Introduction: Trichophyton verrucosum belongs to the dermatophyte fungi, closely related organisms that cause skin infections in animals and humans. T. verrucosum infection has been reported in livestock and people in different countries from all continents. Human cases have been reported in different areas of Pakistan, but there is little information about the animal source of the fungus. Methodology: Dermatological specimens collected in the Chitral district of Pakistan for a study on mange in livestock were retrospectively analyzed for the presence of T. verrucosum. In total, 5,873 animals (1,087 cows, 2,033 goats, and 2,753 sheep) were screened for evidence of dermatological lesions during two surveys performed in the summer and winter seasons. Skin scrapings collected from animals with lesions were analyzed by direct microscopic examination after digestion in sodium hydroxide and a real-time polymerase chain reaction (PCR) targeting pathogenic Trichophyton species. Results: At microscopy, samples from 18 cows (1.6%), 3 sheep (0.1%), and 4 goats (0.2%) were positive for fungal elements consistent with T. verrucosum. PCR confirmed the microscopy results. The prevalence was lower than that reported in other countries in intensive breeding farms. Results agree with the literature regarding factors affecting T. verrucosum diffusion, i.e., infection was more prevalent in cattle, especially in younger animals during the winter season. Conclusions: This study reports, for the first time, the presence of T. verrucosum in livestock in Pakistan. A better knowledge of the animal role in the spread of this fungus may allow the adoption of more efficient control measures and prophylaxis.

Phenomic characterization through digital imaging (DI) can capture the three dimensional variation in wheat grain size and shape using different image orientations. Digital imaging may help identifying genomic regions controlling grain morphology using association mapping with simple sequence repeats (SSRs) markers. Accordingly, seed shape phenotypic data of a core collection of 55 wheat genotypes, previously characterized for osmotic and drought tolerance, were produced using computer based Smart grain software. Measured dimensions included seed volume, area, perimeters, length, width, length to width ratio, circularity, horizontal deviation from ellipse (HDEV), vertical deviation from ellipse (VDEV), factor form density (FFD) etc. The thousand grain weight (TGW) was positively correlated with grain size direct measurements; however, VDEV, FFD and other derived grain attributes showed no or negative correlation with TGW. Digital imaging divided the genotypes correctly into well-defined clusters. The wheat genotypes studied were further grouped into two sub-clusters by the Bayesian structure analysis using unlinked SSR markers. A number of loci over various chromosomal regions were found associated to grain morphology by the genome wide analysis using mixed linear model (MLM) approach. A considerable number of marker-trait associations (MTAs) on chromosomes 1D and 2D may carry new alleles with potential to enhance grain weight due to the use of untapped wild accessions of Aegilops tauschii. Conclusively, we demonstrated the application of multiple approaches including high throughput phenotyping using DI complemented with genome wide association studies to identify candidate genomic regions underlying these traits, which allows a better understanding on molecular genetics of wheat grain weight.

Endopolygalacturonases characterized until now have either low working temperatures, working pH in acidic range, high Michaelis–Menten constant (Km), or a high production cost. These characteristics are a hurdle in the industrial applications of these endopolygalacturonases. The purpose of this work was to characterize a novel endopolygalacturonase produced by Bacillus licheniformis IEB-8. Phylogenetic analysis of Bacillus licheniformis IEB-8 showed that the isolate was unique. Citrus peels were used as the only nutrient source for the growth of Bacillus licheniformis IEB-8, allowing a cheap production of endopolygalacturonase. All the synthetic carbon sources showed a negative impact on the production of endopolygalacturonase, while ammonium sulfate enhanced its production. Among different metal ions, Zn+2 showed a negative effect while Mg+2 and Ca+2 did not have any significant effect on the endopolygalacturonase activity. A Lineweaver-Burk plot was prepared for the characterization of the kinetic parameters including Km and Vmax, which were 0.45 mg/mL and 285.7 µM/min, respectively. A comprehensive comparison of the endopolygalacturonase from this study with the available literature indicated that it is better than the reported and commercially available endopolygalacturonases in having the optimum working temperature of 55 °C, a low Km of 0.57 mg/mL, and pH of 7 to 8, which indicated its novelty.

Garlic (Allium sativum) is an important cash food crop, and the biotechnology industry has considerable interest in the plant because of its medicinal importance. These medicinal properties are attributed to organosulphur compounds as the accumulation of these compounds varies according to genotype, locality, light quality, and cultivation practices. In this study, we compared a newly developed garlic variety NARC-G1 by National Agricultural Research Centre (NARC), Islamabad, Pakistan with three different garlic cultivars and highlighted the distinctive attributes like phenotypic characteristics, the content of allicin, elemental profile, and gene polymorphism. Phenotypic analysis showed NARC-G1 has significantly higher bulb weight (66.36g ± 18.58), single clove weight (5.87g ± 1.041), and clove width (17.41mm ± 0.95) which directly correlates to the size of the garlic. The analytical analysis showed the highest allicin content (4.82 ± 0.001) in NARC-G1. Genotyping of the alliinase in all four cultivars showed indels in the gene resulting in distinguishable changes in organosulphur compounds’ profile. NARC-G1 is unique from other garlic cultivars and could be the best choice for mass production with proper cultivation and irrigation management. Moreover, for Pakistan NARC-G1 could be a potential contender to earn the industrial benefits with inland cultivation instead of importing garlic alleviating the economic burden.

Brucellosis is an important zoonotic disease causing significant economic losses worldwide. Early detection of this disease is essential for its control and eradication. Presently, an Indirect Enzyme Linked Immunosorbent Assay (I-ELISA) was developed using lipopolysaccharide (LPS) as antigen and compared with the commercial kit using one hundred negative and positive sera each from buffaloes. The agreement for the positive result between the developed and commercial I-ELISA was 78% and for the negative it was 100%. At 52.49%, 53.09%, 53.26%, 53.86% and 53.94% cut off the sensitivity was 100%, 100%, 97.53%, 88.93% and 86.42%, while the specificity was 84.03%, 84.87%, 85.71%, 87.39% and 87.39%, respectively, for developed I-ELISA. This developed test can be used for the screening of herds as the relative sensitivity is higher.
