Abscisic acid (ABA) plays a central role in the plant response to water deficit by inducing stomatal closure to conserve water when the soil dries. Exogenous ABA was applied at 45 days after sowing (DAS) as a soil drench, the physiological and seed yield response of soybean to exogenous ABA were examined as the soil was drying. Three experiments were conducted using the drought-tolerant soybean cultivar Jindou 19, grown in pots at the Yuzhong Experimental Station of Lanzhou University, China. In experiment 1, plants were exposed to progressive soil drying and leaf ABA concentration, leaf photosynthesis rate, leaf relative water content (RWC) and osmotic adjustment (OA) were measured. In experiment 2, plants were under progressive soil drying and lethal leaf water potential was measured. In experiment 3, flower production and abortion, and grain yield were measured in plants under well-watered (WW), moderate (MWD) and severe water deficits (SWD). Exogenous ABA application increased ABA accumulation in leaves and reduced the rate of soil drying. It also increased leaf photosynthetic rate, stomatal conductance and transpiration rate at 7–10 days after withholding water. The intrinsic and instantaneous water use efficiency (WUE) was consistently higher with exogenous ABA than without ABA as the soil dried. Exogenous ABA increased OA when the leaf relative water content (RWC) decreased at eight days after withholding water, lowering the lethal leaf water potential by 0.4 MPa. Exogenous ABA reduced water use, increased WUE for grain yield under WW and MWD, and had no effect on flower number, flower abortion or grain yield in any water treatment. We concluded that (1) exogenous ABA induced OA, improved leaf photosynthetic rate, leaf water relations and desiccant tolerance, but did not benefit grain yield in soybean under water deficits; (2) exogenous ABA improved the WUE at the leaf level as soil drying and WUE for grain yield under moderate water deficit.

The effect of the mixture (1:1) of chemical and organic nitrogen (N) fertilizer on crop yield quality and N fertilizer use efficiency remains elusive. A nitrogen field experiment was conducted in the growing seasons of 2020 and 2021 to investigate the effects of the mixture of chemical and organic N fertilizer on the crop yield, crop quality and nitrogen fertilizer use efficiency in a maize–soybean intercropping system in China. Four treatments applied at 150 kg N ha−1 were used: no nitrogen fertilizer (CK), chemical N fertilizer (ChemF), mixture (1:1) of chemical and organic N fertilizer (ChemF + OrgF) and organic N fertilizer (OrgF). The results showed that the yield and aboveground N accumulation of both soybean and maize increased with the application of fertilizer. The ChemF + OrgF treatment had lower maize and soybean seed yields than for ChemF treatment, but higher than the other two treatments in both years, and the maize yield of the (ChemF + OrgF) treatment was significantly higher (14.9%) in 2021 than 2020. Yields were significantly positively correlated with aboveground N accumulation and fertilizer use efficiency, measured using the nitrogen partial productivity (NPP), nitrogen agronomic efficiency (NAE) and nitrogen fertilizer recovery rate (NFRR). The protein content tended to increase and the oil content tended to decrease under (ChemF + OrgF) applications in soybeans. The (ChemF + OrgF) treatment had the lowest starch content in maize. There was no significant difference in the nitrogen harvest index among treatments, while the NPP, NAE and NFRR were the highest for the application of chemical N fertilizer and significantly decreased with the addition of organic N fertilizer. We conclude that the mixture (1:1) of chemical and organic N fertilizer increased the seed yield and quality of maize, but only the seed yield of soybean.

Water stress is the main abiotic factor that limits soybean grain yield. We investigated eight soybean cultivars under well-watered (WW) and terminal drought stress (TDS) conditions to determine the traits associated with water saving and the relationship between water use, root morphology, canopy architecture, flower and tagged-pod number, and yield performance. Under WW conditions, the average grain yield across the new soybean cultivars was significantly higher (18.7 g plant−1 vs. 15.1 g plant−1), but significantly less water was used (36 L plant−1 vs. 47 L plant−1) than in the old soybean cultivars. Under TDS, the four old soybean cultivars failed to produce a measurable grain yield, while the new soybean cultivars Zhonghuang 30 (ZH) and Jindou 19 (J19) produced a measurable grain yield. Water stress significantly reduced the flower numbers and tagged-pod numbers; the four new soybean cultivars on average had low flower and tagged-pod numbers under WW treatment, while they had low flower but high tagged-pod numbers under TDS conditions. ZH and JD exhibited a lower branch number and leaf area under both WW and TDS conditions. Water use during the flowering and podding periods was significantly positively correlated with the flower number and the tagged-pod number under both WW and TDS conditions. Thus, the small canopy size and low root length and root surface area contributed to a water-saving mechanism in the new soybean cultivars and improved the yield under drought conditions.

The circadian system plays a pivotal role in facilitating the ability of crop plants to respond and adapt to fluctuations in their immediate environment effectively. Despite the increasing comprehension of PSEUDO-RESPONSE REGULATORs and their involvement in the regulation of diverse biological processes, including circadian rhythms, photoperiodic control of flowering, and responses to abiotic stress, the transcriptional networks associated with these factors in soybean (Glycine max (L.) Merr.) remain incompletely characterized. In this study, we provide empirical evidence highlighting the significance of GmPRR3b as a crucial mediator in regulating the circadian clock, drought stress response, and abscisic acid (ABA) signaling pathway in soybeans. A comprehensive analysis of DNA affinity purification sequencing and transcriptome data identified 795 putative target genes directly regulated by GmPRR3b. Among them, a total of 570 exhibited a significant correlation with the response to drought, and eight genes were involved in both the biosynthesis and signaling pathways of ABA. Notably, GmPRR3b played a pivotal role in the negative regulation of the drought response in soybeans by suppressing the expression of abscisic acid-responsive element-binding factor 3 (GmABF3). Additionally, the overexpression of GmABF3 exhibited an increased ability to tolerate drought conditions, and it also restored the hypersensitive phenotype of the GmPRR3b overexpressor. Consistently, studies on the manipulation of GmPRR3b gene expression and genome editing in plants revealed contrasting reactions to drought stress. The findings of our study collectively provide compelling evidence that emphasizes the significant contribution of the GmPRR3b-GmABF3 module in enhancing drought tolerance in soybean plants. Moreover, the transcriptional network of GmPRR3b provides valuable insights into the intricate interactions between this gene and the fundamental biological processes associated with plant adaptation to diverse environmental conditions.

Water is the main factor limiting soybean yield and the timely supply of supplemental irrigation could increase the grain yield, but the effects of a supplemental water supply on soybean yields have not been well studied. Field and pot experiments were conducted to compare the grain yield, yield components, water use efficiency for grain yield (WUEG), flower number, filled-pod number, soil water content, and root dry weight at different depths with and without supplemental irrigation at flowering. Field experiments showed that compared to rainfed conditions, 40 mm of water applied during flowering significantly increased grain yield by 26%, WUEG by 12%, filled-pod number by 16%, grain number by 13.3%, and water uptake from soil by 11% in 2011, and increased grain yield by 22%, WUEG by 7%, filled-pod number by 26%, grain number by 27%, and water uptake by 21% in 2012. The soil water content in the subsoil (1.2–2.0 m) layers under the irrigated treatment was lower, indicating greater water extraction, than in the rainfed treatment and water uptake was significantly and positively correlated with yield in both years. In a pot experiment, flower and filled-pod number, water use during flowering and podding were significantly higher in the well-watered (WW) treatment than cyclic water stress (WS) treatment. Flower number and filled-pod number were significantly and positively correlated with water use during flowering and podding, respectively, under both the WW and WS treatments. The root dry weight was higher in the 0.2–0.8 m soil layer in the WW treatment than the WS treatment. We conclude that supplementary water at flowering increased the water uptake from deeper soil layers by increasing the distribution of roots in the subsoil layers that resulted in the production of more flowers and filled pods and increased the WUEG and grain yield.
