The wilting process is required for the preparation of traditional low-moisture silage, which is not only subject to the interference of rainfall during the harvest season in many areas, but also increases labor, economic, and time costs. Therefore, in this experiment, newly harvested alfalfa was supplemented with a complex additive consisting of lactic acid bacteria (LAB), sucrose, and corn flour, and then ensiled directly in order to explore a suitable high-moisture silage processing method. There were four groups: control (CK), 0.01 g/kg LAB plus 2% sugar (S), 0.01 g/kg LAB plus 5% corn flour (C5), and 0.01 g/kg LAB plus 5% corn flour (C10). The results show that all four types of silage had significantly reduced acid detergent fiber, acetic acid content, and ammonia/total nitrogen (p < 0.05). The relative abundance of Lactobacillus spp. was significantly higher (p < 0.05), and that of Weissella, Clostridium sensu stricto 12, and Pantoea genera was significantly lower (p < 0.05) in all four treatments. Group S had significantly increased crude protein content, and groups C5 and C10 had significantly reduced neutral detergent fiber. Overall, group C10 had the highest LA concentration, the highest relative abundance of Lactobacillus spp., and the lowest pH, ammonia/total nitrogen, neutral detergent fiber, and acid detergent fiber. Silage with a complex additive consisting of LAB + 10% corn flour is recommended to achieve good fermentation with high nutritional quality.

Cysteamine (CS), as a feed supplement, can increase the level of growth hormone (GH) in the blood, promote animal growth. However, little attention has been paid to the effects of CS on the rumen microbiome and metabolic profile in cashmere goats. This study aimed to assess the effects of rumen microbiota, metabolites, and plasma antioxidative capacity induced by CS supplementation in cashmere goats. We selected 30 Inner Mongolia white cashmere goat ewes (aged 18 months), and randomly separate the goats into three groups (n = 10 per group) to experiment for 40 days. Oral 0 (control group, CON), 60 (low CS, LCS), or 120 mg/kg BW−1 (high CS, HCS) coated CS hydrochloride every day. Using 16S and internal transcribed spacer (ITS) rRNA gene amplicon sequencing, we identified 12 bacterial and 3 fungal genera with significant changes among the groups, respectively. We found a significant increase in rumen NH3-N and total volatile fatty acid (TVFA) concentrations in the LCS and HCS groups compared with the CON. With untargeted LC–MS/MS metabolomics, we screened 59 rumen differential metabolites. Among the screened metabolites, many unsaturated and saturated fatty acids increased and decreased with CS treatment, respectively. CS supplementation increased the levels of plasma total antioxidant capacity (T-AOC), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), GH, and insulin-like growth factor-1(IGF-1). Spearman correlation analysis revealed that the abundance of U29-B03, Lactococcus, and Brochothrix were positively associated with the levels of δ2-THA, TVFA and antioxidant capacity. In conclusion, CS significantly affected rumen microbiota and fermentation parameters, and ultimately inhibited the biohydrogenation of rumen metabolites, enhanced plasma antioxidant capacity, and regulated some hormones of the GH–IGF-1 axis. This study provides an overall view into the CS application as a strategy to improve health production in cashmere goats.

To explore the effects of different silage inoculants on the silage quality of alfalfa (Medicago sativa L.), the experiment used Alfalfa with a moisture content of 60.00% after harvesting as the raw material. The treatments included a control group containing only distilled water (CK), Xinlaiwang I-straw silage agent (A), Xinlaiwang I-alfalfa silage agent (B), Zhuanglemei silage fermentation agent (C), Baoshiqing (D), and Kangfuqing S lactic acid bacteria inoculant (E), totaling six treatments. After 60 days of normal temperature sealing treatment, the silage fermentation and nutritional indicators of Alfalfa were measured, and the silage fermentation effect was analyzed by the membership function method. The experiment showed that when the moisture content of alfalfa was 60.00%, the neutral detergent fiber (NDF) and acid detergent fiber (ADF) contents of the silage agent treatment groups were significantly lower than those of the CK group (p < 0.05). The lactic acid (LA) content was significantly higher in the treatment group than in the CK group (p < 0.05). The addition of Xinlaiwang I-alfalfa silage agent in group B significantly increased the crude protein (CP) and LA levels in the Alfalfa silage (p < 0.05). It also reduced the neutral detergent fiber (NDF) and acid detergent fiber (ADF) contents. Additionally, the pH and Ammonia Nitrogen/Total Nitrogen (AN/TN) ratio were lowered. Propionic acid (PA) and butyric acid (BA) were not detected. After the membership function calculation, the average membership value of Xinlaiwang I-alfalfa silage agent (B) group was the highest with a score of 0.90, ranking first, and the silage quality was the best. In summary, through quality analysis and membership function calculation, Xinlaiwang I-alfalfa silage agent can effectively improve the silage quality of WL298HQ alfalfa.

Background/Objectives: Research on cashmere goat coat color is crucial for optimizing cashmere goat breeds and increasing their economic value. To identify key genes associated with the formation of cashmere goat coat color and to provide molecular markers for breeding purposes, three healthy, 3-year-old does with similar weights and distinct coat colors—white, black, and light brown—were selected. Methods: Skin samples were collected for transcriptome sequencing, and bioinformatics methods were applied to screen for differentially expressed genes (DEGs) in the skin of cashmere goats with varying coat colors. Real-time fluorescence quantitative PCR (qRT-PCR) and immunofluorescence were subsequently conducted to examine the expression patterns of these DEGs. Results: The results showed that a total of 1153 DEGs were identified across the three groups of cashmere goats. According to GO and KEGG analyses, these DEGs were involved in key biological processes and structures, such as the melanin biosynthetic process (GO:0042438), melanosome membrane (GO:0033162), and melanin biosynthesis from tyrosine (GO:0006583). Employing Cytoscape, a gene interaction network was plotted, highlighting a compact network of DEGs associated with coat color formation. Critical genes identified included TYRP1, TYR, DCT, ASIP, PMEL, LOC102180584, MLANA, TSPAN10, TRPM1, CLDN16, AHCY, LOC106503350, and LOC102175263. qRT-PCR and fluorescence immunohistochemistry further determined that TYRP1, TYR, DCT, and PMEL expression levels were high in black goats (BGs), while ASIP and AHCY expression levels were high in white goats (WGs). The expression levels of these six genes in light brown goats (RGs) were intermediate between those in BGs and WGs. Conclusions: TYRP1, TYR, DCT, and PMEL were believed to play pivotal roles in the formation of black coat color, while ASIP and AHCY regulated the formation of white coat color in cashmere goats.

With the rapid development of the livestock industry, finding new sources of feed has become a critical issue that needs to be addressed urgently. China is one of the top five sunflower producers in the world and generates a massive amount of sunflower stalks annually, yet this resource has not been effectively utilized. Therefore, in order to tap into the potential of sunflower stalks for animal feed, it is essential to explore and develop efficient methods for their utilization. In this study, various proportions of alfalfa and sunflower straw were co-ensiled with the following mixing ratios: 0:10, 2:8, 4:6, 5:5, 6:4, and 8:2, denoted as A0S10, A2S8, A4S6, A5S5, A6S4, and A8S2, respectively. The nutrient composition, fermentation quality, microbial quantity, microbial diversity, and broad-spectrum metabolomics on the 60th day were assessed. The results showed that the treatment groups with more sunflower straw added (A2S8, A4S6) could start fermentation earlier. On the first day of fermentation, Weissella spp.dominated overwhelmingly in these two groups. At the same time, in the early stage of fermentation, the pH in these two groups dropped rapidly, which could effectively reduce the loss of nutrients in the early stage of fermentation. In the later fermentation period, a declining trend in acetic acid levels was observed in A0S10, A2S8, and A4S6, while no butyric acid production was detected in A0S10 and A2S8 throughout the process. In A4S6, butyric acid production was observed only after 30 days of fermentation. From the perspective of metabolites, compared with sunflower ensiling alone, many bioactive substances such as flavonoids, alkaloids, and terpenes are upregulated in mixed ensiling.
