A Gram-stain-negative, light pink-coloured, rod-shaped, flagellated and facultative anaerobic bacterial strain, designated MT2928T, was isolated from deep-sea sediment collected from the Mariana Trench. Growth of strain MT2928T occurred optimally at 28 °C, pH 8.0–9.0 and in the presence of 1.0–2.0 % (w/v) NaCl. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain MT2928T belongs to the genus Pontivivens and has the highest sequence similarity to Pontivivens insulae GYSW-23T (96.6 %). Genomic analysis indicated that strain MT2928T contains a circular chromosome of 4 199 362 bp with G+C content of 67.2 mol%. The strain did not produce bacteriochlorophyll a, but produced carotenoid. The predominant respiratory quinone of MT2928T was ubiquinone-10. The polar lipids of MT2928T contained diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, two unidentified lipids and two unidentified phospholipids. The major fatty acids of strain MT2928T contained summed feature 8 (C18 : 1 ω7c or/and C18 : 1 ω6c), C18 : 0 and summed feature 2 (iso-C16 : 1 I and/or C14 : 0 3-OH). On the basis of phylogenetic, physiological, biochemical and other phenotypic properties, strain MT2928T represents a novel species of the genus Pontivivens, and the name Pontivivens ytuae sp. nov. is proposed with the type species MT2928T (=MCCC 1K05575T=JCM 34320T).
As the largest green macroalgal bloom in the Yellow Sea of China, the overgrowth and degradation of Ulva prolifera (U. prolifera) have a harmful effect on marine organisms and the aquaculture industry. However, the regulation mechanism of U. prolifera stress on the antioxidant capacity and inflammatory response of marine fish is still not completely understood. A 15-day exposure experiment was conducted to evaluate the effects of U. prolifera stress on the antioxidant capacity and inflammatory response of the Japanese flounder (Paralichthys olivaceus) (283.11 ± 6.45 g). The results showed that U. prolifera stress significantly decreased their survival rate. Serum total antioxidant capacity (T-AOC) and non-specific immune-related enzyme activities were significantly impacted under U. prolifera conditions. Moreover, U. prolifera stress significantly decreased T-AOC, superoxide dismutase (SOD) activity, and catalase (CAT) activities in the liver, while malondialdehyde (MDA) contents were significantly increased. Similarly, antioxidant-related gene (cat, nrf2, and keap1) expressions were synchronously downregulated in the liver under U. prolifera stress. Furthermore, U. prolifera stress significantly upregulated pro-inflammatory gene (tnf-α, il-1β, ifn-γ, and p65) expressions and the phosphorylation levels of the p38 and JNK MAPK pathways in the head kidney. In addition, endoplasmic reticulum (ER) stress-related gene and protein expressions were also upregulated in the head kidney. Overall, these results revealed that U. prolifera stress suppressed the antioxidant capacity and induced an inflammatory response in the Japanese flounder. This study could advance the understanding of the adverse effects of U. prolifera stress on marine benthic fish and promote the sustainable development of aquaculture.
The Japanese sardine (Sardinops sagax), a key economic species in the Northwest Pacific Ocean (NWPO), has shown significant increases in both population abundance and catch volume over the past decade. To understand its spatiotemporal dynamics under climate change, this study analyzed light purse seine fishery data (2014–2021) from the NWPO. The results showed that the primary fishing season spans March to December, with peak catches concentrated in 40–43° N, 149–155° E. Annual catches grew steadily, accelerating notably in 2021. The fishing grounds’ center shifted northeastward annually and seasonally (southwest-to-northeast trajectory), driven by directional aggregation. Spatial clustering with global positive autocorrelation was observed, weakening as distance thresholds increased. Resource hotspots migrated northeast, narrowing from 40–42° N (2016) to 42–44° N (2017–2021), while coldspots showed complementary fluctuations. Generalized additive model (GAM) analysis revealed that the catch per unit effort (CPUE) of Japanese sardine in the high seas of the NWPO was governed by temporal–spatial drivers and multivariate environmental determinants. Analytical findings substantiate the significant climate-driven impacts on the spatiotemporal distribution and population dynamics of Japanese sardine. The non-stationary interannual and seasonal patterns of fishing grounds highlight the need for adaptive management strategies.
Rhincodon typus is a keystone and indicator species in marine ecosystems. Meanwhile, R. typus has been listed on the IUCN red list of vulnerable species. Here we used ONT platform to determine the full-length (FL) transcriptome of R. typus and obtained 14,930 FL transcripts. Among all FL transcripts, 14,915 transcripts were covered 11,892 genetic loci and 1,642 novel genetic loci were further found. Meanwhile, we identified 714 novel transcripts by compared FL transcripts with the R. typus genome. Based on FL transcripts, we also predicted the distribution patterns of ASs, LncRNAs, polyAs, CDSs and methylation sites on FL transcriptome of R. typus. Furthermore, a total of 31,021 (97.86%) CDSs can obtained annotation information. Overall, our work firstly provided the FL transcriptome and these sequences complete the annotated R. typus genome information. Furthermore, these information are a potential resource to study biological processes of R. typus.
