Abscisic acid (ABA) regulates plant development, seed germination, and stress responses. The PYR1-like (PYL) proteins are essential for ABA signalling. However, the evolution and expression of PYL genes in potato (Solanum tuberosum) remain poorly understood. Here, we analysed and identified 17 PYL genes in the potato genome, which were categorised into three groups based on phylogenetic analysis. These genes are distributed across nine chromosomes with predicted proteins subcellar localisation primarily in the cytoplasm. These StPYLs revealed conserved exon structures and domains among the groups. Promoter region analysis indicated hormone and stress-related elements in all StPYLs. Protein–protein interactions and microRNA networks predicted that the interactions of StPYLs are crucial components of ABA signalling, underlining their pivotal role in stress management and growth regulation in potato. Expression profiling across different tissues and under various stresses revealed their varied expression pattern. Further, we validated the expression pattern of selected StPYLs through reverse transcription quantitative PCR under drought, salt, and Phytophthora infestans stresses. This revealed consistent upregulation of StPYL6 in these stresses, while StPYL11 exhibited significant downregulation over time. Other genes showed downregulation under drought and salt stresses while upregulation under P. infestans. Overall, our results suggested the potential role of PYL genes in abiotic and biotic stresses.

Carbon dots (CDs) are a class of carbon-based nanomaterials undergoing rapid development with broad potential applications across diverse biomedical fields. These materials are highly attractive for diagnostics, therapeutics, and nanomedicine due to their remarkable optical and physicochemical properties, including photoluminescence, biocompatibility, and aqueous dispersibility. CDs can be synthesized using various techniques, ranging from top-down to bottom-up approaches. Among these, biogenic synthesis, utilizing natural sources and waste materials, presents an eco-friendly and sustainable alternative. CDs have exhibited considerable promise in diagnostics, especially with bioimaging and biosensing, providing both high sensitivity and precise identification. CDs are presently being investigated in the pharmaceutical sector for their potential applications in cancer and infection treatment, as well as in photodynamic and thermal therapies. The advancement of CD composites, through enhanced functionality and broader application, facilitates novel research in nanomedicine. This article highlights the advantages of CDs, focusing on their structural properties, classification, and versatility in synthesis methods. Furthermore, the safety and toxicity profiles of CDs are critically analyzed. In conclusion, the innocuity, adaptability, and multifunctionality of CDs position them as a cornerstone in the advancement of nanotechnology and biomedical applications. With their broad applicability and promising potential, CDs stand poised to drive significant innovation across diagnostics, therapeutics, and other domains, heralding a new era in nanomedicine and sustainable material development.

(1) Background: Gastric cancer (GC) is the fourth leading cause of cancer death worldwide. Silver nanoparticles (Ag-NPs) have been increasingly used in the diagnosis and treatment of cancer due to their physicochemical properties. This study investigated the role of a kind of biosynthetic silver nanoparticle (b-Ag) in the development of GC, the enhancement of 5-fluorouracil (5F), and its mechanism. (2) Methods: X-ray, transmission electron microscopy (TEM), and UV absorbance were used to detect the characterizations of AgNPs. CCK8, Colony formation and a Transwell assay were performed to confirm the malignant behaviors of GC. DCFH-DA and DHE were used to detect intracellular reactive oxygen species (ROS). Quantitative reverse transcription polymerase chain reaction (qRT-PCR) was used to detect the mRNA expression of apoptosis-related genes. (3) Results: Compared with the chemosynthetic silver nanoparticles (c-Ag), b-Ag had a stronger cytokilling effect, and it had a better inhibition on the malignant phenotype of GC when combined with 5F. The b-Ag increased the expression of Bax and P53 while decreasing the expression of Bcl2. It also promoted the generation of intracellular ROS. (4) Conclusions: By promoting cell apoptosis and increasing intracellular ROS, b-Ag inhibited the development of GC and enhanced the inhibition of 5F on GC.
