The aim of this paper was to prepare, by the freeze-drying method, ionically crosslinked chitosan membranes with different contents of pentasodium tripolyphosphate (TPP) and loaded with 1,4-naphthoquinone (NQ14) drug, in order to evaluate how the physical crosslinking affects NQ14 release from chitosan membranes for cancer therapy application. The membranes were characterized by Fourier transform infrared spectroscopy (FTIR), wide-angle X-ray diffraction (WAXD), scanning electron microscopy (SEM), swelling degree, and through in vitro drug release and cytotoxicity studies. According to the results, the molecular structure, porosity and hydrophilicity of the chitosan membranes were affected by TPP concentration and, consequently, the NQ14 drug release behavior from the membranes was also affected. The release of NQ14 from crosslinked chitosan membranes decreased when the cross-linker TPP quantity increased. Thus, depending on the TPP amount, the crosslinked chitosan membranes would be a potential delivery system to control the release of NQ14 for cancer therapy application. Lastly, the inhibitory potential of chitosan membranes ionically crosslinked with TPP and loaded with NQ14 against the B16F10 melanoma cell line was confirmed through in vitro cytotoxicity studies assessed via MTT assay. The anti-proliferative effect of prepared membranes was directly related to the amount of cross-linker and among all membranes prepared, such that one crosslinked with 0.3% of TPP may become a potential delivery system for releasing NQ14 drug for cancer therapy.

Biomaterials stimulate diverse biological responses, including inflammation, wound healing, foreign body reactions, and fibrous encapsulation, all critical for evaluating biocompatibility and effectiveness. These responses are influenced by the material's physicochemical and biological properties, such as composition, texture, and surface characteristics. Adverse reactions, such as severe inflammation or fibrous encapsulation, can hinder tissue integration, jeopardizing patient health and increasing healthcare costs. This review aimed to summarize the current scientific evidence on biological responses to biomaterials. A systematic search was conducted through multiple databases (VHL, PubMed, SCOPUS, EMBASE, and Web of Science) including in vitro and in vivo studies that compared biomaterial interactions with the natural immune response (innate and adaptive). From the 791 articles identified, 25 met strict inclusion criteria. These studies revealed variations in immune responses and material surface characteristics, highlighting advancements made to enhance tissue integration. Bioactive materials demonstrated greater potential for tissue regeneration, while inert materials triggered moderate inflammatory reactions. This variability emphasizes the need for a personalized biomaterial selection, considering both short-term biocompatibility and long-term tissue functionality. This review underscores the importance of comprehensive evaluation to optimize biomaterial performance in clinical applications.

This work aimed to develop chitosan/gelatin scaffolds loaded with ethanolic extract of Jatropha mollissima (EEJM) to evaluate the influence of its content on the properties of these structures. The scaffolds were prepared by freeze-drying, with different EEJM contents (0–10% (w/w)) and crosslinked with genipin (0.5% (w/w)). The EEJM were characterized through High Performance Liquid Chromatography coupled to a Diode Array Detector (HPLC-DAD), and the determination of three secondary metabolites contents was accomplished. The physical, chemical and biological properties of the scaffolds were investigated. From the HPLC-DAD, six main substances were evidenced, and from the quantification of the total concentration, the condensed tannins were the highest (431.68 ± 33.43 mg·g−1). Spectroscopy showed good mixing between the scaffolds’ components. Adding and increasing the EEJM content did not significantly influence the properties of swelling and porosity, but did affect the biodegradation and average pore size. The enzymatic biodegradation test showed a maximum weight loss of 42.89 within 28 days and reinforced the efficiency of genipin in crosslinking chitosan-based materials. The addition of the extract promoted the average pore sizes at a range of 138.44–227.67 µm, which is compatible with those reported for skin regeneration. All of the scaffolds proved to be biocompatible for L929 cells, supporting their potential application as skin tissue engineering materials.

The objective of this study was to develop a chitosan-based biomaterial with calcium hydroxide and 2% chlorhexidine for intracanal treatment application and, consequently, to diminish the number of microorganisms in the root canal system. The chitosan solution was prepared by dissolving it in 2% and 4% acetic acid (v/v) for 1 h at room temperature (25 °C) with magnetic agitation (430 rpm). Calcium hydroxide was obtained in two stages: the first was the synthesis of the calcium oxide—CaO, and the second was that of the calcium hydroxide—Ca(OH)2. The samples were developed using different concentrations of chitosan, calcium hydroxide, and chlorhexidine 2%. They were codified as Ca(OH)2 + Q2% (M1), Ca(OH)2 + Q4% (M2), Ca(OH)2 + Q2% + CLX (M3), Ca(OH)2 + Q4% + CLX (M4), Ca(OH)2 + Q2% + PEG (M5), and Ca(OH)2 + Q4% + PEG (M6). They were characterized through Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and rheological measurement, and the antimicrobial activity was evaluated in vitro. Characteristic absorption bands of the source materials used in this research were observed in the FTIR spectra. The X-ray diffraction technique indicated that the material has a semi-crystalline structure and that the presence of calcium hydroxide made the biomaterial more crystalline. The viscosity measurement showed a pseudoplastic behavior of the studied samples. The microbiologic analysis was positive for all samples tested, with bigger inhibition zones for the samples M3 and M4. As a result, we conclude that the formulation developed based on chitosan is promising and has potential to be an intracanal medication.

In this study, blends based on a high density polyethylene (HDPE) and chitosan (CS) were successfully prepared by melt processing, in a laboratory internal mixer. The CS biopolymer content effect (up to maximum of 40%), and, the addition of bentonite clay modified with quaternary ammonium salt (CTAB) impregnated chitosan as a compatibilizing agent, on the properties of the blends was analyzed by Fourier transform-infrared spectroscopy (FT-IR), wide angle X-ray diffraction (WAXD), differential scanning calorimetry (DSC), thermogravimetric analyses (TG), tensile strength, and scanning electron microscopy (SEM). The use of clay modified with CTAB impregnated chitosan, employing a method developed here, improved the compatibility of HDPE with chitosan, and therefore the thermal and some of the mechanical properties were enhanced, making HDPE/chitosan blends suitable candidates for food packaging. It was possible to obtain products of synthetic polymer, HDPE, with natural polymer, chitosan, using a method very used industrially, with acceptable and more friendly properties to the environment, when compared to conventional synthetic polymers. In addition, due to the possibility of impregnated chitosan with quaternary ammonium salt exhibit higher antibacterial activity than neat chitosan, the HDPE/chitosan/organobentonite blends may be potentially applied in food containers to favor the preservation of food for a longer time in comparison to conventional materials.

Chitosan/montmorillonite nanocomposite films were prepared by the solvent evaporation method to immobilize the drug ibuprofen (IBU) and delay its release in a medium that simulates the environment of the gastrointestinal tract. The effects of montmorillonite, at different mass proportions (10, 20, and 50%), on the morphological and physical properties of the films were studied. The samples were characterized by X-ray diffraction (XRD), Infrared Spectroscopy (FTIR), Scanning Electron Microscopy (SEM), degree of swelling, drug encapsulation, and drug release efficiency. According to the XRD it was evidenced that the incorporation of montmorillonite to chitosan led to the formation of nanocomposites of ordered morphology. The infrared spectra confirmed the good interaction between montmorillonite and chitosan by the formation of nanocomposites. This fact, which favored the imprisonment of the IBU, reduced the diffusion coefficient in the studied systems. The micrographs comproved the formation of dense and uniform films. The controlled release profile, especially for the nanocomposite with 10% clay mass, showed a slow drug release rate. The incorporation of montmorillonite at different proportions produced different morphologies, with good encapsulation efficiency and an adequate profile for the controlled release of the drug.
