Potassium (K) has been recognized as an essential element in intensive agricultural production systems, and deficiency of K usually results in a decrease in crop yields. The utilization of potassium-solubilizing bacteria (KSB) to increase the soluble K content in soil has been regarded as a desirable pathway to increase plant yields. Following the inoculation of KSB in the soil, potassium can be released (in the form of K+) and consumed by plants. This study aims to investigate and compare the distribution characteristics of potassium-solubilizing bacteria between forest and plantation soils in Myanmar. In this study, 14 KSB strains were isolated from rhizosphere samples collected from forest soil, as well as fertilized rubber tree rhizosphere soil and fertilized bare soil from a plantation. Broadleaf forests with high levels of canopy cover mainly comprised the forest environment, and rubber trees were planted in the plantation environment. The Chao and abundance-based coverage estimator (ACE) indices showed that the microbial abundance of the plantation soil was higher than that of the forest soil. According to the Illumina MiSeq sequencing analysis results, the Shannon index of the forest soil was lower while the Simpson index was higher, which demonstrated that the microbial diversity of the forest soil was higher than that of the plantation soil. Potassium-solubilizing test results showed that the strains E, I, M, and N were the most effective KSB under liquid cultivation conditions. Additionally, KSB only accounted for less than 5.47% of the total bacteria detected in either of the sample types, and the distribution of dominant KSB varied with the soil samples. As another result, the abundance of Pseudomonas spp. in S1 was higher than in S2 and S3, indicating a negative impact on the growth of Pseudomonas in the fertilized rubber tree rhizosphere soil. The significance of our research is that it proves that the increasing use of KSB for restoring soil is a good way to reduce the use of chemical fertilizers, which could further provide a relatively stable environment for plant growth.

Nodularin (NOD) is greatly produced by Nodularia spumigena and released into the environment when toxic cyanobacterial blooms happened in natural water body, which is seriously harmful to human and animals. The promising bacterial strain of Sphingopyxis sp. USTB-05 was found to have an ability in biodegrading NOD. Initially, 11.6 mg/L of NOD could be completely eliminated within 72 h by whole cells of USTB-05, and within 36 h by its crude enzymes (CEs) of 570 mg/L, respectively. During the enzymatic biodegradation process of NOD, two products were observed on the profiles of HPLC. Based on the analysis of m/z ratios of NOD and its two products on a rapid-resolution liquid chromatogram-mass spectrum (RRLC-MS), we suggested that at least two enzymes of USTB-05 participated in biodegrading NOD. The first enzyme hydrolyzed Arg-Adda peptide bond of cyclic NOD and converted it to linear NOD as the first product. The second enzyme was found to cut off the target peptide bond between Adda and Glu of linearized NOD, and Adda was produced as a second and dead-end product. This finding is very important in both basic research and the application of USTB-05 on the removal of NOD from a water environment.

β-Nicotinamide mononucleotide (NMN), as a key precursor of an essential coenzyme nicotinamide adenine dinucleotide (NAD+), is most recognized for its pathological treatment effects and anti-aging functions. Here, the biosynthesis of NMN from the inexpensive feedstock substrate nicotinamide (Nam) using previously isolated Saccharomyces boulardii-YS01 was investigated. Ultra-high performance liquid chromatography coupled to triple quadrupole tandem mass spectrometry (UPLC-ESI-QqQ-MS/MS) was established for the determination and targeted analysis of NMN, nicotinamide riboside (NR), nicotinic acid (NA), Nam, and NAD+ in YS01 cells. Satisfactory precision and accuracy values were achieved with recoveries above 70% for five analytes. A 5~100 times higher content of NMN in YS01 (0.24~103.40 mg/kg) than in some common foods (0.0~18.8 mg/kg) was found. Combined with genome sequencing and enzyme function annotation, target-acting enzymes, including nudC, ISN1, URH1, PNP, and SIR2, were identified, and the biosynthetic pathway of NMN via Nam was suggested. The initial addition of 3 g/L Nam in the culture medium effectively promoted the generation of NMN, which raised the content of NMN by 39%. This work supplements an alternative resource for NMN production and lays the theoretical foundation for the further construction of NMN transgenic synthesis hosts.
