One of the wild fruit species with a natural distribution in Türkiye, and historically used for medicinal purposes due to its rich composition, is Berberis crataegina DC. Various parts of the plant, including its roots, bark, leaves, flowers, and fruits, have been utilized in traditional medicine, while its fruits are also consumed in various forms as food. This study aimed to characterize the morphological, biochemical, and molecular traits of B. crataegina genotypes naturally growing in the Kayseri region, located in central Türkiye. The fruit weight of the genotypes ranged from 0.047 to 0.137 g, fruit width from 3.06 to 4.64 mm, and fruit length from 5.80 to 9.05 mm. Similarly, the leaf traits of the genotypes exhibited wide variation. The total phenolic content ranged from 190.53 to 297.55 mg GAE/100 g, total flavonoid content from 82.03 to 203.89 mg QE/100 g, total anthocyanin content from 4.54 to 11.76 mg cyn-3 gluc/100 g, and total antioxidant capacity between 57.76 and 87.93%. A principal component analysis (PCA) of 11 traits identified four principal components with eigenvalues greater than 1. The first four components accounted for 71.89% of the total variation, with PCA1 explaining 23.48%, PCA2 18.68%, PCA3 16.39%, and PCA4 13.34%. ISSR molecular analysis using nine markers revealed a band count ranging from 4 to 13, polymorphic band count between 3 and 10, and polymorphism rates from 61.54 to 100%, with band lengths ranging from 200 to 1000 base pairs. According to the UPGMA dendrogram based on molecular analyses, the genetic similarity between the genotypes ranged from 0.64 to 1.00, with B2 and B13 being the most similar genotypes. In conclusion, the Kayseri region is rich in B. crataegina genotypes with wide genetic variation. The genotypes identified in this region may serve as valuable genetic resources for future studies.

Hawthorn (Cratageus monogyna Jacq.), one of the important wild fruit species in Turkey, is an important fruit species in many countries due to its use in traditional medicine, landscaping studies, and the food and beverage industry. In this study, morphological characterization revealed significant variation among genotypes, with fruit weight ranging from 0.55 g to 2.87 g, fruit width from 9.74 mm to 18.39 mm, and leaf width from 19.53 mm to 41.19 mm. Genotype-dependent variations were observed in the total phenolic content (151.34 mg–491.88 mg GAE 100 g−1 flesh), the total flavonoid content (16.74–48.13 mg CAE 100 g−1 flesh), the total anthocyanin content (26.34–79.79 mg cyn-3 gluc 100 g−1 flesh), and the antioxidant activity (29.20% to 56.70%). HPLC-based phenolic profiling identified substantial variations in key bioactive compounds, with the highest levels of chlorogenic acid (16.16 mg 100 g−1 fw), caffeic acid (10.21 mg 100 g−1 fw), epicatechin (13.83 mg 100 g−1 fw), rutin (74.05 mg 100 g−1 fw), and protocatechuic acid (2.00 mg 100 g−1 fw). ISSR marker-based molecular analysis revealed a high degree of genetic polymorphism (89.12%), with 55 out of 62 bands classified as polymorphic. The polymorphic information content values ranged from 0.34 to 0.44. The Jaccard similarity coefficient ranged from 0.04 (M9 and M16) to 0.63 (M17 and M3), indicating substantial genetic variability. The tanglegram analysis comparing genetic and morphological–biochemical dendrograms yielded an entanglement score of 0.714, indicating an alignment between molecular and phenotypic data. These findings show that hawthorn genotypes from the Kelkit Valley exhibit extensive genetic and biochemical diversity, which is critical for conservation efforts, breeding programs, and the development of high-value medicinal and functional food products.

Türkiye has a unique position globally as it connects the Asian and European continents. In this study, the genetic diversity of a total of 27 mulberry (Morus alba) genotypes collected from northeastern Türkiye was evaluated in terms of morphological, biochemical, and molecular characteristics. In the fruits of the mulberry genotypes, the fruit weight (FW) ranged from 2.00 to 5.13 g, fruit width (Fw) ranged from 12.64 to 16.93 mm, and fruit length (FL) ranged from 20.89 to 32.34 mm. Total soluble solids (TSS) ranged from 16.56% to 26.00%, fruit pH ranged from 3.87 to 5.10, total phenolic content (TPC) ranged from 72.69 to 171.34 mg GAE/100 g, total flavonoid content (TFC) ranged from 75.55 to 86.85 mg QE 100/g, and antioxidant activity (AA) ranged from 42.35% to 74.93%. According to principal component analysis (PCA), the first three principal components (eigenvalue ≥ 1.00) explain 54.05% of the total variation. In general, the fruit and leaf characteristics were more effective in the first two principal components, while the biochemical traits were more effective in the third principal component. As a result of molecular analysis using 13 inter-simple sequence repeat (ISSR) primers, 94.23% of the total of 76 bands was polymorphic. Molecular variance analysis (AMOVA) showed that within the population, genetic variation was 79%, and among the populations, the variation was 21%. The similarities of the genotypes in the UPGMA dendrogram created according to the results of the molecular analysis varied between 0.74 and 0.98, and the genotypes were divided into two main groups. This study guides breeders in future studies by evaluating mulberry genotypes from morphological, biochemical, and molecular perspectives and explaining the wide variation among genotypes.

Apple leaves are a rich source of bioactive compounds such as phenolics, flavonoids, and essential minerals, which exhibit significant antioxidant and therapeutic properties. This study focuses on comparing the biochemical composition, antioxidant capacity, and mineral contents of Malus domestica Borkh. cultivars and M. kirghisorum Al. Fed. & Fed. genotypes. The goal is to identify potential health-promoting compounds and establish a basis for utilizing apple leaves as a sustainable resource in the food, pharmaceutical, and cosmetic industries. The study revealed significant biochemical and nutritional variation among the genotypes. Total antioxidant capacity ranged from 36.00 in ‘A12’ to 59.50% in ‘Starking Delicious’. Total phenolic content varied between 70.42 in ‘A28’ and 147.45 mg GAE/100 g in ‘Granny Smith’, while total flavonoid content ranged from 15.43 in ‘A28’ to 38.66 mg QE/100 g in ‘A16’, demonstrating considerable variability in bioactive compound composition. Correlation matrix analysis identified several significant relationships. Total phenolics and total flavonoids showed a positive correlation (r = 0.52**), while calcium strongly correlated with magnesium (r = 0.79**), potassium (r = 0.52**), and phosphorus (r = 0.52**), underscoring their physiological interconnections. Multiple regression analysis clarified key traits. Total phenolic content was positively influenced by total flavonoids (β = 0.52, p < 0.00). Calcium was strongly associated with magnesium (β = 0.52, p < 0.00) and sodium (β = 0.46, p < 0.00), reflecting their synergistic roles in cellular and metabolic functions. Principal component analysis revealed that the first three components explained 57.80% of the total variation. PC1 (30.56% variance) was predominantly associated with calcium, potassium, phosphorus, and magnesium. PC2 (14.16%) highlighted the relationship between manganese and total antioxidant capacity, while PC3 (13.08%) reflected the influence of lead, total phenolics, and total flavonoids. Heat map analysis indicated that the calcium, phosphorus, sulfur, phenolic compounds, and antioxidant activities in subgroup A1 suggest that the genotypes may be beneficial for health. Additionally, the accumulation of heavy metals such as lead, nickel, and aluminum in subgroup B1 could pose a health risk; however, the genotypes ‘A18’, ‘A21’, ‘A21-1’, and ‘A22’ possess the capacity to reduce this accumulation. The results highlight the nutritional and therapeutic potential of apple leaves as a natural source of antioxidants and essential minerals. In particular, the genotypes ‘A21-1’ and ‘A16’ stand out due to their high content of bioactive compounds and nutrients, offering promising prospects for further research and applications. These findings contribute to the conservation of wild apple genetic resources and their potential for industrial use. Not applicable.
