Background: Cervical cancer continues to be a major public health problem worldwide, and Cisplatin is used as first-line chemotherapy for this cancer; however, malignant cells exposed to CISplatin (CIS) become insensitive to the effects of this drug. PenToXifylline (PTX) is a xanthine that sensitizes several types of tumor cells to apoptosis induced by antitumor drugs, such as Adriamycin, Carboplatin, and CIS. The effects of PTX on tumor cells have been related to the disruption of the NF-κB pathway, thus preventing the activation of cell survival mechanisms such as the expression of anti-apoptotic genes, the secretion of proinflammatory interleukins, and growth factors. Objective: In this work, we studied the antitumor proprieties of PTX in human SiHa cervical carcinoma cells resistant to CIS.Materials and Methods: SiHa and HeLa cervical cancer cells and their CIS-resistant derived cell lines (SiHaCIS-R and HeLaCIS-R, respectively) were used as in-vitro models. We studied the effects of PTX alone or in combination with CIS on cell viability, apoptosis, caspase-3, caspase-8, and caspase-9 activity, cleaved PARP-1, anti-apoptotic protein (Bcl-2 and Bcl-xL) levels, p65 phosphorylation, cadmium chloride (CdCl2) sensitivity, Platinum (Pt) accumulation, and glutathione (GSH) levels, as well as on the gene expression of GSH and drug transporters (influx and efflux). Results: PTX sensitized SiHaCIS-R cells to the effects of CIS by inducing apoptosis, caspase activation, and PARP-1 cleavage. PTX treatment also decreased p65 phosphorylation, increased Pt levels, depleted GSH, and downregulated the expression of the ATP7A, ATP7B, GSR, and MGST1 genes. Conclusion: PTX reverses the acquired phenotype of CIS resistance close to the sensitivity of parental SiHa cells.

A balanced and varied diet provides diverse beneficial effects on health, such as adequate micronutrient availability and a gut microbiome in homeostasis. Besides their participation in biochemical processes as cofactors and coenzymes, vitamins and minerals have an immunoregulatory function; meanwhile, gut microbiota and its metabolites coordinate directly and indirectly the cell response through the interaction with the host receptors. Malnourishment is a crucial risk factor for several pathologies, and its involvement during the Coronavirus Disease 2019 pandemic has been reported. This pandemic has caused a significant decline in the worldwide population, especially those with chronic diseases, reduced physical activity, and elder age. Diet and gut microbiota composition are probable causes for this susceptibility, and its supplementation can play a role in reestablishing microbial homeostasis and improving immunity response against Coronavirus Disease 2019 infection and recovery. This study reviews the role of micronutrients and microbiomes in the risk of infection, the severity of disease, and the Coronavirus Disease 2019 sequelae.

Cervical cancer (CC) is one of the most common and deadly types of female cancer worldwide. Late diagnosis in CC increases the risk of tumor cells spreading to distant organs (metastasis). The epithelial-mesenchymal transition (EMT) is a fundamental process of cancer metastasis. Inflammation can lead to tumor progression, EMT induction, and metastasis. The inflammatory microenvironment is a potent inducer of EMT; inflammatory cytokines such as Tumor Necrosis Factor-alpha (TNF-α) and Transforming growth factor-beta (TGF-β1) activate transcriptional factors such as STAT3, Snail, Smad, and the Nuclear Factor kappa light-chain-enhancer of activated beta cells (NF-κΒ), which drive EMT. Anti-inflammatory compounds may be an option in the disruption of EMT. PenToXifylline (PTX) possesses potent anti-inflammatory effects by inhibiting NF-κB activity. In addition, PTX exerts an anti-fibrotic effect by decreasing Smad2/3/4. We hypothesize that PTX could exert anti-EMT effects. CaSki human cervical tumor cells were exposed to TNF-α 10 ng/mL and TGF-β1 alone or in combination for 5 days. Our results revealed that TNF-α and TGF-β1 induced N-cadherin and Vimentin, confirming the induction of EMT. Furthermore, the combination of cytokines synergized the expression of mesenchymal proteins, enhanced IκBα and p65 phosphorylation, and upregulated Serpin family E member 1 (SERPINE1) mRNA. PTX pretreatment prior to the addition of TNF-α and TGF-β1 significantly reduced N-cadherin and Vimentin levels. To our knowledge, this is the first time that this effect of PTX has been reported. Additionally, PTX reduced the phosphorylation of IκB-α and p65 and significantly decreased SERPINE1 expression, cell proliferation, migration, and invasion. In conclusion, PTX may counteract EMT in cervical cancer cells by decreasing the NF-κB and SERPINE1.

Background/Aims. Prostate cancer (PCa) is one of the neoplasms with the highest incidence and mortality rate in men worldwide. Advanced stages of the disease are usually very aggressive, and most are treated with chemotherapeutic drugs that generally cause side effects in these patients. However, additional therapeutic targets such as the IL6R/STAT-3 axis and TIGIT have been proposed, mainly due to their relevance in the development of PCa and regulation of NK cell-mediated cytotoxicity. Here, we evaluate the effect of inhibitors directed against these therapeutic targets primarily via an analysis of NK cell function versus prostate cancer cells. Methods. We analyzed the secretion of cytokines, chemokines, and growth factors in 22Rv1, LNCaP, and DU145 cells. In these cells, we also evaluated the expression of NK ligands, IL6R, STAT-3, and phosporylated STAT-3. In NK-92 cells, we evaluated the effects of Stattic (Stt) and tocilizumab (Tcz) on NK receptors. In addition, we assessed if the disruption of the IL6R/STAT-3 pathway and blockade of TIGIT potentiated the cytotoxicity of NK-92 cells versus DU145 cells. Results. DU145 abundantly secretes M-CSF, VEGF, IL-6, CXCL8, and TGF-β. Furthermore, the expression of CD155 was found to increase in accordance with aggressiveness and metastatic status in the prostate cancer cells. Stt and Tcz induce a decrease in STAT-3 phosphorylation in the DU145 cells and, in turn, induce an increase of NKp46 and a decrease of TIGIT expression in NK-92 cells. Finally, the disruption of the IL6R/STAT-3 axis in prostate cancer cells and the blocking of TIGIT on NK-92 were observed to increase the cytotoxicity of NK-92 cells against DU145 cells through an increase in sFasL, granzyme A, granzyme B, and granulysin. Conclusions. Our results reveal that the combined use of inhibitors directed against the IL6R/STAT-3 axis and TIGIT enhances the functional activity of NK cells against castration-resistant prostate cancer cells.

Background/Objectives: Whether the COVID-19 pandemic uniformly increased antimicrobial resistance (AMR) is contested, and aggregate analyses may mask organism-specific patterns. The objective of the study was to characterize changes in the level and slope of monthly multidrug-resistant (MDR), possible extensively drug-resistant (pXDR), possible pandrug-resistant (pPDR), vancomycin-resistant enterococci (VRE), and methicillin-resistant Staphylococcus aureus (MRSA) positivity at a Mexican tertiary-care hospital, and to assess heterogeneity by organism, antimicrobial class, specimen type, and hospital service. Methods: Retrospective interrupted time-series of deduplicated bacterial isolates (October 2017 to January 2026), analyzed by segmented regression across pre-pandemic, pandemic, and post-peak phases. Results: Cohort-wide MDR positivity had been declining before 2020 and rose abruptly at pandemic onset (level shift +6.81 percentage points, p = 0.0092) with a simultaneous slope reversal, reaching 24.27 percentage points above the extrapolated pre-pandemic trend by February 2022 (95% CI 14.26–34.28, p < 0.0001). The decline resumed after the pandemic peak, but the two slope changes did not offset one another (joint p = 0.0064). pXDR and pPDR moved in the same direction (+8.93, p = 0.0233; +15.40, p = 0.0113). VRE showed no significant departure, whereas MRSA declined in every phase yet departed significantly from its extrapolated trajectory (+23.68, p = 0.0040). Among species, Escherichia coli showed the largest departure from its counterfactual (+48.82 percentage points, p < 0.0001) and Klebsiella pneumoniae had the largest abrupt level rise (+14.46 percentage points, p = 0.0055), while S. aureus declined in every phase despite an abrupt rise in level at pandemic onset. At the class level, carbapenem and third-/fourth-generation cephalosporin non-susceptibility rose at pandemic onset and returned to baseline afterwards (19.1% to 28.0% to 18.7%, and 51.7% to 59.4% to 52.5%; all q < 0.001), whereas fluoroquinolone non-susceptibility rose and did not revert, climbing further in E. coli (61.5% to 65.2% to 72.7%, q = 0.0012) and plateauing at an elevated level in K. pneumoniae (37.6% to 46.5% to 49.3%, q = 0.1919 post-peak). Conclusions: Resistance trajectories across pandemic-related phases were divergent and, for the Gram-negative organisms, incompletely reversed, with the highest burden of advanced resistance in high-intensity settings. Organism-resolved surveillance and sustained stewardship are needed.

Pancreatic ductal adenocarcinoma (PDAC) is rarely detected before the disease is advanced, and existing serum markers are insufficient for early detection. Its systemic immune alterations are incompletely characterized and rarely examined by sex. We measured 19 serum cytokines by bead-based multiplex assay in 22 patients with PDAC and 16 controls, comparing groups overall and separately in women and men, and examined co-regulation among them. In the full cohort, interleukin-7 (IL-7) was the only cytokine to discriminate patients from controls after correction for multiple testing. Adjustment for age strengthened this association, and IL-7 matched the discrimination of carbohydrate antigen 19-9 (CA19-9). After stratification by sex, IL-7 remained the only cytokine higher in women, though it did not survive correction within that subgroup, whereas in men, eight innate, T-helper 17 (Th17) and type I interferon cytokines (IL-1α, IL-1RA, IL-31, IFN-α, IL-17A, IL-17F, TNF-α, and TNF-β) separated patients from controls, although the male control group was small. The correlation network present in controls was largely absent in patients, which instead showed few associations centered on IL-2, IL-5 and IL-6. These findings identify IL-7 as a candidate discriminator of PDAC and show that the cytokines separating patients from controls differed by sex, warranting validation in a larger, sex-balanced cohort that also measures sex hormones.
