Communities are often associated with important structural characteristics of a complex network system, therefore detecting communities is considered to be a fundamental problem in network analysis. With the development of data collection technology and platform, more and more sources of network data are acquired, which makes the form of network as well as the related data more complex. To achieve integrative community detection of a multi-layer attributed network that involves multiple network layers together with their attribute data, effectively utilizing the information from the multiple networks and the attributes may greatly enhance the accuracy of community detection. To this end, in this article, we study the integrative community detection problem of a multi-layer attributed network from the perspective of matrix factorization, and propose a penalized alternative factorization (PAF) algorithm to resolve the corresponding optimization problem, followed by the convergence analysis of the PAF algorithm. Results of the numerical study, as well as an empirical analysis, demonstrate the advantages of the PAF algorithm in community discovery accuracy and compatibility with multiple types of network-related data.

Next-generation sequencing studies on cancer somatic mutations have discovered that driver mutations tend to appear in most tumor samples, but they barely overlap in any single tumor sample, presumably because a single driver mutation can perturb the whole pathway. Based on the corresponding new concepts of coverage and mutual exclusivity, new methods can be designed for de novo discovery of mutated driver pathways in cancer. Since the computational problem is a combinatorial optimization with an objective function involving a discontinuous indicator function in high dimension, many existing optimization algorithms, such as a brute force enumeration, gradient descent and Newton’s methods, are practically infeasible or directly inapplicable. We develop a new algorithm based on a novel formulation of the problem as nonconvex programming and nonconvex regularization. The method is computationally more efficient, effective and scalable than existing Monte Carlo searching and several other algorithms, which have been applied to The Cancer Genome Atlas (TCGA) project. We also extend the new method for integrative analysis of both mutation and gene expression data. We demonstrate the promising performance of the new methods with applications to three cancer datasets to discover de novo mutated driver pathways.

We consider the one sample location testing problem for high-dimensional data, where the data dimension is potentially much larger than the sample size. We devise a novel inverse norm sign test (INST) that is consistent and has much improved power than many existing popular tests. We further construct a general class of weighted spatial sign tests which includes these existing tests, and show that INST is the optimal member within this class, in that it is consistent and is uniformly more powerful than all other members. We establish the asymptotic null distribution and local power property of the class of tests rigorously. Extensive numerical experiments demonstrate the superiority of INST in terms of both efficiency and robustness.
