Replacing partial chemical fertilizers with organic fertilizer can increase organic carbon input, change soil nutrient stoichiometry and microbial metabolism, and then affect soil organic carbon (SOC) storage. A 6-year field experiment was used to explore the mechanism of SOC storage under different ratios of manure substitution in northeast China, with treatments including chemical fertilizer application alone (nitrogen, phosphorus, and potassium, NPK) and replacing 1/4 (1/4M), 2/4 (2/4M), 3/4 (3/4M), and 4/4 (4/4M) of chemical fertilizer N with manure N. Soil nutrients, enzymatic activity, and SOC fractions were analyzed to evaluate the effect of different manure substitution ratios on SOC storage. A high ratio of manure substitution (>1/4) significantly increased soil total N, total P, total K, and available nutrients (NO3−-N, available P, and available K), and the 4/4M greatly decreased the C/N ratio compared to the NPK. Manure incorporation increased microbial biomass carbon (MBC) by 18.3–53.0%. Treatments with 50%, 75%, and 100% manure substitution (2/4M, 3/4M, and 4/4M) enhanced bacterial necromass carbon (BNC), fungal necromass carbon (FNC), and total microbial necromass carbon (MNC) by 31.9–63.5%, 25.5–107.1%, and 27.4–94.2%, respectively, compared to the NPK treatment. Notably, the increase in FNC was greater than that of BNC as the manure substitution ratio increased. The increasing manure substitution significantly enhanced particulate organic C (POC) and total SOC but did not affect mineral-associated organic C (MAOC). High soil N and P supplies decreased leucine aminopeptidases (LAPs) and alkaline phosphatase activities but increased the activity ratio of β-glucosidase (BG)/(N-acetyl-glucosaminidase (NAG) + LAP). Treatments with 25% manure substitution (1/4M) maintained maize and soybean yield, but with increasing manure rate, the maize yield decreased gradually. Overall, the high ratio of manure substitution enhanced SOC storage via increasing POC and MNC, and decreasing the decomposition potential of manure C and soil C resulting from low N- and P-requiring enzyme activities under high nutrient supplies. This study provides empirical evidence that the rational substitution of chemical fertilizers with manure is an effective measure to improve the availability of nutrients, and its effect on increasing crop yields still needs to be continuously observed, which is still a beneficial choice for enhancing black soil fertility.

The long-term use of chemical N fertilization may have a negative impact upon soil fertility and quality. On the contrary, organic fertilization is considered a sustainable development agricultural strategy. However, the remediation effect of organic fertilization on agroecosystems remains unclear. This study was conducted in a long-term (1979–2020) field experiment to investigate the influence of organic and chemical fertilizers on the soil microbiome assembly processes. The experiment consisted of six treatments: chemical N fertilization (N), double N fertilization (N2), organic fertilization (M), organic and N fertilization (MN), double organic and N fertilization (M2N2), and unfertilized control. The chemical N fertilization (N and N2) treatments significantly decreased soil microbial diversity, as well as soil pH, compared to the Control treatments (p < 0.05). MN and M2N2 treatments increased microbial diversity compared to that of N and N2 treatments. The combination of organic and chemical N fertilizer recovered the decreased microbial diversity to the level of the Control and M treatments, but the application of double organic fertilizer (M2N2) still showed a significantly lower microbial diversity than that of the Control and M treatments. From the results of the microbial community assembly processes, it was found that environmental filtering was induced by N fertilization, while organic fertilization developed a stochastic process and mitigated the role of environmental filtering in the community assembly process. An ecological network analysis showed that the decrease in Acidobacteria in organic fertilization treatments played a key role in mitigating the soil acidification induced by 40-year chemical N fertilization. It indicated that organic fertilizer could mitigate the decrease in soil fertility induced by chemical N fertilization. Higher environmental filtering effects in M2N2 than those in MN treatments suggested that the mitigation effect of organic fertilizer was weakened when double chemical N fertilization was applied in black soils. These results are helpful for a unified understanding of the ecological processes for microbial communities in the development of sustainable agriculture.

Stalk rot is one of the most destructive and widely distributed diseases in maize plants worldwide. Research on the performance and resistance mechanisms of maize against stem rot is constantly improving. In this study, among 120 inbred maize lines infected by Fusarium graminearum using the injection method, 4 lines (3.33%) were highly resistant to stalk rot, 28 lines (23.33%) were resistant, 57 lines (47.50%) were susceptible, and 31 lines (25.84%) were highly susceptible. The inbred lines 18N10118 and 18N10370 were the most resistant and susceptible with disease indices of 7.5 and 75.6, respectively. Treatment of resistant and susceptible maize inbred seedlings with F. graminearum showed that root hair growth of the susceptible inbred lines was significantly inhibited, and a large number of hyphae attached and adsorbed multiple conidia near the root system. However, the resistant inbred lines were delayed and inconspicuous, with only a few hyphae and spores appearing near the root system. Compared with susceptible inbred lines, resistant maize inbred line seedlings treated with F. graminearum exhibited elevated activities of catalase, phenylalanine ammonia-lyase, polyphenol oxidase, and superoxide dismutase. We identified 153 genes related to disease resistance by transcriptome analysis. The mitogen-activated protein kinase signaling and peroxisome pathways mainly regulated the resistance mechanism of maize inbred lines to F. graminearum infection. These two pathways might play an important role in the disease resistance mechanism, and the function of genes in the two pathways must be further studied, which might provide a theoretical basis for further understanding the molecular resistance mechanism of stalk rot and resistance gene mining.

Soybean root rot, a soil-borne fungal disease, is caused by multiple pathogens that seriously affect soybean production. During spring 2021, 92 pathogenic fungal strains were isolated from soybean plants with root rot in Hailun City, Heilongjiang Province, China. Through morphological and molecular identification, these strains were identified as Fusarium oxysporum (39.1%), F. asiaticum (30.4%), F. graminearum (13.0%), Pythium macrosporum (8.7%), and Rhizoctonia solani (8.7%). Among them, F. oxysporum was the dominant species, and F. asiaticum, not previously reported as a soybean root rot pathogen in Northeast China. Approximately 50% of the F. asiaticum isolates were moderately pathogenic. In addition, F. asiaticum had a wide host range, infecting black soybean, French bean, white hyacinth bean, mung bean, and adzuki bean but not corn, peanut, rice, and oat roots. Regarding field management, fludioxonil and pyraclostrobin had the best control effects of 73.8% and 69.4%, with EC50 values of 0.0029–0.0071 μg·mL−1 and 0.0045–0.0076 μg·mL−1, respectively. The study reported that F. asiaticum is a pathogen causing soybean root rot in northeast China. The application of chemical fungicides and non-host crop rotation can effectively control the disease caused by F. asiaticum.

Soil erosion results in dissolved organic matter (DOM) loss and is one of the main paths of soil carbon loss. Bacteria affect the generation and transformation of DOM. However, the effect of bacteria on the composition and slope distribution of DOM has rarely been investigated under field conditions. Based on a long-term experiment of three gradients (3°, 5°, 8°) in a black soil erosion area of Northeast China, the content, composition, and source of DOM were studied. The results showed that the DOM of the 3° and 5° slope was enriched midslope, and the DOM of the 8° slope was enriched downslope. Parallel factor (PARAFAC) analysis indicated that the main substances in DOM were fulvic-like acid, humic-like acid, tryptophan-like protein, and soluble microbial metabolites. The upslope and downslope soils of 3° and 5° slopes showed high DOM bioavailability, while the downslope soil of the 8° slope showed high DOM bioavailability. The content of new DOM in downslope soil increased with the gradient. Bacteria played an important role in the synthesis and transformation of DOM and affected its composition and slope distribution. Verrucomicrobiota, Firmicutes, Planctomycetota, and Gemmatimonadota were the main factors affecting soil DOM. The results could be helpful in understanding the loss mechanism of DOM in eroded black soil and provide support for soil carbon sequestration.

The degradation of farmland in China underscores the need for developing and utilizing saline-alkali soil. Soil health relies on microbial activity, which aids in the restoration of the land’s ecosystem, and hence it is important to understand microbial diversity. In the present study, two Gram-stain-positive strains HR 1-10T and J-A-003T were isolated from saline-alkali soil. Preliminary analysis suggested that these strains could be a novel species. Therefore, the taxonomic positions of these strains were evaluated using polyphasic analysis. Phylogenetic and 16S rRNA gene sequence analysis indicated that these strains should be assigned to the genus Halalkalibacter. Cell wall contained meso-2,6-diaminopimelic acid. The polar lipids present in both strains were diphosphatidyl-glycerol, phosphatidylglycerol, and an unidentified phospholipid. The major fatty acids (>10%) were anteiso-C15:0, C16:0 and iso-C15:0. Average nucleotide identity and digital DNA#x2013;DNA hybridization values were below the threshold values (95% and 70%, respectively) for species delineation. Based on the above results, the strains represent two novel species of the genus Halalkalibacter, for which the names Halalkalibacter flavus sp. nov., and Halalkalibacter lacteus sp. nov., are proposed. The type strains are HR 1-10T (=GDMCC 1.2946T = MCCC 1K08312T = JCM 36285T), and J-A-003T (=GDMCC 1.2949T = MCCC 1K08417T = JCM 36286T).
