Background: Shikonin, a natural naphthoquinone, is abundant in Chinese herb medicine Zicao (purple gromwell) and has a wide range of biological activities, especially for cancer. Shikonin and its analogues have been reported to induce cell-cycle arrest, but target information is still unclear. We hypothesized that shikonin, with a structure similar to that of quinone-type compounds, which are inhibitors of cell division cycle 25 (Cdc25) phosphatases, will have similar effects on Cdc25s. To test this hypothesis, the effects of shikonin on Cdc25s and cell-cycle progression were determined in this paper. Methods: The in vitro effects of shikonin and its analogues on Cdc25s were detected by fluorometric assay kit. The binding mode between shikonin and Cdc25B was modelled by molecular docking. The dephosphorylating level of cyclin-dependent kinase 1 (CDK1), a natural substrate of Cdc25B, was tested by Western blotting. The effect of shikonin on cell cycle progression was investigated by flow cytometry analysis. We also tested the anti-proliferation activity of shikonin on cancer cell lines by MTT assay. Moreover, in vivo anti-proliferation activity was tested in a mouse xenograft tumour model. Results: Shikonin and its analogues inhibited recombinant human Cdc25 A, B, and C phosphatase with IC50 values ranging from 2.14 ± 0.21 to 13.45 ± 1.45 μM irreversibly. The molecular modelling results showed that shikonin bound to the inhibitor binding pocket of Cdc25B with a favourable binding mode through hydrophobic interactions and hydrogen bonds. In addition, an accumulation of the tyrosine 15-phosphorylated form of CDK1 was induced by shikonin in a concentration-dependent manner in vitro and in vivo. We also confirmed that shikonin showed an anti-proliferation effect on three cancer cell lines with IC50 values ranging from 6.15 ± 0.46 to 9.56 ± 1.03 μM. Furthermore, shikonin showed a promising anti-proliferation effect on a K562 mouse xenograph tumour model. Conclusion: In this study, we provide evidence for how shikonin induces cell cycle arrest and functions as a Cdc25s inhibitor. It shows an anti-proliferation effect both in vitro and in vivo by mediating Cdc25s.

The potato plant is one of the main crops in the world. However, relatively little is known about key virulence factors of major fungal and bacterial diseases in potatoes, biocontrol measures to improve activity and stability, and the core driving forces in the control process. Here, we focus on analyzing the mechanisms by which genes, proteins, or (and) metabolites of potato pathogens as key virulence factors. Then, the single strain biocontrol agents, synthetic microbial communities, microbial microcapsule strategies were introduced, and the latter two strategies can improve stability and activity in biocontrol. Meanwhile, summarized the defense mechanisms of biocontrol and their specific issues in practical applications. Furthermore, explore how potato crop management, soil management, and climate effects, as crucial driving forces affect potato biocontrol in the system. Dynamic and systematic research, excavation of biocontrol strain resources, find the causes of regional disease resistance and exploration of biocontrol mechanism will provide promising solutions for biotic stress faced by potato in the future.

In order to isolated and identified the bacterial strains from wheat rhizosphere and evaluated the effect of different concentration of bacterial fermentation broth on the wild oats weed growth. This experiment carried out the separation and purification of dominant bacterial strains from the wheat rhizosphere soil, and performed the fermentation broth biological activity assessment by measured the seed germination and plant growth from 20 wheat varieties. The results had shown that the bacterial fermentation broth inhibits the growth of wild oat seedlings and plants to varying degrees, bacterial strains of X3, X4, X8, X12, X16 and X20 has certain level of inhibition activity and X20 has the highest herbicidal effectiveness. According to molecular biology identification, obtained superior bacterial strains X20 was Bacillus as potentially inhibitor for developing of bacterial-based bioherbicides for wild oats weed control management in the wheat field. In this study, an effective analytical method for simultaneous determination of thiamethoxam and its metabolite clothianidin in goji berry and soil was developed by liquid chromatography-tandem mass spectrometry (LC-MS/MS). The recoveries of the compounds in goji berry and soil at the levels of 0.005, 0.02, and 0.1 μg kg−1 were 84.7–98.9% and the relative standard deviations (RSDs) were 0.9–3.2%. The limits of detection (LOD) for both compounds in goji berry and soil matrices were 0.001 mg kg−1; the limits of quantification (LOQ) were 0.005 mg kg−1 for both compounds in two matrices. The dissipation and final residual experiments in 2016 with the commercial formulation of dinotefuran • thiamethoxam 30% suspension concentrate (SC) was conducted in goji berries in northwest China (Qinghai, Gansu, Inner Mongolia, and Ningxia). Thiamethoxam was dissipated fast in goji plant ecosystem with half-lives were 1.08–1.01 and 2.04–4.25 days in goji berry and soil. The final residues of thiamethoxam were <0.005–0.382 and <0.005–1.120 mg kg−1 in goji berry and soil, respectively.

In this study, a reliable and efficient residue analytical method to determine dinotefuran and its metabolites [1-methyl-3-(tetrahydro-3-furylmethyl) guanidine (DN) and 1-methyl-3-(tetrahydro-3-furylmethyl) urea (UF)] in wolfberry and soil was established. The mean recoveries from soil and wolfberry matrices ranged from 84.7 to 98.9%, with intra-day relative standard deviations (RSDr) in the range of 0.9–2.5% for three test compounds. The limits of quantification (LOQs) were 0.005 mg kg−1 for dinotefuran, DN, and UF in wolfberry and soil matrices, and the limits of detection (LODs) were 0.0001 mg kg−1 for three compounds in two matrices. The dissipation and residual assay in 2016 with the commercial formulation of dinotefuran • thiamethoxam 30% suspension concentrate (SC) was conducted at four locations of Qinghai, Gansu, Inner Mongolia, and Ningxia province. The dissipation rates of dinotefuran were rapid in wolfberry with the half-lives were 1.4–3.1 days. The dissipation in soil were relatively slow with half-lives 7.5–17.4 days. According to the results of dietary risk assessment, the residue of dinotefuran at the recommended dosage is safe for human

Effective storage of potatoes is very important for the food industry. Given the problems involving rotten potatoes and low quality during storage, harvested potatoes from the main potato-producing areas in the Qinghai Plateau were treated by selection and air drying (Group “A”) and the others were stored directly as controls (Group “C”). Then, the microbial community structure and diversity of diseased potato tubers from four main production areas were analyzed by high-throughput sequencing technology in different storage stages. The results showed that the community composition and diversity of microbes in different regions and storage periods were different, and the dominant fungi in diseased potato tubers were Boeremia in Huangyuan (HY), Maying (MY) and Zhongling (ZL) and Apiotrichum in Huangzhong (HZ) at the genus level. The dominant bacterial genus was Pseudomonas, but its abundance varied in samples from different regions and storage periods. In the analysis of indicator species, there were some common species and endemic species in each region and period, and the period with the largest number of different species was the third period. Among the four storage periods, the region with the largest number of different species was HZ. Some fungi, especially Fusarium and other potato pathogens, were more abundant in control Group “C” than in treatment Group “A.” In the diversity analysis, the α diversity of fungi in Group “C” was higher than that in Group “A,” but the α diversity of bacteria in Group “A” was higher than that in Group “C,” and there was no obvious regularity with storage time. The β diversity varied significantly among different regions. In addition, through functional prediction analysis, it was found that a plant pathogen was one of the main nutritional types of fungi, which indicated that treatment by selection and drying could significantly reduce phytopathogenic microbe and other microorganisms and could be used as an effective measure for potato storage compared with the prevention and control by drugs that can cause environmental pollution. Further analysis of co-occurrence network showed that pathogenic fungi Fusarium was negatively correlated with pathogenic bacteria Erwinia, and there is also a negative correlation between pathogens and antagonistic microorganisms indicated that there were various symbiotic relationships among microorganisms in diseased potatoes. This study may provide a theoretical basis for biological control of potato cellar diseases and the maintenance of potato quality during long-term storage.
