Intracerebral hemorrhage (ICH) is a neurological disorder resulting from the nontraumatic rupture of blood vessels in the brain. Ferroptosis is a newly identified form of programmed cell death, which is an important pathological feature of ICH injury. At present, the therapeutic efficacy of ICH treatment is far from satisfactory, so it is urgent to develop a safer and more effective method to treat ICH injury. Resveratrol (Res), a widely used nonflavonoid polyphenol compound, plays a neuroprotective role in many diseases. However, its poor oral bioavailability limits its clinical application in ICH. Polymer nanoparticles (NPs) are a commonly used drug delivery matrix material with good biocompatibility. To improve its bioavailability and accumulation in the brain, we used NPs to encapsulate Res. These spherical Res nanoparticles (Res-NPs) had a particle size of 297.57 ± 7.07 nm, a PDI of 0.23 ± 0.02 and a zeta potential of −5.45 ± 0.27 mV. They could be taken up by Madin–Darby canine kidney (MDCK) cells through a variety of nonspecific endocytosis mechanisms, mainly mediated by clathrin and plasma membrane microcapsules. After entering the cell, Res-NPs tend to accumulate in the endoplasmic reticulum and lysosomes. In a zebrafish model, we observed that Res-NPs could transport across physiological barriers. In a Sprague-Dawley (SD) rat model, we found that Res-NPs had more desirable improvements in Res accumulation within the plasma and brain. Moreover, we demonstrated that Res-NPs were able to inhibit ferroptosis induced by erastin in HT22 mouse hippocampal cells, which are commonly used in in vitro studies to examine neuronal differentiation and neurotoxicity implicated in brain injuries or neurological diseases. Finally, in an ICH mouse model, we confirmed that Res-NPs are a safer and effective treatment for ICH injury. Collectively, Res-NPs are effective to improve Res brain delivery and its therapeutic efficacy in ICH treatment.

Aim: β-asarone, an active component of Acori graminei rhizome, has been reported to have neuroprotective effects in Alzheimer’s disease. As the underlying mechanism is not known, we investigated the neuroprotective effects of β-asarone in an APP/PS1 double transgenic mouse model and in NG108 cells. Materials and methods: APPswe/PS1dE9 double transgenic male mice were randomly assigned to a model group, β-asarone treatment groups (21.2, 42.4, or 84.8 mg/kg/d), or donepezil treatment group (2 mg/kg/d). Donepezil treatment was a positive control, and background- and age-matched wild-type B6 mice were an external control group. β-asarone (95.6% purity) was dissolved in 0.8% Tween 80 and administered by gavage once daily for 2.5 months. Control and model animals received an equal volume of vehicle. After 2.5 months of treatment, behavior of all animals was evaluated in a Morris water maze. Expression of synaptophysin (SYP) and glutamatergic receptor 1 (G1uR1) in the hippocampus and cortex of the double transgenic mice was assayed by Western blotting. The antagonistic effects of β-asarone against amyloid-β peptide (Aβ) were investigated in vitro in the NG108-15 cell line. After 24 hours of incubation, cells were treated with 10 µm Aβ with or without β-asarone at different concentrations (6.25, 12.5, or 25 µM) for an additional 36 hours. The cytotoxicity of β-asarone was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay of cell viability, and cell morphology was evaluated by bright-field microscopy after 24 hours of treatment. The expression of SYP and GluR1 in cells was detected by Western blot assay in the hippocampus and brain cortex tissues of mice. Results: β-asarone at a high dose reduced escape latency and upregulated SYP and GluR1 expression at both medium and high doses. Cell morphology evaluation showed that β-asarone treatment did not result in obvious cell surface spots and cytoplasmic granularity. β-asarone had a dose-dependent effect on cell proliferation. Conclusion: β-asarone antagonized the Aβ neurotoxicity in vivo, improved the learning and memory ability of APP/PS1 mice, and increased the expression of SYP and GluR1 both in vivo and in vitro. Thus, β-asarone may be a potential drug for the treatment of Alzheimer’s disease.

Sodium Tanshinone IIA sulfonate (STS) is a derivative of Tanshinone IIA (Tan IIA). Tan IIA has been reported to possess neuroprotective effects against Alzheimer’s disease (AD). However, whether STS possesses effect on AD remains unclear. This study aims to estimate whether STS could protect against scopolamine- (SCOP-) induced learning and memory deficit in Kunming mice. Morris water maze results showed that oral administration of STS (10 mg/kg and 20 mg/kg) and Donepezil shortened escape latency, increased crossing times of the original position of the platform, and increased the time spent in the target quadrant. STS decreased the activity of acetylcholinesterase (AChE) and increased the activity of choline acetyltransferase (ChAT) in the hippocampus and cortex of SCOP-treated mice. Oxidative stress results showed that STS increased the activity of superoxide dismutase (SOD) and decreased the levels of malondialdehyde (MDA) and reactive oxygen species (ROS) in hippocampus and cortex. In addition, western blot was carried out to detect the expression of apoptosis related proteins (Bcl-2, Bax, and Caspase-3). STS upregulated the protein expression of Bcl-2 and downregulated the proteins expression of Bax and Caspase-3. These results indicated that STS might become a promising therapeutic candidate for attenuating AD-like pathological dysfunction.
