This study investigated the effects of astaxanthin (Ax) on the growth, body color, and immunity of Macrobrachium rosenbergii . Four different concentrations of Ax (0, 50, 100, and 200 mg kg −1 ) were supplemented in the basal diet to assess their impact on the growth, coloration, immunity, and antioxidant capacity in M. rosenbergii . After 10 and 20 weeks of feeding, compared to the control group, the addition of 100 mg kg⁻ 1 of Ax significantly increased the weight gain rate (WGR) and specific growth rate (SGR; p < 0.05) and shortened the molting cycle ( p < 0.05). Addition of Ax increased the redness ( value) and yellowness ( value) of live and cooked M. rosenbergii ( p < 0.05), and the muscle Ax content increased as the amount of added Ax increased. The enzyme activity results showed that after 20 weeks of feeding, the inclusion of 200 mg kg −1 of Ax significantly increased the activities of superoxide dismutase (SOD) and alkaline phosphatase (AKP) in M. rosenbergii ( p < 0.05) and significantly decreased the activity of malondialdehyde (MDA; p < 0.05). Compared with control group, the addition of 100 mg kg −1 of Ax significantly increased the expression levels of genes related to immunity and antioxidant activity ( p < 0.05). Additionally, there were significant differences in the expression levels of molting‐related genes (ecdysteroid receptor ( EcR ) and retinoid X receptor ( RXR )), across the different Ax supplementation levels and molting stages. Considering all factors, the addition of 100 mg kg −1 of Ax to the feed not only improved the growth performance and body color of M. rosenbergii but also enhanced its immune and antioxidant capabilities. This study provides a reference for the application of Ax in crustacean feed.

The Asian giant soft-shelled turtle, Pelochelys cantorii (Trionychidae), is one of the largest aquatic turtles in China and was designated as a First-Grade Protected Animal in China in 1989. Previous investigation based on a combination of Illumina short-read, PacBio long-read and Hi-C scaffolding technologies acquired a high-quality chromosome-level genome of Pc. cantorii. In this study, comparative genomic analysis between Pc. cantorii and 16 other vertebrate genomes indicated that turtles separated from the ancestor of archosaurians approximately 256.6 (95% highest posterior density interval, 263.6–251.9) million years ago (Mya) (Upper Permian to Triassic) and that Pc. cantorii separated from the ancestor of Pd. sinensis and R. swinhoei approximately 59.3 (95% highest posterior density interval, 64.3–54.3) Mya. Moreover, several candidate genes, such as VWA5A, ABCG2, A2M and IGSF1, associated with tumor suppression, growth and age were expanded, implicating their potential roles in the exceptional longevity of turtles. This new chromosome-level assembly has important scientific value in the study of conservation of Pc. cantorii and also enriches the evolutionary investigation of turtle species.

The spectrum is a key environmental factor, and light‐emitting diodes (LEDs) can influence the growth and development of crustaceans by altering the composition of the spectrum. This study conducted a 30‐day experiment to investigate the effects of five LED spectra (red, yellow, blue, green, and white light) on the growth, antioxidant and immune enzyme activities, stress hormone levels, and the expression of α‐amylase (α-AMY), ecdysone receptor (EcR) and retinoid X receptor (RXR) genes in juvenile redclaw crayfish (Cherax quadricarinatus). The results show that the survival rate of juveniles is markedly higher in the yellow and red‐light groups than in the other three groups (P < 0.05). The green light group exhibits the lowest survival rate, yet it demonstrates the highest weight gain rate and specific growth rate. Regarding enzyme activity and hormone levels, the yellow light group shows the lowest malondialdehyde content, with higher superoxide dismutase and acid phosphatase activity than the other groups; no significant differences are observed in lysozyme activity among the groups (P > 0.05). The melatonin content in the green and blue light groups is significantly higher than that in the other three groups (P < 0.01). In terms of growth gene expression, the expression of α-AMY, EcR, and RXR in juvenile C. quadricarinatus is regulated by the spectrum. In conclusion, when raised under the yellow light spectrum, juvenile C. quadricarinatus displays elevated survival rates, rapid growth, and robust antioxidant and immune defenses. This study provides important technical parameters for optimizing and enhancing the industrial cultivation of juvenile C. quadricarinatus.

17α-methyltestosterone (MT) is known for its ability to suppress ovaries and induce spermatogenesis; yet, its effects in crustaceans are underexplored. This study investigates the impact of varying MT concentrations on the gonadal development and intestinal microbiota of juvenile Macrobrachium rosenbergii. Feeds containing different MT doses were provided, and the sex ratios, histological observations, reproductive gene expression, and intestinal microbial composition were analyzed. The results revealed short-term feeding (60 days) of 1000 mg/kg MT resulted in the highest male ratio, while long-term feeding (150 days) of 500 mg/kg MT achieved the same outcome. Conversely, long-term feeding of 1500 mg/kg MT led to the lowest male ratio and retarded male germ cell development. An intestinal microbiota analysis showed that MT supplementation significantly increased microbial abundance and altered the intestinal microbial community structure. Additionally, MT suppressed the expression of female reproductive-related genes. This study provides insights into the effects of MT on reproductive development and gut microbiota in juvenile prawns, offering a valuable reference for the application of MT in crustacean aquaculture.

Hibernation in turtle species is an adaptive survival strategy to colder winter conditions or food restrictions. However, the mechanisms underlying seasonal adaptions remain unclear. In the present study, we collected hemocytes from Pelochelys cantorii and compared the molecular signature of these cells between the active state and hibernation period based on single-cell RNA sequencing (scRNA-seq) analysis. We found six cell types and identified a list of new marker genes for each cell subpopulation. Moreover, several heat shock genes, including the Hsp40 family chaperone gene (DNAJ) and HSP temperature-responsive genes (HSPs), were upregulated during the hibernation period, which predicted these genes may play crucial roles in the stress response during hibernation. Additionally, compared to hemocytes in the active state, several upregulated differentially expressed immune-related genes, such as stat1, traf3, and socs6, were identified in hemocytes during the hibernation period, thus indicating the important immune function of hemocytes. Therefore, our findings provide a unified classification of P. cantorii hemocytes and identify the genes related to the stress response, thereby providing a better understanding of the adaptive mechanisms of hibernation.

The giant freshwater prawn, Macrobrachium rosenbergii, is native to Southeast Asia and is used in aquacultural practices worldwide. It is considered advantageous because of its rapid growth, high nutritional value, and economic benefits. As one of the three major freshwater aquaculture shrimp sources in China, a high-quality genome resource is of great significance for promoting the germplasm improvement of varieties. This study presents a high-quality chromosome-level genome assembly of M. rosenbergii that was generated by combining PacBio, MGI, and Hi-C reads. The assembled genome was 2.96 Gb in size, with a contig N50 of 0.64 Mb and a scaffold N50 of 55.76 Mb, which was positioned on 59 pseudo-chromosomes. The Benchmarking Universal Single-Copy Orthologs (BUSCO) analysis for genome assembly reached 94.37%. In total, 27,111 protein-coding genes were identified, of which 25,470 were functionally annotated. These results provide a foundation for future research into adaptive evolution, genomics, and molecular breeding in M. rosenbergii.
