Isoëtes sabatina is the rarest aquatic quillwort in Europe. Although recently found (2013) in Lake Bracciano (central Italy), the species is just one step away from extinction with an estimated population not exceeding 400 individuals and a spatial range of a few hundred square metres. Lake Bracciano is a deep, oligo-mesotrophic Mediterranean volcanic lake that has been subjected to human activities. From January to October 2017, the lake experienced a dramatic water level decrease (up to −1.50 m), which significantly affected the littoral zone and the habitat of I. sabatina. To improve the chances of survival of I. sabatina, the first eco-taxonomic investigation on this species was carried out to describe its genetic distinctness, physical and chemical requirements and companion species. The phylogenetic position of I. sabatina was investigated by applying standard DNA barcoding methods. Simultaneously, during summer 2019, the physical and chemical features of water and sediments of the I. sabatina population and five small Alpine lakes colonized by Isoëtes echinospora – a supposed close relative – were characterized. These data were then compared with the available data on the trophic requirements of the target obligate aquatic Isoëtes, together with Isoëtes lacustris and Isoëtes malinverniana. The present survey confirmed the taxonomic and ecological distinctness of I. sabatina – providing the first evidence of genetic differentiation from I. echinospora. Isoëtes sabatina grows in waters with temperature, conductivity and total alkalinity up to 30°C, 561 μS cm−1 and 3.45 meq L−1, respectively. The edaphic requirements of I. sabatina confirm its outstanding conservation value, and this study offers a basic understanding of how to prevent its extinction. Now, all possible actions must be taken immediately to save this species.

Here, successful germination protocols for the following four annual pansies are presented:Viola arvensis subsp. arvensis, V. hymettia, V. kitaibeliana, andV. tricolor subsp. tricolor. Seedswere collected mainly in Central Italy (Tuscany, Umbria, and Latium) and in Sicily. The germi-nation ability was tested at the Tuscia Germplasm Bank (BGT) using several methods for dor-mancy-breaking. Our results show high germination percentages and germination rate for thetested species.

Isoëtes sabatina is an aquatic quillwort endemic to Italy. It is one of the rarest quillworts in Europe, and is critically endangered due to restricted range and to the continuous decline of both population and habitat quality. This study aims to develop an optimized protocol to reproduce and grow I. sabatina sporelings. Mature and immature megaspores were mixed with mature microspores to evaluate the influence of the developmental stage on germination and sporeling development. Two substrates, distilled water and water-agar medium, were tested for germination and sporeling emergence, and three substrates, sand, lake sediment and water-agar, were tested for transplants. A high percentage of megaspore germination (a total of 79.1%) was obtained in both substrates, higher for mature than immature spores. A total of 351 sporelings were produced in distilled water and water-agar cultures, with similar percentages (64.5% and 69.6%, respectively). The development stage of the megaspores affected both germination and sporeling development. Sporeling emergence showed significantly higher percentages in mature megaspores than immature ones (69.6% vs. 11.6%, respectively), with 85% of germinated spores developing sporelings. Only transplants over water-agar medium were successful. This protocol could be useful for the propagation of sporelings as the key step towards the planning of in situ actions to save this Mediterranean quillwort from extinction.

The germination ability was tested at the Tuscia Germplasm Bank(BGT) at six constant temperatures (from 5 to 30°C), under both light (with a 12/12h pho-toperiod) and total dark. Our results show a high germination ability and germination ratefor the tested species.

The census of the loci classici of 1,227 native, non-endemic vascular plants described from Italy is here presented and described. The effective place of publication of accepted names, basionyms and homotypic synonyms were identified and critically verified. The geographic information on the loci classici was excerpted from the protologues, as well as information on typification for the taxa described before 1 January 1958. The names without a holotype are 1,165. For 591 names a lecto- or neo-typification is available in literature, while 572 currently accepted taxa still need a type designation. For ten of these names showing previous ineffective typification, nomenclatural types are designated here (Allium savii, A. tenuiflorum, Anemone millefoliata, Catapodium tuberculosum, Cynosurus siculus, Filago congesta, Saponaria bellidifolia, Sclerochloa patens, S. zwierleinii, and Vicia leucantha). A new combination (Trisetaria aurea comb. nov.) is proposed. The general picture of the currently accepted taxa of vascular plants described from Italy, including endemics, amounts to 2,631, i.e. about 32% of the native flora currently recorded for the country.

This integrated study provides new insights into pollen and seed morphology and pollen heteromorphism of four closely related annual taxa of Viola sect. Melanium. The plant material, both fresh and dried, was collected in Italy and studied using light and scanning electron microscope. Palynological data for V. hymettia together with a detailed comparative analysis of seed morphology and micromorphology of the four species are reported for the first time. Results of this work highlight some pollen and seed features as useful diagnostic characters. The pollen size proves to be of diagnostic value to easily separate V. kitaibeliana, having the smallest pollen grains, from the others, especially from V. arvensis with the largest ones. Exine ornamentation is microreticulate, showing no relevant differences among species. We can partially confirm the diagnostic value of the prevailing pollen morph as it can be useful only for V. arvensis (five-aperturate) versus V. tricolor and V. hymettia (four-aperturate). The macro- and micromorphology of seeds provide additional useful distinguishing characters. Particularly, seed size was found to be a good delimitating character, especially to distinguish V. kitaibeliana (with the smallest seeds) from V. arvensis, and easy to be measured with no need of particular equipment.

The need to optimize seed banking efforts has stimulated research for rapid methods to estimate quality in seed-lots. For terrestrial orchids, viability testing using tetrazolium (TTC) staining requires chemical scarification, as seeds have an impermeable testa. Different seed-coat permeability may affect TTC staining, thus affecting the results. The aim of this study was to perform a permeability test to assess the effectiveness of the used scarification method and its usefulness to correct TTC viability results. Mature seeds of Anacamptis laxiflora were subjected to eight scarification treatments with sodium hypochlorite solutions with different concentration and duration. Viability tests were performed using the basic TTC methodology, followed by a permeability test performed by means of trypan blue dye. The different scarification methods resulted in estimated TTC viability ranging from 0% and 94% for the same seed lot of A. laxiflora seeds. Our results proved that the used scarification protocol significantly affects both seed coat permeability and subsequent TTC staining (two-way ANOVA, p < 0.0001). We describe a new rapid protocol that can be used to test terrestrial orchid seed viability. This double-staining method, providing rapid information on seed coat permeability, can be useful to avoid under-estimation of TTC results.

Taxa endemic to a country are key elements for setting national conservation priorities and for driving conservation strategies, since their persistence is entirely dependent on national policy. We applied the IUCN Red List categories to all Italian endemic vascular plants (1340 taxa) to assess their current risk of extinction and to highlight their major threats. Our results revealed that six taxa are already extinct and that 22.4% (300 taxa) are threatened with extinction, while 18.4% (247; especially belonging to apomictic groups) have been categorized as Data Deficient. Italian endemic vascular plants are primarily threatened by natural habitat modification due to agriculture, residential and tourism development. Taxa occurring in coastal areas and lowlands, where anthropogenic impacts and habitat destruction are concentrated, display the greatest population decline and extinction. The national network of protected areas could be considered effective in protecting endemic-rich areas (ERAs) and endemic taxa, but ineffective in protecting narrow endemic-rich areas (NERAs), accordingly changes to the existing network may increase the effectiveness of protection. For the first time in the Mediterranean Basin biodiversity hotspot, we present a comprehensive extinction assessment for endemic plants under the full responsibility of a single country. This would provide an important step towards the prioritization and conservation of threatened endemic flora at Italian, European, and Mediterranean level. A successful conservation strategy of the Italian endemic vascular flora should implement the protected area system, solve some taxonomical criticism in poorly known genera, and should rely on monitoring threatened species, and on developing species-specific action plans.

Good‐quality dry seeds of some orchids have the potential to survive for decades under conventional seed‐bank conditions, but further research is needed to fill existing gaps in knowledge regarding seed behaviour under long‐term dry storage. The objectives of this study were to evaluate germination ability on two asymbiotic culture media with different nitrogen source; to assess seed desiccation tolerance needed for the storage at sub‐zero temperatures; to study the effects of dry storage at low temperature. Asymbiotic seed germination tests of four Anacamptis species were carried out to evaluate the effects of different culture media, dehydration, and dry storage on germination ability. Viability of 4‐year‐stored seeds was assessed by means of tetrazolium test. Generalised Linear Models (GLM) analysis detected significant effects (P < 0.01) of the species, medium, and storage time on total germination, while dehydration did not significantly affect it. Except for A. palustris, germination percentage had a minimum after 1‐month storage and increasing values with longer storage periods. Tetrazolium viability tests detected high percentages of viable seeds (> 90%) following 4‐year storage, in three out of four species. Seeds of the four Anacamptis species showed to be desiccation tolerant and to have an orthodox storage behaviour. The consequence of these findings is of interest to practical conservation approaches of orchids as seed‐banking. The results highlight the importance of multiple assessments of seed quality, both viability and germination, to understand seed storage behaviour. This article is protected by copyright.
