that CreditCoin is efficient and practical in simulations of smart transportation.

Recent developments in high-throughput technologies have accelerated the accumulation of massive amounts of omics data from multiple sources: genome, epigenome, transcriptome, proteome, metabolome, etc. Traditionally, data from each source (e.g., genome) is analyzed in isolation using statistical and machine learning (ML) methods. Integrative analysis of multi-omics and clinical data is key to new biomedical discoveries and advancements in precision medicine. However, data integration poses new computational challenges as well as exacerbates the ones associated with single-omics studies. Specialized computational approaches are required to effectively and efficiently perform integrative analysis of biomedical data acquired from diverse modalities. In this review, we discuss state-of-the-art ML-based approaches for tackling five specific computational challenges associated with integrative analysis: curse of dimensionality, data heterogeneity, missing data, class imbalance and scalability issues.
