The performance of a soybean genotype under water deficit stress at an early vegetative stage might be related to its general tolerance to drought. To investigate the plasticity of root and shoot seedling traits in response to drought at an early vegetative stage, a set of 32 soybean genotypes adapted to southeast European growing conditions was grown under polyethylene glycol (PEG) 8000-induced drought stress and well-watered control conditions. Under drought, mean tap root length (RL), shoot length (SL), root fresh weight (RFW) and shoot fresh weight (SFW) decreased significantly by 11, 17, 38 and 34%, respectively, while root dry matter (RDM) and shoot dry matter (SDM) increased significantly by 13 and 11%, respectively. Of the four derived traits, the ratios of RL/SL, RL/RFW and SL/SFW increased significantly by 8, 45 and 28%, respectively, under drought, while the ratio of RFW/SFW decreased significantly by 4%. However, a wide variation between genotypes was observed for all 10 studied seedling traits under both control and drought conditions. Broad sense heritability ranged from 0.53 (RL) to 0.97 (SL) under control conditions and from 0.56 (RL/RFW) to 0.96 (SL) under drought conditions. The correlation coefficients between the traits were either weak or moderate, indicating that the studied traits can be modified independently by selection.

Breeding resistant wheat cultivars to Fusarium head blight (FHB), caused by Fusarium spp., is the best method for controlling the disease. The aim of this study was to estimate general combining ability (GCA) and specific combining ability (SCA) for FHB resistance in a set of eight genetically diverse winter wheat cultivars to identify potential donors of FHB resistance for crossing. FHB resistance of parents and F1 crosses produced by the half diallel scheme was evaluated under the conditions of artificial inoculation with F. graminearum and natural infection. Four FHB related traits were assessed: visual rating index (VRI), Fusarium damaged kernels (FDK), and deoxynivalenol and zearalenone content in the harvested grain samples. Significant GCA effects for FHB resistance were observed for the parental cultivars with high FHB resistance for all studied FHB resistance related traits. The significant SCA and mid-parent heterosis effects for FHB resistance were rare under both artificial inoculation and natural infection conditions and involved crosses between parents with low FHB resistance. A significant negative correlation between grain yield under natural conditions and VRI (r = −0.43) and FDK (r = −0.47) under conditions of artificial inoculation was observed in the set of the studied F1 crosses. Some crosses showed high yield and high FHB resistance, indicating that breeding of FHB resistant genotypes could be performed without yield penalty. These crosses involved resistant cultivars with significant GCA effects for FHB resistance indicating that that they could be used as good donors of FHB resistance.

Trypsin inhibitors (TI) in raw soybean grain, mainly represented by the Kunitz trypsin inhibitor protein (KTI), prevent the normal activity of the digestive enzymes trypsin and chymotrypsin in humans and monogastric livestock. The inactivation of TI is achieved through costly and time-consuming heat treatment. Thermal processing also impairs the solubility and availability of the soybean grain protein. Therefore, the genetic elimination of KTI has been proposed as a suitable alternative to heat treatment. The aim of this study was to screen the collection of European soybean cultivars with six genetic markers (one SSR marker and five SNP markers) previously proposed as tightly linked to the KTI3 gene encoding the major Kunitz trypsin inhibitor seed protein of soybean and validate their usability for marker-assisted selection (MAS). The six markers were validated on a subset of 38 cultivars with wide variability in KTI content and in the F2 and F3:5 progenies of two crosses between the known high- and low-KTI cultivars. Three genetic markers (SSR Satt228 and two SNP markers, Gm08_45317135_T/G and Gm08_45541906_A/C) were significantly associated with KTI content in a subset of 38 cultivars. Low-KTI alleles were detected in both low- and high-KTI genotypes and vice versa, high-KTI alleles were found in both high- and low-KTI genotypes, indicating a tight but not perfect association of these markers with the KTI3 gene. The genetic marker SSR Satt228 showed a significant association with KTI content in the F2 progeny, while the SNP markers Gm08_45317135_T/G and Gm08_45541906_A/C allowed significant discrimination between progeny with high- vs. low-KTI progenies in the F3:5 generation. These three markers could be applied in MAS for low-KTI content but not without the additional phenotyping step to extract the desired low-KTI genotypes.

The development of maize varieties with enhanced tolerance to drought stress has become a high-priority goal for maize breeding programs worldwide. In order to assess the variability of root and shoot traits in response to drought at an early vegetative stage, a set of 32 maize single-cross hybrids was grown under polyethylene glycol 8000-induced drought stress and well-watered control treatments. Drought stress significantly reduced hybrid seedling root and shoot lengths (RL and SL) as well as root and shoot fresh weights (RFW and SFW), while an increase in seedling root and shoot dry matter (RDM and SDM) and root fresh weight-to-shoot fresh weight ratio (RFW/SFW) was observed. The high heritability estimates for the four directly and easily measured traits, namely, RL, SL, RFW, and SFW (0.83, 0.83, 0.74, and 0.74, respectively), and medium-to-very-strong positive correlations among these traits under drought conditions indicate their applicability for the assessment of maize drought tolerance at the seedling stage and may represent a practical contribution to maize breeding programs for improved drought tolerance. Among the studied hybrids, hybrids 30, 3, and 23 were characterized by the largest RL under drought conditions and small relative change in RL between control and drought treatments. Hybrid 30 also showed one of the smallest relative reductions in SL, RFW, and SFW between the two treatments, while hybrids 3 and 23 were among those which exhibited the highest relative RL/SL and RFW/SFW increase between the two treatments, which supports their potential as parental lines in drought-tolerant breeding.

Maize is considered a promising candidate for biofortification through breeding, given its widespread cultivation and significance as a food crop. This cost-effective and sustainable approach could be used to increase the content of different tocol compounds, i.e., vitamin E, in maize grain due to the well-documented genetic variability. In the present study, an evaluation of the content of nine different tocol traits was performed in a genebank collection of 88 inbred lines of temperate maize grown at two locations in Croatia in 2019. A large genotypic variability within the studied material was observed for α-tocopherol, γ-tocopherol, δ-tocopherol, total tocopherols, α-/γ-tocopherol ratio, α-tocotrienol, γ-tocotrienol, total tocotrienols and total tocols with corresponding coefficients of variation of 52, 61, 51, 45, 106, 24, 54, 33 and 38%, respectively. Heritability estimates ranged from 0.66 for α-tocotrienol to 0.95 for γ-tocopherol. The content of α-tocopherol, which has the highest vitamin E activity and is therefore most interesting for selection, was not significantly correlated with either grain color or grain type. Comparison of the effects of simulated selection with an intensity of 20% on increased α-tocopherol content using the two selection criteria, absolute α-tocopherol content and α-/γ-tocopherol ratio, showed that the highest absolute α-tocopherol content was a better selection criterion. Indeed, simulated selection based on the absolute α-tocopherol content resulted in a 64% increase in this compound without significantly affecting the total tocopherols, the total tocotrienols, and the α-/γ-tocopherol ratio.

Background/Objectives: A critical part of the maize life cycle takes place during the summer, and due to climate change, its growth and development are increasingly exposed to the irregular and unpredictable effects of drought stress. Developing and using new cultivars with increased drought tolerance for farmers is the easiest and cheapest solution. One of the concepts to screen for drought tolerance is to expose germplasm to various growth scenarios (environments), expecting that random drought will occur in some of them. Methods: In the present study, thirty-two maize hybrids belonging to four FAO maturity groups were tested for grain yield at six locations over two consecutive years. In parallel, data of the basic meteorological elements such as air temperature, relative humidity and precipitation were collected and used to compute two indices, scPDSI (Self-calibrating Palmer Drought Severity Index) and VPD (Vapor Pressure Deficit), that were assessed as indicators of drought (water deficit) severity during the vegetation period. Practical implementation of these indices was carried out indirectly by first analyzing yield data using a factor analytic model to detect latent environmental variables affecting yield and then correlating those latent variables with drought indices. Results: The first latent variable, which explained 47.97% of the total variability, was correlated with VPD (r = −0.58); the second latent variable explained 9.57% of the total variability and was correlated with scPDSI (r = −0.74). Furthermore, latent regression coefficients (i.e., genotypic sensitivities to latent environmental variables) were correlated with genotypic drought tolerance. Conclusions: This could be considered an indication that there were two different acting mechanisms in which drought affected yield.
