Zearalenone (ZEA) is a nonsteroidal estrogenic mycotoxin found in several food commodities worldwide. ZEA causes reproductive disorders, genotoxicity, and testicular toxicity in animals. However, little is known about the functions of apoptosis and autophagy after exposure to ZEA in granulosa cells. This study investigated the effects of ZEA on chicken granulosa cells. The results show that ZEA at different doses significantly inhibited the growth of chicken granulosa cells by inducing apoptosis. ZEA treatment up-regulated Bax and downregulated Bcl-2 expression, promoted cytochrome c release into the cytosol, and triggered mitochondria-mediated apoptosis. Consequently, caspase-9 and downstream effector caspase-3 were activated, resulting in chicken granulosa cells apoptosis. ZEA treatment also upregulated LC3-II and Beclin-1 expression, suggesting that ZEA induced a high level of autophagy. Pretreatment with chloroquine (an autophagy inhibitor) and rapamycin (an autophagy inducer) increased and decreased the rate of apoptosis, respectively, in contrast with other ZEA-treated groups. Autophagy delayed apoptosis in the ZEA-treated cells. Therefore, autophagy may prevent cells from undergoing apoptosis by reducing ZEA-induced cytotoxicity. In addition, our results further show that the autophagy was stimulated by ZEA through PI3K-AKT-mTOR and MAPK signaling pathways in chicken granulosa cells.

Skeletal muscle plays important roles in animal locomotion, metabolism, and meat production in farm animals. Current studies showed that non-coding RNAs, especially the circular RNA (circRNA) play an indispensable role in skeletal muscle development. Our previous study revealed that several differentially expressed circRNAs among fast muscle growing broilers (FMGB) and slow muscle growing layers (SMGL) may regulate muscle development in the chicken. In this study, a novel differentially expressed circPPP1R13B was identified. Molecular mechanism analysis indicated that circPPP1R13B targets miR-9-5p and negatively regulates the expression of miR-9-5p, which was previously reported to be an inhibitor of skeletal muscle development. In addition, circPPP1R13B positively regulated the expression of miR-9-5p target gene insulin like growth factor 2 mRNA binding protein 3 (IGF2BP3) and further activated the downstream insulin like growth factors (IGF)/phosphatidylinositol 3-kinase (PI3K)/AKT serine/threonine kinase (AKT) signaling pathway. The results also showed that the knockdown of circPPP1R13B inhibits chicken skeletal muscle satellite cells (SMSCs) proliferation and differentiation, and the overexpression of circPPP1R13B promotes the proliferation and differentiation of chicken SMSCs. Furthermore, the overexpression of circPPP1R13B could block the inhibitory effect of miR-9-5p on chicken SMSC proliferation and differentiation. In summary, our results suggested that circPPP1R13B promotes chicken SMSC proliferation and differentiation by targeting miR-9-5p and activating IGF/PI3K/AKT signaling pathway.

Skeletal muscle satellite cell growth and development is a complicated process driven by multiple genes. The PDZ and LIM domain 5 (PDLIM5) gene has been proven to function in C2C12 myoblast differentiation and is involved in the regulation of skeletal muscle development. The role of PDLIM5 in chicken skeletal muscle satellite cells, however, is unclear. In this study, in order to determine whether the PDLIM5 gene has a function in chicken skeletal muscle satellite cells, we examined the changes in proliferation and differentiation of chicken skeletal muscle satellite cells (SMSCs) after interfering and overexpressing PDLIM5 in cells. In addition, the molecular pathways of the PDLIM5 gene regulating SMSC proliferation and differentiation were analyzed by transcriptome sequencing. Our results show that PDLIM5 can promote the proliferation and differentiation of SMSCs; furthermore, through transcriptome sequencing, it can be found that the differential genes are enriched in the MAPK signaling pathway after knocking down PDLIM5. Finally, it was verified that PDLIM5 played an active role in the proliferation and differentiation of chicken SMSCs by activating the p38-MAPK signaling pathway. These results indicate that PDLIM5 may be involved in the growth and development of chicken skeletal muscle.

Spring wheat–summer maize (SWSM) annual crop systems were formed to satisfy the maize grain mechanized harvest thermal requirement in the thermal–resource–limited region of the North China Plain. However, the nitrogen (N) application rate effect on SWSM annual yield formation, N accumulation and utilization were barely evaluated. Two–year field experiments were conducted to evaluate the effects of the N application rate on the annual yield of SWSM, observe N accumulation and utilization, and identify the optimized N application. The experiments were conducted under 5 N levels of 0 (N0), 180 (N180), 240 (N240), 300 (N300), and 360 (N360) kg ha−1. The results showed that spring wheat, summer maize and annual cereal yield under the N240 and N480 treatments obtained the highest grain yield (GY) of 5038, 1282 and 16,320 kg ha−1, respectively, and the optimal N application rate was estimated using a linear–plateau model to be 231–307, 222–337 and 463–571 kg ha−1 with maximum GY of 4654–5317, 11,727–12,003 and 16,349–16,658 kg ha−1, respectively. With the increase in the N application rate, the dry matter accumulation (DM) were significantly increased by 16.9–173.5% for spring wheat and 11.1≈–76.8% for summer maize, respectively; and the annual cereal DM was 15.1–179.7% greater than that with N0 treatment, respectively. Spring wheat, summer maize and the annual cereal total N accumulation (TN) under N360 and N720 treatments were significantly increased by 5.4–19.1%, 16.6–32.3% and 11.5–26.2%, respectively, compared to the other treatments; however, N use efficiency for biomass and grain production (NUEbms and NUEg) were decreased significantly by 10.9–13.6% and 8.9–20.7%, 6.8–13.8% and 12.2–15.6%, and 5.5–11.7% and 10.0–16.0%, respectively. Meanwhile, the N partial factor productivity (PFPN), N agronomy use efficiency (ANUE), N recovery efficiency (NRE) and N uptake efficiency (NEupk) under the N240 treatment for spring wheat and summer maize obtained high levels of 20.99 and 47.01 kg−1, 9.27 and 16.35 kg−1, 32.53% and 32.44%, and 0.85 and 0.72 kg−1, respectively. Correlation analysis showed that the N application rate, TN and NEupk played significantly positive roles on GY, spring wheat spilke grain number, summer maize ear grain number and 1000–grain weight, DM LAImax and SPADmax, while NUEbms, NUEg, PFPN and ANUE always played negative effects. These results demonstrate that spring wheat, summer maize and annual cereal obtained the highest GY being 4654–5317, 11,727–12,003 and 16,349–16,658 kg ha−1 with the optimal N application rate 231–307, 222–337 and 463–571 kg ha−1, respectively, which provide N application guidance to farmer for spring wheat–summer maize crop systems to achieve annual mechanical harvesting in the thermal–resource–limited region of the North China Plain.

Soybean is an important source of high-quality vegetable protein with various health-improving properties, and its main bioactive substances are small peptides produced by in vitro enzymatic hydrolytic processes. In traditional layer breeding, the nutritional health of roosters is frequently neglected, ultimately affecting the quality and quantity of offspring. This study investigated the effects of various quantities (0%, 0.15%, 0.30%, 0.45%, and 0.60%) of soybean bioactive peptide (SBP) feed additives on immunological and antioxidant functions, gut health, and reproductive performance of roosters. SBP supplementation significantly improved male growth and reproductive performance, including growth rate, feed conversion ratio, reproductive organ development, and semen quality. SBP also increased immune and antioxidant levels, boosted the integrity of the small intestinal physiological structure and barrier function, and diversity of cecal microbes, and decreased the apoptotic ratio of small intestinal epithelial cells. The effects of SBP on various functions of males showed a quadratic trend, with the optimal concentration determined to be 0.45%.

Natural mating colony cages are crucial in poultry breeding, yet breed-specific management requires further investigation. We evaluated the effects of sex ratios, stocking densities, and cohabitation age on Lohmann Pink-shell breeders’ performance. A total of 6126 birds were randomly allocated to experimental groups with varying ratios (1:8–1:13), densities (582–748 cm2/bird), and cohabitation ages (120/140 days), each containing six replicates. We monitored male mating frequencies at 50 weeks in 1:8 and 1:10 ratio groups. All 120-day-old groups showed delayed production onset and superior male weight compliance (p < 0.01), with reduced egg breakage and increased healthy chick output (p < 0.01). Lower stocking densities (748/694 cm2/bird) showed lower breakage rate and uniformity than 582 cm2/bird (p < 0.05). The 1:10 sex ratio achieved optimal egg production and fertilization rate (p < 0.05). Male mating peaked between 16:00 and 18:00. Optimal parameters were 120-day age of cohabitation, 694 cm2/bird density, and 1:10 sex ratio, providing theoretical guidance for natural mating colony cage development in layer breeding.
