The objective of this study was to evaluate the effect of body condition score (BCS), body weight (BW), average daily weight gain (ADG), and age at first calving (AFC) of Holstein heifers on production and reproduction parameters in the 3 subsequent lactations. The data set consisted of 780 Holstein heifers calved at 2 dairy farms in the Czech Republic from 2007 to 2011. Their BW and BCS were measured at monthly intervals during the rearing period (5 to 18 mo of age), and the milk production and reproduction data of the first 3 lactations were collected over an 8-yr period (2005 to 2012). The highest milk yield in the first lactation was found in the group with medium ADG (5 to 14 mo of age; 0.949 to 0.850 kg of ADG). The highest average milk yield over lifetime performance was detected in heifers with the highest total ADG (≥ 0.950 kg/d). The difference in milk yield between the evaluated groups of highest ADG (in total and postpubertal growth ≥ 0.950 kg/d and in prepubertal growth ≥ 0.970 kg/d) and the lowest ADG (≤ 0.849 kg/d) was approximately 1,000 kg/305 d per cow. The highest milk yield in the first lactation was found in the group with the highest AFC ≥ 751 d, for which fat and protein content in the milk was not reduced. Postpubertal growth (11 to 14 mo of age) had the greatest effect on AFC. The group with lowest AFC ≤ 699 d showed a negative effect on milk yield but only in the first 100 d of the first parity. The highest ADG was detrimental to reproduction parameters in the first lactation. The highest BW at 14 mo (≥ 420 kg) led to lower AFC. Groups according to BCS at 14 mo showed no differences in AFC or milk yield in the first lactation or lifetime average production per lactation. We concluded that low AFC ≤ 699 d did not show a negative effect on subsequent production and reproduction parameters. Therefore, a shorter rearing period is recommended for dairy herds with suitable management.

We are entering an exciting epoch in livestock biotechnology during which the fundamental approaches (such as transgenesis, spermatozoa cryopreservation and artificial insemination) will be enhanced based on the modern understanding of the biology of spermatogonial stem cells (SSCs) combined with the outstanding recent advances in genomic editing technologies and in vitro cell culture systems. The general aim of this review is to outline comprehensively the promising applications of SSC manipulation that could in the nearest future find practical application in livestock breeding. Here, we will focus on 1) the basics of mammalian SSC biology; 2) the approaches for SSC isolation and purification; 3) the available in vitro systems for the stable expansion of isolated SSCs; 4) a discussion of how the manipulation of SSCs can accelerate livestock transgenesis; 5) a thorough overview of the techniques of SSC transplantation in livestock species (including the preparation of recipients for SSC transplantation, the ultrasonographic-guided SSC transplantation technique in large farm animals, and the perspectives to improve further the SSC transplantation efficiency), and finally, 6) why SSC transplantation is valuable to extend the techniques of spermatozoa cryopreservation and/or artificial insemination. For situations where no reliable data have yet been obtained for a particular livestock species, we will rely on the data obtained from studies conducted in rodents because the knowledge gained from rodent research is translatable to livestock species to a great extent. On the other hand, we will draw special attention to situations where such translation is not possible.

The objectives were to evaluate the effects of sire and content of fresh or ionised egg yolk in extenders on sperm motility and share of live and dead sperm in collected ejaculate, in thawed artificial insemination (AI) doses, and during thermodynamic testing compared to extenders not containing egg yolk. Ejaculates were collected once a week from 4 Holstein bulls. Each of the 20 ejaculate samples from each bull was diluted with 4 different extenders. AndroMed and Bioxcell (no egg yolk) and Triladyl and Optidyl (fresh, ionised egg yolk) were used. A total of 640 AI doses were analysed. The volume of samples, sperm concentration, and percentage of motile spermatozoa were evaluated after collection, as was sperm motility after thawing of AI doses and during thermodynamic testing. Percentages of live and dead sperm were also evaluated. The data set was analysed using SAS/STAT 9.1 (SAS Institute Inc., Cary, NC, USA). The results confirmed significant (P<0.05–0.01) between-sire differences in the volume, density, and activity of sperm as well as in share of live and dead sperm after collection; in decline of sperm motility in the fresh ejaculate, after thawing, and during the entire thermodynamic test, as well as in the share of live and dead sperm after thawing. The extenders ranked by sperm motility are: Optidyl, Triladyl, AndroMed, and Bioxcell, demonstrating the higher quality of AI doses produced using egg yolk extenders. Differences in sperm motility were significant (P<0.05–0.01) during the entirety of thermodynamic testing. Egg yolk extenders had a significantly (P<0.05–0.01) higher percentages of live sperm after thawing.

The present study compared the fatty acid profile and some physical parameters of eggs from hens reared according to the organic system at the beginning of the first and second laying cycle. A total of 1080 eggs were analysed at the beginning of the first (from the 28th to 30th week of age) and the second (from the 78th to 80th week of age) laying cycle. It was found that the hen ages influenced the egg weight, egg surface area, yolk proportion, and eggshell colour. Albumen and eggshell proportion, albumen, yolk index, Haugh unit score, and eggshell strength were lower in eggs from older hens compared with those produced from younger layers. Monounsaturated fatty acids were found in higher amounts than saturated fatty acids and polyunsaturated fatty acids in egg yolks of eggs from layers only at the beginning of the second laying cycle. The PUFAn-6/n-3 ratio, saturation, atherogenic, and thrombogenic indices were significantly lower in the egg yolks from older hens compared to younger layers. These findings (regarding the eggs from the older ones) prove that it is practical to utilize them in the organic farming system during a period of two years.
