In precision viticulture, the intra-field spatial variability characterization is a crucial step to efficiently use natural resources by lowering the environmental impact. In recent years, technologies such as Unmanned Aerial Vehicles (UAVs), Mobile Laser Scanners (MLS), multispectral sensors, Mobile Apps (MA) and Structure from Motion (SfM) techniques enabled the possibility to characterize this variability with low efforts. The study aims to evaluate, compare and cross-validate the potentiality and the limits of several tools (UAV, MA, MLS) to assess the vine canopy size parameters (thickness, height, volume) by processing 3D point clouds. Three trials were carried out to test the different tools in a vineyard located in the Chianti Classico area (Tuscany, Italy). Each test was made of a UAV flight, an MLS scanning over the vineyard and a MA acquisition over 48 geo-referenced vines. The Leaf Area Index (LAI) were also assessed and taken as reference value. The results showed that the analyzed tools were able to correctly discriminate between zones with different canopy size characteristics. In particular, the R2 between the canopy volumes acquired with the different tools was higher than 0.7, being the highest value of R2 = 0.78 with a RMSE = 0.057 m3 for the UAV vs. MLS comparison. The highest correlations were found between the height data, being the highest value of R2 = 0.86 with a RMSE = 0.105 m for the MA vs. MLS comparison. For the thickness data, the correlations were weaker, being the lowest value of R2 = 0.48 with a RMSE = 0.052 m for the UAV vs. MLS comparison. The correlation between the LAI and the canopy volumes was moderately strong for all the tools with the highest value of R2 = 0.74 for the LAI vs. V_MLS data and the lowest value of R2 = 0.69 for the LAI vs. V_UAV data.

Crop protection strategies based on cupric products and mainly adopted in organic viticulture produce a consistent environmental impact due to the persistence of copper in soils and its negative effects on edaphic biodiversity. In this work, trials were carried out during the crop years 2018–2020 in a vineyard with an organic management by a low-copper strategy and in a conventional IPM management with an IPM strategy with reduced use of fungicides. Phytosanitary treatments have been strictly planned according to forecasting models, and fungicides have been partially substituted with substances improving the resistance mechanisms of plants. Different strategies of green manure management, in order to improve the health of vines, were also adopted. Results suggest the efficacy of the “GreenGrapes” plant protection strategy in conditions of low downy mildew pressure. Furthermore, no declines in the production quality have been recorded; conversely, the synergic effect of the green manure and the tested biostimulant substances (“GreenGrapes” protocols) and the green manure management improved yield and grape quality, compared with conventional conduction (IPM and Organic) with a grass covering.

Grapevine downy mildew, caused by Plasmopara viticola, is one of the most destructive vineyard diseases worldwide, with the potential to devastate up to 90% of harvests under adverse conditions. Traditional chemical-based control strategies are increasingly restricted due to environmental concerns and the development of resistant pathogen strains, prompting the search for sustainable alternatives. This study evaluated the efficacy of two yeast-based biostimulants (YE1, an experimental formulation, and YE2, Romeo, a commercial preparation containing Cerevisane®) in mitigating P. viticola infections in Vitis vinifera cv Cabernet Sauvignon under open field conditions. Field trials were designed to monitor the effects of these bioproducts on grapevine metabolism, with a focus on the biosynthesis of phenolic compounds linked to plant defense responses. Both biostimulants demonstrated significant efficacy against downy mildew, achieving protection levels above 93% during critical growth stages. Metabolic analyses of leaves revealed distinct impacts of YE1 and YE2 on secondary metabolite dynamics. YE1 primarily promoted the accumulation of flavonols, such as quercetin glycosides and kaempferol glucosides, which are known to contribute to pathogen resistance through antimicrobial activity and ROS detoxification. Compared to the untreated sample, quercetin glycosides levels in YE1 leaves were approximately 700 µg/g higher (+9.6%), in comparison to control conditions, on a dry weight basis after the first treatment in 2019 and about 900 µg/g higher (+13.7%) after the final treatment in 2020. A similar trend was observed for kaempferol concentrations, which were 70 µg/g (+98.7%) and 100 µg/g higher (+24.6%) on the same dates. In contrast, YE2 enhanced the biosynthesis of caftaric and coutaric acids, known for their antioxidant and antimicrobial properties. These compounds were induced by YE2 over time and, after the last treatment, in 2019 caftaric acid levels were approximately 345 µg/g higher (+25.9%), than control condition, while coutaric acid levels were 30 µg/g higher (+33.8%). The stilbene piceid was also induced at early stages by treatments, indicating its role in enhancing plant defenses. In 2019, after the first treatment, both YE1 and YE2 applications induced an increase in piceid concentration, compared to the control, of approximately 2.5 µg/g (+104.8%) for YE1 and 1.3 µg/g for Y2 (+55.0%). In contrast, in 2020, the effect was more pronounced for YE2, with concentrations exceeding the control by 2 µg/g (+46.2%). The results suggest that these yeast-based bioproducts act as elicitors, effectively enhancing grapevine immunity and reducing reliance on synthetic inputs. This study provides novel insights into the metabolic mechanisms underlying the efficacy of yeast-based biostimulants against downy mildew and offers practical guidance for their strategic application.

The research work carried out aimed at verifying the efficacy ofTrichoderma-based products on the fertility maintenance in vineyard soils, in case of replanting. The presence of a hypofertile horizon, in fact, can cause problems in the engrafting of rooted cuttings and a slowing down in their vegetative development. The trial was carried out at the experimental farm of CREA − Research Center for Viticulture and Enology of Arezzo, during the setting up of a new Sangiovese vineyard. Rooted grafts treated with three different formulations containing someTrichoderma spp.strains were planted, considering an untreated negative control. In addition, treatments with the same formulations were repeated in the 2014-2016 three-year period, by spraying the soil, with an injector pole. Every year the shoot length and the pruning wood weight were evaluated, as well as the presence/absence ofTrichodermaspp. on the root systems. Moreover, in 2016 the photosynthetic efficiency of the vines was verified. The presence ofTrichodermaspp. within the rhizosphere of the vines treated was stable over time; the differences found between treated and untreated thesis were considerable, while only minor differences emerged among the plants subjected to the three different treatments. According to the results obtained, the application ofTrichoderma-based products can be considered a valid practice to be used during vineyard implantation in case of difficult edaphic conditions.

Late spring frosts represent a major threat to grapevine (Vitis vinifera L.), a risk increasingly exacerbated by climate change-driven shifts in phenology. To explore sustainable strategies for frost mitigation, this study investigated the effect of a natural polysaccharide-based biogel, derived from carob (Ceratonia siliqua L.), on the molecular response of grapevine buds exposed to severe cold stress. To this aim, a preliminary RNA-Seq analysis was carried out to compare the transcriptomes of biogel-treated frozen buds (BIOGEL), untreated frozen buds (NTF), and unstressed controls (TNT). The transcriptomic analysis revealed extensive reprogramming of gene expression under freezing stress, highlighting the involvement of pathways related to membrane stabilization, osmotic adjustment, and metabolic regulation. Interestingly, the biogel treatment appeared to attenuate the modulation of several cold-responsive genes, particularly those associated with membrane functionality. Based on these preliminary transcriptomic data, twelve candidate genes, representative of the functional classes affected by biogel treatment, were selected for qRT-PCR validation. The expression patterns confirmed the RNA-Seq trends, further suggesting that biogel application might mitigate the typical transcriptional activation induced by frost, while supporting genes involved in cellular protection and integrity maintenance. The overall analyses suggest that the biogel may act through a dual mechanism: (i) providing a physical barrier that reduces cold-induced cellular damage and stress perception, and (ii) promoting a selective adjustment of gene expression that restrains excessive defense activation while enhancing membrane stability. Although further field validation is required, this natural and biodegradable formulation represents a promising and sustainable tool for mitigating late frost injuries in viticulture.
