The classical Osborne wheat protein fractions (albumins, globulins, gliadins, and glutenins), as well as several proteins from each of the four subunits of gliadin using SDS-PAGE analyses, were determined in the grain of five bread (T. aestivum L.) and five durum wheat (T. durum Desf.) genotypes. In addition, content of tryptophan and wet gluten were analyzed. Gliadins and glutenins comprise from 58.17% to 65.27% and 56.25% to 64.48% of total proteins and as such account for both quantity and quality of the bread and durum wheat grain proteins, respectively. The ratio of gliadin/total glutenin varied from 0.49 to 1.01 and 0.57 to 1.06 among the bread and durum genotypes, respectively. According to SDS-PAGE analysis, bread wheat genotypes had a higher concentration of α + β + γ-subunits of gliadin (on average 61.54% of extractable proteins) than durum wheat (on average 55.32% of extractable proteins). However, low concentration of ω-subunit was found in both bread (0.50% to 2.53% of extractable proteins) and durum (3.65% to 6.99% of extractable proteins) wheat genotypes. On average, durum wheat contained significantly higher amounts of tryptophan and wet gluten (0.163% dry weight (d.w.) and 26.96% d.w., respectively) than bread wheat (0.147% d.w. and 24.18% d.w., respectively).
During post-harvest maturation, different species vary in the length of dormancy breaking or germination increases. Seed dormancy and slow seedling development often limit establishment of forage grass stands. Seed germination and seedling vigour of Italian ryegrass (Lolium italicum A. Braun, Synonym Lolium multiflorum L.), cocksfoot (Dactylis glomerata L.) and timothy (Phleum pretense L.) were observed after harvest and storage. After harvest in June, seeds were stored under standard storage conditions and sampled every 30 days after harvest (DAH), up to 270 DAH, and then every 60 days up to 990 DAH. At each date, seeds were tested for final germination percentage and for seedling vigour traits. Timothy seeds had a maximum germination (88%) and the best seedlings vigour at 90 DAH, which implies that early autumn (September-October) is the best sowing period for freshly harvested seeds of timothy. Timothy seed germination was poor from 270 DAH (73%). The best germination and vigour of Italian ryegrass and cocksfoot seedlings were between 270 and 330 DAH, which equates to spring sowing time (March-April) in the succeeding year. Cocksfoot and Italian ryegrass seeds maintained satisfactory germination levels up to 630 DAH (81%) and 810 DAH (81%), respectively. The data can serve for the determination of a proper storage duration management between harvest and sowing of the tested species under ambient conditions of south-eastern Europe.
When water stress develops post-anthesis, wheat (Triticum aestivum L.) plants have to rely increasingly on remobilization of previously stored assimilates to maintain grain filling. In two-year field trials, we studied more than 20 agronomic and developmental traits in 61 wheat genotypes (27 F4:5 families, 17 parents used for the crosses and 17 standards), comparing plants that were defoliated (DP) by cutting off all leaf blades 10 days after anthesis with intact control plants (CP). Estimated contributions of stem and sheath assimilate reserves to grain weight/spike were from 10–54% and from 24–84% in CP and DP plants, respectively. Stem-related traits were among key traits determining stem reserve contribution (SRC). The most important genetic variables in differentiating genotypes for stress tolerance were biomass/stem, stem reserves mobilization efficiency and grain filling rate (GFR). Balance among traits related to yield maintenance in DP were more important than their high values. In general F4:5 families (FAM), that had been crossed to combine typical breeding traits such as biomass and yield components, showed better tolerance under moderate stress than standards and parents.

SUMMARY: Assimilated carbon during vegetative and early reproductive growth in wheat is temporarily stored in stem internodes and leaf sheaths (LSs), and can later be remobilized and transported to developing grain. The present study was undertaken to determine the effect of several peduncle (the uppermost internode) morpho-anatomical and biochemical traits on grain weight, and to assess the contribution of the peduncle water-soluble carbohydrate (WSC) reserves shortly after anthesis to its variation. In 2-year field trials, 61 wheat genotypes were used (27 F4:5families, 17 parents used for the crosses and the 17 current best standards), comparing intact control plants (CP) with plants that were defoliated (DP) by cutting off all leaf blades 10 days after anthesis. Estimated contributions of peduncle (culm (C) and flag LS) assimilate reserves to grain weight/spike were from 0·06 to 0·31 and from 0·11 to 0·45 in CP and DP plants, respectively. In both CP and DP plants, a higher contribution was from the LS than from the C. High peduncle reserve mobilization efficiency, a longer exposed part of the peduncle and larger C storage capacity (through higher parenchyma and/or lower lignified area) were of specific benefit for maintaining grain weight in defoliated plants. F4:5families had higher transport capacity in the peduncle, but without any improvement in WSC-related traits compared with the best standards.
Post-anthesis drought affects wheat production worldwide, primarily through the limitation of grain filling. The enhanced remobilization of stem reserves induced by drought can provide considerable carbon sources for grain filling. The aim of this study, which was part of an ongoing wheat-breeding program targeting drought tolerance, was to assess the ability of 20 contrasting common wheat genotypes (2 cultivars, 8 F6:7 families (FAM), and 10 parent genotypes (PAR) used to make the families) to remobilize stem dry matter under water-stressed conditions simulated via defoliation 10 days after anthesis, and to estimate the contribution of stem dry matter remobilization to grain weight. In two-year field trials, the genotypes were scored for agro-physiological and peduncle morpho-anatomical traits. Stem reserve contribution to grain weight per spike was significantly enhanced in defoliated plants but did not differ amongst the groups of genotypes. F6:7 families had higher grain-filling rate and 1000-grain weight but without improvement in grain weight per spike under defoliation compared with parental groups. The total area of chlorenchyma, phloem-area-related traits, and stem reserve contribution to grain weight were positively associated with grain weight per spike under defoliation, whilst in both treatments, the grain-filling rate was determined by stem height. These results imply that the grain-filling rate is a trait desirable for drought tolerance that can be improved during the breeding process.

The objectives of this work were to determine whether and at which seed physiological maturity stage the diquat desiccant affects the tall oat-grass (Arrhenatherum elatius) seed quality and yield, as well as the proper storage period between harvest and sowing. Diquat desiccation was evaluated in applications during milk or dough seed maturation stages. Seeds conventionally produced and stored under traditional storage conditions were used for the analyses. Seed samples were drawn every 30th day after harvest (DAH). After the 240th DAH, samples were drawn every 90th day up the to 690th DAH. The highest yield were obtained by desiccation applied at the beginning of the seed dough stage, with a satisfactory seed quality. Both final germination and seedling growth parameters achieved their maximum values between 180th and 240th DAH. Oat-grass seeds preserved satisfactory level of final germination (75%) up to 420th DAH. The application of diquat desiccant at the beginning of seed dough maturity stage can be a good solution for seed production of tall oat-grass. Early spring is the best sowing period for freshly harvested seeds of tall oat-grass regarding germination and seedling growth.
